    I personally noticed whoever that takes Glucosamine, is usually a health conscious or an athletic or very active. And this is an observational study. Not sure if it’s just a correlation or causation. But probably at least there is no harm taking it. Thanks !

      Agreed. Likely no harm in taking glucosamine, but is there a true benefit? Hopefully further research will answer that question

      @@DrBradStanfield please see my comment I have found that MSM works more powerful for my broken cartilage from my neck injury that has been with me all of my life. However I do take glucosamine chondroitin turmeric bioperine and other things with it. Thanks for the great review very insightful and informative all of your reviews I love them thank you.

      @@DrBradStanfield I'm a bit critical of the glucosamine studies in general because they don't distinguish between pain and the progression of the OA disease, that is, they tend to judge the effectiveness of glucosamine based solely on the pain factor. Years ago I started taking glucosamine because of a study done in England based on x-rays of the patients knees. Before and after x-rays of the patients were taken, and in the control group, the after x-rays showed continued degradation of their knees, but the before and after x-rays of the glucosamine group showed little or no deterioration. There may be even some thought that it might reverse OA to some minor degree in some patients. I'm skeptical of the pain studies partly because OA and bursitis are separate diseases, but similar in symptoms, or do I have that wrong?

    Glucosamine binds to and inactivates Gliadin, WG Agglutinin and other lectins thus preventing damage to GI damage. If glucosamine is consumed with meals it should decrease the inflammation caused by regular intake of wheat and other lectin rich foods.

      @@g.s.5868 I could only found one study where they used plain glucosamine.
      Isolation of a Glucosamine Binding Leguminous Lectin with Mitogenic Activity towards Splenocytes and Anti-Proliferative Activity towards Tumor Cells
      Glucosamine binds to that specific brown bean lectin.
      Lectins have very specific binding affinities and i guess some may bind to glucosamine sulphate. But yeah, most of the research is done on NAG, which is also available as a supplement that supposedly treats autoimmune disorders.

    Dear Dr Brad. Thank you for such an informative video. I was diagnosed with knee arthritis in my late 30s. I did some research which led me to start talking Glucosamine and Chondroitin 1500 mg. I have been taking this supplement for years now. I recently went for a full body check up including MRI scan and other bones tests and the results were quite clear that I do not have arthritis anymore and my bones are biological younger than my chronological 55 years of age. I also suggested to my neighbour to take the same supplement because she was limping for years due to an injury in her knee. She has now totally recovered and told me that taking this supplement has changed her life for the better in general like nothing has ever done. Hope this helps. Lots of love from London

      I've been taking Glucosamine everyday for the last 25 years, (it stopped the pain I suffered when I sat on a hard surface), & Chondroitin for about 15 years, (which stopped the heel pain I had endured for 16 years - And when I have tried to stop these supplements, both ailments return within days.

      @@Booklivre - Have been taking Gluco for so long, it's hard to compare the difference - Having said that, I am light skinned, & when i compare myself to people of a similar age 66, I think many look red-faced in comparison - Hope that helps a little.

    I've been taking Glucosamine Sulphate for several years to stop my hips hurting, probably due to arthritis just starting.
    After starting NMN many of my aches and pains went so I stopped the Glucosamine.
    After a few days the hip pain came back but not as strongly. I started back on the Glucosamine and now, no pain. I did miss one day of both tablets accidently, without realising it and noticed in the night some pain in my hips for the first time in three weeks, nexr morning I realised that I had missed the previous days tablets.
    Keep up the trials!!

    How is about 2010 year study that says that "High doses or prolonged use of glucosamine causes the death of pancreatic cells and could increase the risk of developing diabetes, according to a team of researchers at Université Laval's Faculty of Pharmacy"??? Details of this discovery were published on the website of the Journal of Endocrinology 2010...

      I think those results were from an in vitro study with insanely high doses of glucosamine. I asked a doctor and he said even just water at huge amounts would be disastrous to cells. Drugs are also a matter of dosing. That study pissed me off because what’s the point of using such a high dosage in vitro? I stopped taking glucosamine after that study scared me even I thought it’s non sense but i remaniés uncomfortable with taking it, because why would you create a new issue with a drug that might not even cure your other issue

    I would be curious if they adjusted for sulfur content (glucosamine sulfate for example) . I suspect that many of the benefits people ascribe to glucosamine actually are due to the sulfate. Many glucosamine supplements include MSM (methylsulfonylmethane), a form of sulfur

    due to knee pain i could not sleep whatever position, Gluco & Condro taken for weeks, pain gone! My mother-in-low was recommended for knee surgery, took both G&C 1400mg daily and weeks after became pain free and could walk without crutches

    Years back in the 1990s I went to a chiropractor who told me of a study that aired on 60 Minutes which was done on patients that had back pain and joint pain and these patients were put on glucosamine and the results were amazing 85% of them significant improvements.big pharma didn't like that someone was eating there lunch😢

    I have a friend who is a research scientist for Hills pet foods. He was telling me about testing Glucosamine in dogs and were having significant benefits. So I started giving my dogs this supplement. Also they got good quality Hills pet food and little junk food, daily walks. They both lived 17 years, which is at the upper boundry for dachshunds. I know Hills food scientists believe in this molecule and with dogs you can rule out placebo.

      dachshunds are known to live very long. the dachshund on our street was 19th. he was very stiff lately. probably he had never received glucosamine...😂

    In the nineties there were studies of glucosamine sulfate and it had the effect of decreasing the frequency of coronary events by 7 times

    Another excellent video Brad - thank you. I fear chances are your hunch will prove correct: RCT and interventions trial will not show a positive result. I know almost nothing about medicine. However, I build low signal-to-noise ratio (SNR) models for living. My biased opinion is that adjusting the observational results for confounding factors (sex, income, lifestyle etc) is impossible to do with a degree of certainty that would make 15% improvement (post adjustments) a reliable indicator. If that number was say 90% or higher, that would have been another matter. Only 15% is too low.

    N-acetyl glucosamine is taken for osteoarthritis and inflammatory bowel disease (IBD), including ulcerative colitis and Crohn's disease.

    Thank you for this exhaustive analyze! Again a video that explains a lot.
    This is very interesting, at least if it might extend life maybe means it might not worsen blood sugar as previously suspected. I recently started taking it again and I will continue for now because I would take anything that helps with joint health and tendonitis. In the past I took up to 5 grams per day and this is when I felt some slight improvement but I can’t be sure if it was in my head or due to other factors. I am now taking 1-2 grams a day and unsure to go up to 5. The safety for different dosages and long term are always a concern.

    It would be fascinating to do a kind of meta study on observational studies. By that I mean checking for a number of different research areas how well the conclusions of observational studies aligned with the conclusions of RCTs. In this way you could quantify how helpful observational studies are.
    I find it very hard to put much stock in observational trials-there's just so many possible confounds that they seem almost pointless. I think doing a study like this could make some serious changes in how scientists interpret the results of observational studies across fields.

    I have a son with a life threatening illness. Many years ago when he was a young child I had a chat with one of his doctors. I told him I had found information suggesting a readily available food supplement might help offset some of the side effects of the medications he was taking. After telling me that theoretically the supplement I suggested might indeed help, and telling me that knew of no research to confirm this, he offered this perspective. He said, if it might help and it won't hurt and it doesn't come at a great cost, then there's no reason not to do it. So when I heard about this study knowing that I have had some cardiac issues myself, my father had cardiac issues, and his father died of a heart attack in his late 50's... and finding that a Glucosamine supplement was not cost prohibitive and it might help especially for those of us with heart issues, it seemed like a no brainer. So I've been taking a glucosamine/chondroitin/MSM supplement ever since. I've informed my primary care physician and my cardiologist who have not expressed any concerns about taking it.

    I have a similar personal experience with taking 1500mg Glucosamine a day. I started taking 20years ago introduced by a friend in his 50s who was representing his country in senior field hockey. He also took the supplement since his 30s as a preventative measure. I am now in my 50s and no sign of joint pain not any health issues.

But just with the fact that there is no way to know if Craig doesn't have joint pain because he has taken glucosamine or just because many other people don't have joint pain at that age without doing anything special. As you stated yourself, "correlation is not causation".

    My contribution to this topic, is that glucosamine can potentially give rise to higher IOP (intraocular pressure). The significance of this is that high IOP is a marker of glaucoma, it tends to increase as we age. This effect is more marked in older patients, which ironically is the group most likely to suffer from a variety of aches and pains, this prone to taking glucosamine.
    If I were to start glucosamine, I'd get s baseline IOP, and retest every couple months to see if there was an increase. Glaucoma is no joke.

    After being on a Chondroitin/Glucosamine/Boswellia Acid/MSM stack for a few weeks, good at first but then relief tapering off, I use this stack now in place of NSAIDs. In other words, I use them when pain is at its worst but otherwise taper off. Seems to work at least at the moment.

    If youd suffer from knee pain due to OA every day like me, even if there is 1% to help your would take it, believe me. And I have read so many books describing why it helps in OA (with chondroitin, MSM and ASU) - the KEY is the right dosage and most supplements are to low.
    If it also increases lifespan and there is no known side effects, this one is no brainer for me.

    There are some key people I follow/ed for health these included some of big keto channels and Sinclair. Further research has shown the sources to be not as trust worthy as I had presumed. Are there any particular channels/individuals you would recommend following?

    I certainly hope it’s true it extends health. I’ve been taking about 800MG a day of Glucosamine (Chondroitin and MSM) for my joints for 20+ years. No joint pain at 64.

    It's good at combatting low grade chronic inflammation, certain aspects of the integrated stress response, and how we process carbs.
    I wouldn't call it a breakthrough, and more a way to combat the side effects of a terrible diet.

    Can you make a video on chronic insomnia and what meds will cause me to start dreaming again? Currently on xywav for the restorative qualities but the best sleep of my life was when I had deep sleep as well as sleep where I had rem and had dreams.

    One other mechanism that might have to do with placebo etc, is that arthritic patients who are suffering less joint pain will lead more active lifestyles, (and more activity extends life).

    I like your critical approach to reading studies. Like me, after 40 years in pharma, i always remain skeptical of observational studies because of low HR and cofounding factors. So we never used them in the design of our trials. Like you mentioned the placebo effect in observational studies is so high that results are confusing.
    Also on the resveratrol study, you were totally right to call Mr Sinclair. I will not take supplements like those but i will gladly continue to drink my red wine, because of taste not health benefits.

      I have read that any attempt to get resveratrol, whether it is beneficial or not, from red wine must be done with grapes that have not been sprayed with anti-mold chemicals, which means the red wine must be organic to guarantee this.

    Really interesting. I was wondering whether it's not useful to recommend Glucosamine just for the Placebo effect. If the placebo effect is that big, it seems like a good argument, even if it is not glucosamine in the end (as long as there are no side effects)?

    I've been taking it since tearing a mensicus in my knee in 2003 when I was 42. I was told at the time that I would eventually have arthritis in my knees. A few years later I ran across a study that suggested Glucosamine Sulfate was the type that was shown to work in studies and trials. So I've been taking both kinds. Starting to experience that arthritis 19 years later at the age of 60. Don't really know if the Glucosasmine delayed the symptoms or not. Maybe it helped, maybe it just enriched the supplement industry.

    I’ve taken glucosamine, chondroitin and msm for years to support my joints. I’m a runner who Year’s ago was overweight 320 lbs, now since 2016, 220lbs. It seems to help as I’ve never had any joint issues.

      Not every supplement. Glucosamine is one of the least surprising to have shown a human lifespan increase. Glucosamine has previously shown lifespan increases in mice and c elegans. N Acetyl Glucosamine was able to extend c elegans lifespan by 50%, which is greater than almost any other substance tested.

    My experience with G: I took G. for around 3-4 years almost every day (due mostly to art. problems I have from sport). I stopped recently taking it because I realised my glycemy level went way too high compared to several years ago when it was normal. I am not sure that this is the result of taking G, or other factors, but considering I had a normal -ish life/diet, I have to take this into consideration.

    I have been taking glucosamine tablets for years now, I believe it has a beneficial effect for helping alleviate joint pain and soreness.

    Over 20 years ago, I started taking Glucosamine/Chondroitin for carpal tunnel symptoms resulting from spending far too much time at the keyboard every day. Now I'm 80. I still spend far too much time at the keyboard but the carpal tunnel syndrome has never come back. I understand the concepts that correlation does not guarantee causation and that humans are prone to the placebo effect. But it is good to hear from you that my Glucosamine/Chondroitin habit is unlikely to be doing harm.

    Observational studies like this I think are so prone to so many confounding factors they’re even less informative than in-vitro studies for new drugs.

    I gave glucosamine a try 3 times in my life for knee arthritis. Each time I took supplements as prescribed for several months. I’ve had so many testimonials from friends touting it’s benefits, but it just didn’t do anything to improve my knee pain.

    For me, it's a cost/benefit argument. As I get up in years (early 60s) and having suffered some joint injuries, I found (imagined?) that my use of Glucosamine helped with my knee "loosens/instability". But my Glucosamine is mixed with chondroitin, plus I started Glucosamine when I started other things like hyaluronic acid, so I can't disentangle their effects. In the end, Glucosamine is inexpensive so that clinches the argument. Once again, a nice video, thanks Dr Stanfield!
    Are there studies that clearly show that Glucosamine indeed HAS an anti-inflammatory effect?!? That argument felt like "hand waving".

    I took relatively high doses (9 - 12 grams/day) of glucosamine and chondroitin proactively since my 30's. Developed severe arthritis in both knees and both shoulders in my late 50's anyway. Had to stop running completely and sharply curtail my weight lifting. So far I've tried cortisone shots, PRP, stem cells, and hyaluronic acid with little success while trying to avoid joint replacements.

    I have been taking glucosamine for 22 years. A surgeon told me after knee surgery that I had a better chance of not having knee replacement after an arthroscopic procedure if I took glucosamine. I was told at that time my knee had significant degradation of the cartilage. 20 years later I had knee x-rays in 2021 at the age of 54. The doctor was impressed with how much space I had remaining in the joint for my age and activity level. In 2022 I had to have a flap removed from the meniscus. Again the surgeon said after surgery that my knee was in amazing shape for my age. I also have noticeable improvement in joint comfort when I am using glucosamine. I have failed to take it a few times in the years because I ran out and did not get to the store for a few weeks. I really notice when I come off of it for a period of time. I am a believer.

    Can you please make suggestion on how to find quality creatine supplements cause a lot of brands state it’s 99% pure however products are hardly tested by independent agencies for purity, also a lot of creatine comes from China where synthesis of creatine is poorly tested/regulated.     Pycnogenol helps with joints too amongst many other things as well as inflammation. Also doesn’t lemons help with inflammation plus a bunch of other benefits? Seeing inflammation seems to maybe be the big problem..

    Sometimes observational studies are the best at revealing results.After hearing a lot of great things through non-observational studies about Resveratrol,I decided it would be a good investment.After taking R for a while,I "observed" myself feeling puny,weak,and tired more often than usual.I stopped taking R based on this personal observation and now all is well,lol.We need to stop basing what is good for humans based on what mice or other animal species can tolerate.

      There are also studies that prove taking resveratrol every other day is providing max benefits. Basically, taking resveratrol daily is not that good as opposed to taking it every two days.
      Keep in mind that reveratrol is a signaling molecule that tells your body that times are tough and it should start repairing. You don't want to be in this state every day.

    Had severe knee pain from arthritis in one knee doctor recommended naproxen which did nothing except give me gastritis and a druged up feeling. Dr said to stop and recommended steroids. I said nope what else have you got and then was told to try glucosamine. I got 100% relief in 5 days that's lasted for 4 years so far. Would a placebo effect last that long? I will keep taking it daily unless things change but so far fantastic results with no side effects.

    How or why does glucosamine supplementation focus exclusively on the knee? I believe you said it was used to reduce inflammation in general. Also ingeneral, I have started "pulsing" my supplements so not to take too much of anything on a regular bases. I think the same approach could be taken with diet and exercise.

      Because the arthiritc knee is one of largest and most common issues that come with ageing and can come relatively young as well with shive is oven life . It’s also very difficult to help because majority of the e cartilage is non vascular . People often take a glucosamine chindriotin suplemt together specifically fo this reason and chrondoiton is a major component in cartilage . It’s been nearly decade since I sat through my tissue engineering lecture on cartilage

    6:08 Dr. Stanfield mentions an ITP trial and a large randomized trial showing metformin didn't benefit people without diabetes. I wasn't able to find those studies. Can someone who knows provide links to them? Thanks.

    I found a study by Yunjeong Gwon, Jisun Oh & Jong-Sang Kim in 2020 that, as far as I understand it, says that sulforaphane actually increases the growth of colon cancer cells caused by a P53 mutation.
    So how safe is it to take sulforaphane prophylactically if you don't know if you currently have P53 cancer cells in your colon? I am afraid that I will even accelerate the growth of mutated cells through the increased consumption of sulforaphane.

    I've been taking glucosamine to minimize joint risk. The hard thing with these supplements are that if they prevent something, you won't be able to notice it. I have never had any joint pains or injuries. I'm 43 years old and lift heavy ( close to 500 pounds squat) without any kind of steroid use. By the way, Dr. Stanfield, is there any knowledge about cycloastragenol? I used it for couple of months 2 years ago, and while I can't tell if it had any longetivity effect, my eyesight got noticeably better.

    We need trials about sooooo many supplements. The issue I see with this one is that multivariate adjustments in no way reflect the nonlinear interactions of the possible confounds. Finally, a 15 % reduction of mortality rates just between ages of roughly 60 and 70 is NOT a 15% (12 years!!!) increase in lifespan. In fact I bet there's a catchup effect on that preserved cohort not very far down the line.

    Do you respond to messages usually? I want to donate but I want to talk to you first. I think that you have a great potential for being a major player in the longevity field. To do that just being a researcher is not enough. We are about to move much faster with way more money than 400k you require for the study. Try to stay relevant and keep looking at your fan base questions and individual comments.

    I took Glucosamine for years & stopped because of increased muscle camps & weight gain.This was after emergency heart surgery. I stopped and noticed improvement after a couple of months without it & no notable change in joint pain or other issues. I am happier with out it.☮🕊

    @Dr Brad Stanfield Could you one day cover the carnivore diet? It is a very interesting topic. Anthropologically, it makes sense for us to be optimized for it. The literature seems to go against this due to the whole MTor debacle and yet empirical evidence seems to fight it. For example, Centenarians tend to eat high protein diets. The argument I keep hearing is that insulin (due to carbs) tends to cause higher MTor activation than protein. Any thoughts on that?

    There was a RCT back in 2013 which showed that glucosamine use was associated with a statistically significant rise in intraocular pressure, especially in the elderly. Sample size of 88 though. I don't believe this has been replicated, but could this be cause for concern?

      Thanks for sharing. I'm not aware of that as a side effect. If it was a massive concern, it should have shown up in the data by now (such as this large observational study)

      I should read the comments before I do. Just posted about this. I had been using the supplement when this research came out, bc I was so active in the gym and wanted to support my joints. I stopped immediately when the news came out, and yes, bc IOP had been creeping up while my eye doctor said nothing. I now have to use drops to control my IOP. May be a coincidence, but glaucoma does not run in my family.

      @@DrBradStanfield They may not have checked IOP? They may not have been aware of the study, or had reason to ignore it. There's no pain, redness or other obvious symptoms associated with elevated IOP, which is why glaucoma is so insidious, often the first sign is loss of peripheral vision.

      It seems as though risk increases as we age. So you may not have seen an increase in earlier years, but may in the future.

      No.
      Degenerative conditions like emphysema can't be treated with glucosamine or any other drug/supplement. However there *is* one thing that can reverse a degenerative condition, and that's your own body's autophagy. Prolonged fasting (fast longer than 24 hours) is what triggers your body to start doing autophagy.

    I heard and read some stuff about it and ijust start a week ago but many years ago I see the neighbor dog and barely walking and limping and the dog was sad to look at and 3 months later I see the dog running and he looks full of energy then I ran on the street to see the neighbor and ask him what the hell have you been doing to your dog and look at me and say I m giving glucosamine i was totally stunned by the results.

    I have tried glucosamine, but didn't feel any difference. I have rheumatoid arthritis and have endeavored to relieve pain other ways besides pain medication. There are other supplements that do help. I focus on those.
