      Niacin (NAD precursor)+ apigenin (cd38 inhibitor) = increased blood NAD. The unanswered question is ... does this increase translate to greater cellular NAD throughout the body for better health and longevity?

      Do you have data for dosages based on the amount of the supplement per kilogram body weight? (For example, the way Ivermectin is dosed at 2mg per kg body weight? Or 1.5 grams Sodium Ascorbate per kg body weight?)
      The dosages you are citing seem rather low.

    This is FANTASTIC (1) analysis, (2) story composition and (3) delivery.
    Life saving innovation would accelerate FAR more quickly than its current glacial pace if more people followed this example.
    For this reason alone, we should all work to spread the influence of this channel.

    I've tried lots of supplements over the years (56 now) and I've bee taking NMN for about 3 years now and some of the expected the effects are pretty clear like more energy, not feeling like having a nap in the afternoon, strength at the gym, libido.....I don't contest the studies and I still like to see studies with different results......but yeah, I can't deny how I physically feel too.

      I’ll attest to this. I’ve taken it for a month. Mainly because of my knees giving out and drastically effecting my work out routine. Also hormonal weight gain that was fast and furious.
      After four weeks I’m back to running twice a day plus an entire squat routine afterwards. Waist is visible again. My skin is glowing and I don’t feel that 3pm energy crash I usually do. I’ve even survived without coffee this entire time. Which is unheard of for me.
      So I can’t really agree this is some sort of placebo scam. I can’t just be ‘imagining’ my knees just being able to work again.

      I can also ascertain positive effects on myself after taking it - immediately, after 2 months and even now after regular daily intake after 2 years: I can MEASURE my muscle tonus/strength mostly on ring muscles around eyes and palms. That alone, is worth it.
      I am 65+, sedentary, but on intermediate 18/6 diet

      Any benefits are probably from the niacin in the NMN (maybe you're an over-methylator). But niacin is hundreds of times cheaper than NMN. NMN has been conclusively proven to be a scam by Dr. Sinclair. Look for other videos explaining it. This isn't the one.

      I have also had positive results, but this may have something to do with my hypothyroidism, the nmn appears to be bridging the gap where my body is falling short on the T3 & T4.

    This is awesome. I was really looking for the nuance on this. My own experience is similar. No change in blood numbers (maybe increase in cholesterol? or just getting old?) but after 6-8 months, a huge boost in endurance.

      It increased my endurance and respiratory and cardiovascular capacity greatly. I noticed it markedly improved my running time and endurance, especially for long runs like marathons.

    I am 61 and have been taking nmn for almost 4 years now, and love it. My walking speed has increased, I have angina but don't need isosorbide mononitrate as often as I do without nmn. My skin is amazing. I think there needs to be more adequate tests done using correct doses/age, and or longer periods of time. Having said that, nmn alone is not a wonder drug but one of several that should be taken in combination. For example you should be taking TMG in equal doses. I also take supplements such as quercetin which activates sirt6, AKG, Berberine and more. Also, NAD helps generate ATP, thereby restoring mitochondrial function. I've noticed that when I started taking NMN it seemed to go where it was needed most. I believe that more thorough studies are needed.

      I have angina too and am 70 but I think for people like us it may be a placebo effect to get us up off our buts and get us moving.Hope is a powerful thing!

      @@candrad I can assure you it's not placebo effect. My angina was unstable when I started NMN, by the second day I was zooming about all over the place. I had no expectancy from NMN in less than a month. Four years later, my skin is smooth, black 'panda' eyes have disappeared. Thinking has sharpened. You really do need to try it for yourself.

    Improved blood values aren't the main selling point of NMN though. It's decreased genetic age. Any information on that?

    I personally take nmn one brand did nothing for me the other brand did wonders from nmnbio. Pulled me out of chronic fatigue gave me way more energy and helped recovery time from my running. I am over 60

    The main two reasons I have not taken NMN is the cost, and that I personally think Dr. Sinclair is more of a used car salesman than a caring longevity scientist. There are other supplements that are less costly that show benefits, such as GlyNAC and Taurine. Great video!

    What I'm confused about NMN is what exactly is it supposed to do. If it boosts the production of NAD+, then the studies should look at likely markers of success, such as products of 3-way enzymes that require NAD+ as their power source. Why are these studies looking at stupid run of the mill blood test items like total cholesterol and all those meaningless numbers created for the sake of marketing cholesterol control drugs? Who gives a damn about cholesterol anymore? I want to know about things that actually depend on NAD+, and how they improve, or not; and nothing else. Same as if a drug was marketed as a fat loss drug I'd want to know how much fat people lose on it, and not how much cholesterol they got, or whether they had less headaches, or more hairs, or whether their marriages lasted more, or less. None of those markers seem to me related to NAD+, but I could be wrong, of course.

      same thoughts. I have no idea how to measure energy metabolism in the blood - but the classic markers aren't it.
      Maybe more practical tests (like endurance/strength/cognitive) would be more suitable in case there are no reliable blood markers for measuring energy metabolism in the blood yet

    Hey Buddy I am 77 y/o Ex military with 100% disability and have taken NMN for almost 3 years. I do it for health span not life span. Felt energy effects on the first day I tried it. Recent 10 vacation left it home felt tired and weak. If placebo then all good to rock and roll.

    I took NR for 6 months with zero effect. I switched to NMN. I took 1 gram per day for one year. The energy boost alone was worth it. It took a few weeks to reach its maximum. I am a fit 69 year old male. I no longer take NMN because I can’t afford it.

      I'm 45m, and it gave me the energy and optimism I had when I was 10. I kid you not (lol). This stuff changed my life. I've been using caffeine for years, and tried other stuff like pomegranate to keep T levels up. I used creatine for months, and it made a minor difference. But nothing like NMN. I've even tried designer amphetamine some time, and the effect is comparable, but NMN is not only neurological, and you don't "borrow tomorrow's energy".. Hormone production, and metabolism start running full speed again. I love it.

      I didn’t get any energy boost either, but I have a chronic illness so really nothing makes me feel better 🤷‍♀️

    I will be there for the Sinclair study. I just turned 60 and have greatly improved my lifestyle with fasting and am beginning to take supplements introducing them one at a time in low dosages. Since I have always been research-oriented, I research a supplement or vitamin do death before deciding to take it. I even chose a cardiologist as my primary care physician since heart disease is so common. Keep up the good work. My son has assigned one of your videos for his biology class to watch and do further research. They are high school students, but it is never too late to get them interested in science.

    One very important aspect here, it is actually really important which NMN supplement was used. As many other researchers have already found out, internet marketplaces are filled with very low grade or even completely fake NMN supplements, which contain very low level or no level of NMN at all.
    In the NMN community there are only few supplement companies that are trusted and third party lab tested regularly to actually contain the advertised amount of NMN.
    This is really a big problem as so many people actually spend large amount of money on fake supplements, I'm not aware that there is some pharmaceutical grade NMN. And this is true for many supplements, as very few do go through FDA or even third party testing and are verified to contain the advertised ingreditients.
    I'm in my thirties and can confirm NMN does work, no wonders, but strongly notable improvements in daily function overall.
    It works best if taken with TMG as many studies do show taking NMN alone is not very effective.

      Correct, almost everything on Amazon was fake. For me NMN does do wonders, but I see there's everything from non-responders to people that respond even stronger than I do. For instance people with hypothyroidism.

      correct, and many NMN are not stabilized, which means they degrade easily, and when then used actually give a negative effect, not merely neutral.

    6 months ago I was genuinely ready to call it quits. Life was uncomfortable and really not pleasant. In desperation I started taking nmn, resveratrol and tmg. it took a few months, but these days I feel fine again. The difference is fundamental. I really don't care if it is placebo effect or something else - life is ok, versus being absolutely not ok. That's the end of the argument for me. (I am well over 60, but generally in good health, don't smoke, don't drink.)

It's getting better and better in terms of delivery, production, etc. I might start directing my students to some of these videos as examples of paper analyses. You might want to consider a first read "stream of consciousness" approach some time. Takes a long time to get through, but my students find it really helpful to see "what it takes" to analyse a paper.

    I took NR for a month back in 2020 and I started having dreams I remembered again. Also, my memory from watching UA-cam videos and work skills got better too. It’s not a placebo since I doubted that it would do anything. Now starting this year in 2023, I started taking NMN and I feel just as great. I don’t give a damn about how “greedy” Sinclair is ow whatever, I’m gonna keep taking it since I’ve felt incredible because of it

      I really could not wrap my head around the many non-result studies that came out recently.

      Same here. I have been taking Lopo NMN for almost 2 years. My health and energy levels feel great - relative to where I was previously. My friends even comment on how much younger I look (fresher / less tired skin).
      Also, I had a kidney transplant 5 years ago. Over time with 500-1000 mg / day (5days a week) my creatine levels (a measure of my kidney health. & function) has dropped from 1.9 to 1.5!
      My nephrologist is astounded!
      I don’t care what this guy says. I am seeing results.
      I recommend 1000 mg/ day, But dammmm… it is expensive.

      Best to assume health research is fraudulent until proven otherwise. Not my words but those of former editor of the British Medical Journal. Big farmer wants you sick and on a bunch of their pills. So of course "the science" will poo-poo any new supplement which has the potential to drastically improve people's lives. And of course the FDA banned its commercial sale. NMN made a huge difference for me, particularly running endurance. I'll continue taking it.

    I didn’t think NAD+ precursors like NR or NMN would do shit. But after taking it for a few weeks, my sleep dramatically got better and my memory is better too. I’m gonna keep taking it

    I am 74(and kidney transplanted since I was 60)and been talking nmn plus a few of the other supporting supplements and I must say it has increased my energy and well being in an unbelievable way! I believe in everything Dr David Sinclair says! Any critics should demonstrate double research time than what he did before I change my mind..

    I had golfers elbow for a couple of years. It wasn't healing and my ability to exercise with my arm under load was very limited.
    I took nmn out of desperation and within a month my elbow was much improved. Within two months totally healed. For me it's an absolute miracle drug. I'd tried everything to get that elbow to heal and nmn was the key.

    I don’t know BUT my doctor told me about it. Said he’s taking it and I should try. I bought a reputable brand and only take 125 mg a day. I’m 71 years old and I’m suddenly FLYING. Amazing energy and focus.

    I’m wondering if any study that includes diabetics or pre diabetics is a fair study of any supplement. Diabetes inhibits uptake of a number of nutrients if I’m not mistaken, so adding the extra burden of having to be a “magic pill “ that overcomes a bad diet seems to be an unreasonable expectation.

    Thank you for this video. I'm glad there is someone out there keeping an eye on the scientific literature to keep everybody honest.
    I'm currently taking NMN every few days as part of a rotation of supplements I take. But this is in addition to a good regimen of exercise, sleep, and a healthy diet, free of ultra-processed foods.
    I don't know if any of the supplements I take will make any true difference in longevity. However, since animal studies have discovered some pathways can lead to longer life in animals a few of the supplements I'm taking might move the needle in some way and give a few extra years of health in old age.
    I'm keeping an eye on the research, and just hedging my bets until they find what really works. My bet is being active and having a good diet will move that needle more than any supplement, but you never know.
    One thing I know for sure. Those people out there who think they can eat unhealthy ultra-processed foods and sit and the couch all day and just take one pill and live to 120 are wrong. It will take supplements plus a very healthy lifestyle to have any chance at a healthy old age.

    Thank you, Nick, great video. I've been taking a quality NMN product (3rd party tested) for the last 6 months. My dosage varies from 500mg to 1g. daily, depending on my financial circumstances that month. I have never felt that it's improved my quality of life either physically, mentally or emotionally. My background is: I'm 73, on no medications, good blood work, great A1c, 14% body fat, lift weights 5 times a week, walk 5-6 miles a day and consider myself a (relatively) good male specimen. Protocols such as NMN may be beneficial to individuals with some medical problem or deficiency, but it's apparently not that beneficial to everyone. Thanks again.

      Taking NMN alone was never ever the solution. It seems no one on here was paying attention to David Sinclairs protocol. Without something like resveratrol NMN simply cannot complete it's job

      Hi Paul, thanks for responding. You bring up a valid point, NMN may not be the whole picture. I commented on my NMN experience because that was the topic, but my personal protocol is much more extensive. I have been taking Resveratrol for some time. I've taken Spermidine and Quercetin for extended periods in the past. My personal health protocol currently consists of some 37 different vitamins, minerals and supplements, which I can post on request. My point was that for many people, there is no "Magic Bullet", no wonder drug. @@paulharrisonadventuregearm5457

      @@carlocroma1093 Carlo, I started taking Renew by Science and did that for 6 months. Then I switched over to DoNotAge because it was on sale, and I've been with them for the last 6-8 months. They're both high quality, top shelf products, but I haven't felt anything outstanding after over 1 year of use.

    What fails to be said is that there are a number of people online who have conducted their own experiments and found quite substantial changes in the epigenetic outcomes. Endurance, brain fog, blood sugar, weight loss, eye vision correction and much much more. I've found studies staying substantial changes. Furthermore, David Sinclair said you must take resveratrol at bare minimum to get the changes NMN in regards to NAD+ effects. Taking NMN alone is effective but not nearly as effective as when combined with resveratrol. I can also vouch for using NMN alongside resveratrol in that it reduced my cytokine levels and took away my brain fog as well as giving me extra energy. NMN won't work unless resveratrol turns on sirtuins which then allows NMN to do it's miracles. These studies seem to forget that that is how NMN works

    Thanks for this. I started taking NMN a few months ago largely due to some of the claims by Sinclair and others regarding anti-aging. This gives me pause for thought, since I'm generally in decent shape and the supplement is _extremely_ expensive for the amount you get. Looking forward to that next video.

      also, from another angle, he straight up does not seem healthy, he seems frail, sure he might live long, but who wants to spend the last 50 years of their life with poor health like him? i know plenty of people who are way healthier than he is at his age using just simple diet and exercise, strong, attractive and full of energy.

      Dr Sinclair seems to me being too much of a spindoctor. Everything he does seems to be up in the air. He isn't the kind of cautious researcher who base his talks on actual research results. But more on research ideas.
      We all know that epigenetic changes has to do with aging. That's what epigenetic clocks tells us.
      But just using Yamanaka factors to change it is a wild dream. We don't know how that will end.
      Dr Siclairs talk about a "backup copy" might be just wishful thinking.
      Extraordinary claims requires extraordinary proofs. And Dr Sinclair does claims so much better than proofs.

    1) NMN should be applied to those that need it:
    - old people or with equivalent mitochondrial disability / permanent effect of poisoning from diverse sources (long term bad diets, carbon monoxide poisoning, etc.)
    - athletes
    2) NMN needs to be legit and of 500 mg/day dosage
    If you want, supplement TMG alongside it although I don't think it's necessary if you don't go above 500 mg.
    If you don't feel the effect within 30 minutes of your first dose, then you probably don't need NMN.
    For me it made a huge shift in well-being 2 years ago. Got rid of chronic fatigue and anxiety, along with poor body/mouth odor. That is not remotely possible from a placebo and the impact to my wellbeing was massive. I very rarely skip a day but the effect is persistent over at least a 3 day period.

    Started taking NMN @ 39 for about 9 mths now. I take it with TMG. What I've noticed is a boost in motivation, mood and no pain post exercise. I can exercise every single day now and don't need a recovery day. I've always been sore the next day after exercise since my 20s but not anymore which is one of the more odd benefits. I had also got glasses for the first time @ 38 and no longer need them so it would seem to have had some effect on macular degeneration too. I think the best bet is try it, if it does nothing for you don't use it - It's always better to stay open minded about these things.

    Thank you for the video. The supplement is often touted for promoting longevity. While it boosts muscle energy and improves alertness, somewhat like caffeine but with a different mechanism, its true benefits for increased physical activity require further study. Notably, some UA-camrs haven't observed significant improvements in long-term cardiovascular health markers. Overall, it seems beneficial but might not live up to all its claims.

    Can't wait to hear your take on Sinclair's latest study. Thank you for this one. I've been taking NMN for a little over a year. I had my biological age tested before starting and am about to retest to see if it's made a difference - although I went down the longevity supplement rabbit hole, so if it does show an improvement I won't be sure what did it. With the controversy involving David Sinclair both with Resveratrol and NMN, I'm getting an ever increasing "sketchy" vibe regarding him.

      I take NMN with TMG and resveratrol. I've started cutting back on the resveratrol as I read a few negative thing about it. I kinda went down the rabbit hole too. I started with gynac and quercetin about a year ago but started taking NMN + TMG+ resveratrol. I felt immediate benefit from that stack (perhaps too much. My libido a bit wild and I couldn't sleep well). I stopped taking that stack for a month and when I started agao (about 3 weeks ago) I never got the same 'hit' but I do feel benefits in general aches and pains that have died off and still feel some energy boost... libido is back to normal :0( lol

      @@Tony-un3vf Dr. Brad Stanfield has a video titled "Resveratrol - The Unfortunate (& Scandalous) Story" that goes into detail regarding the back story and issues. One of the biggest takeaways from this video for me is that GlaxoSmithKline paid I think 750 million to obtain the rights to create a drug using resveratrol. Then they spent who knows how much more testing it. And in the end they chose not to pursue it.
      Also, how it effects your cells is dose dependent according to another doctor. His channel is "Wellness Messiah". (I'm honestly not a fan of anyone calling themselves a Messiah, that whole god complex thing, but he does deep dives into the papers and research so I let that slide.) He gave an analogy using a laptop that is broken. Do you fix the laptop in hopes it lasting a while longer, or do you replace it. Each decision has it's pros and cons. Resveratrol in smaller doses repairs the cell. In larger it doses facilitates replacing the damaged cell. The former better when you are younger, the latter when you are older. I kept going back and forth on which I should do.
      Also Resveratrol can blunt the positive effects of exercise. Hell to the no on that! Lol. In the end I decided that there were other, better researched supplements that would help me achieve the health span benefits I'm looking for. Good luck Tony!!

    NMN has had an amazing effect on me. I am 51 and now play three games of field hockey per week. Since taking NMN, I never have any aches or pains from sprinting around the field. I can run faster and longer, and hockey ball impact bruises take two days to clear, not a week. I feel like i want to run every day, i have spring in my step, my mind is clearer. Never have i felt so able bodied. It literally feels like my body is 28 again. When i turned 33 my metabolism slowed down. I had to eat less or exercise more to control my weight. That has now been reversed. Best thing I have ever taken. It took about 3 weeks before i noticed significant changes and now 3 months in, I feel great.

    I’ve been taking it on and off for almost 2 years… started after reading Sinclair’s book. I notice more energy when I’m on it- need less sleep. Biomarkers have all improved but can’t put that down to NMN because I made across the board changes after reading that book. Won’t throw the NMN out yet!

    I take NMN for the extra NAD+ and saw an increase in general energy and ability to exercise, one that NR alone doesn’t seem to provide. I speculate that this may be because the half-life of NMN in the blood is rather long compared to NR. It’s not a holy grail molecule, but that’s an issue of inflated expectations, not necessarily effectiveness: if it does one thing and does it well, who cares that it doesn’t also do 99 other things.

      Completely ancedotal, but I am a physician and have used nmn for about a year. I have seen improvements, then I started my wife and my in laws on it. We all had similar provements in energy levels. Will it help me live longer probably not, but it has improved quality of life.

      That's the basis yes. But creatine for muscles (and you'll also need them for longevity), caffeine can also be used. Plenty of other stuff as well.

      On another note: Mother Nature designed everything on this planet to function in sync. Food for all species is meant to be consumed raw, in its living state. Living foods have enzymes. Cooking or heating foods over 117 degrees kills all enzymes. Food without enzymes is dead food. Consuming dead food over a long period of time robs us of our enzyme potential. This creates premature aging. Metabolic enzymes which are designed to repair the body, have to be converted into digestive enzymes to digest dead foods. This causes bodily repairs to be placed on the waiting list because we eat 3 meals a day, not counting snacks in between meals.
      Dr. Edward Howell stated that when man discovered fire, disease was born.
      Instinct tells animals what they are supposed to eat. Humans bypass instinct and have to read books in order to determine what our natural foods are. We are also imprinted with all the massive propaganda generated by the food industry telling us what we should eat to stay healthy. The meat industry wants us to drink more milk. The meat industry along with the cattleman’s association wants us to be carnivores and eat more meat. I guess we should be weary of scientific studies paid for by industries. 🤔

    Doesn't vitamin B3 itself does better job than NMN for boosting NAD ?

      Apparently it does in some individuals. This could be due to the fact that B3 is a precursor and can help other deficiencies further down the line before NAD Plus synthesized.
      For the same reason, it may be less effective in others as their natural deficiency may be after niacin but before NAD.
      If any of them work at all, it appears to be highly variable on the person. This data sure is eye opening though. Sinclair is going to put a hit out on our boy here 😂

    I first became aware of nicotinimide back in 2008. My ex girlfriend' s cousin was doing research at Berkeley on the role of Nicotimide in ameliorating apoptosis, the induced cascade of cell failure and damage caused by reperfusion after ischemic stroke. They were examining the role of PARP 1 and it's interaction with nicotinimide noting the positive effects it has in limiting cell damage and death in a ischemic reperfusion scenario in vitro. I didn't know anything about all this but I googled his name and research before we first met so that I could maximise the opportunity to learn and have an interesting conversation when we met.

    I'm just throwing in my experience. I always approach new supplements skeptically. What do I take? Mostly things I know with certainty I am low on. Supporting my immune system is number one, followed by everything else; I know what it is like to be in full autoimmunity.
    Sample of one. 😅 I am 66. As I said, my endocrine system was failing due to autoimmunity from food allergies and that was discovered around 2003, so I have had time to recover as much as possible. I also had pretty substantial back injuries in1994, so I experience chronic pain daily and have since. This definitely drains my energy, so I was willing to give NMN a try. I am a very analytical person; everything I take is based on studies, generally. NMN was pretty new, and my KREB cycle suffers, and given it was a precursor to NAD I decided to give it a go anyway.
    I began NMN about a year and a couple of months ago at 65. For two months I saw zero effect. It wasn't until month three that I began to see a difference. I don't know what is behind that delay, nor would I speculate other than to say through tests I know for a fact that my KREB cycle was highly compromised. The difference I noted during the third month was substantial. I take a gram a day plus Glycine, both are in raw powder/granular form. Now my wife is taking it as well.
    Here's my main observation: people who suffer from low energy and have difficulty getting through the day are going to benefit. I am not at all surprised that all of the measurements taken regarding blood tests showing no hard improvements in cholesterol and other significant aspects like blood sugar - it's more related to cellular energy than anything else. Insulin resistance gets handled through other means. NMN was never this miracle end of aging for me; I just was looking for improvement in energy, and there is zero question that it provided that. For me the difference is pretty stark; I see it as improving our life quality experience as we age. Some young persons do suffer from low KREB output, and in that case, I am pretty sure that NMN will benefit them as well.
    For those who have chronic back pain where overactive neural systems in the lower back are involved, I suggest looking into tests done on Thiamine and Augmatine Sulfate. I use Benfotiamine, a lipid soluble form of Thiamine. This is NOT a pain reliever; it calms the overactive nerves. Look into the studies and understand it before deciding.
    I have had a lot of years understanding my pain condition and methods for getting relief. The pain sources have independent characteristics and therefore independent treatments. Inflammation, neural pain (sometimes caused by inflammation), neural pain caused by a chronic condition where the nerve becomes overactive and the brain responds, and muscle spasm.
    CBD (which is not yet widely available) works really well for inflammation. I already mentioned treatment for overactive nerves to calm them. For spasms there is no substitute for magnesium. I take Magnesium twice daily and recommend that regimen for anyone who suffers from Spasms. I also use a cream that has magnesium in it for cramps - Hyland's leg cramp cream. It works well on large back spasms. Of course, heat also works and other methods like far Infra-red treatment like therapeutic saunas using far infra-red.
    For me, anything I consume is backed by research generally. In the case of NMN, based on the studies showing results by indicating normal bloodwork without improvement really is focusing on the wrong areas. Such studies can have the effect of making future studies less likely to get funds, which is not good. I am not sure why those were not just preliminary studies to understand and narrow down where the NMN WAS actually beneficial.
    Personally, I don't see the benefit at all to looking in the wrong place and concluding based on that data, nor do I think it is beneficial to bring those results to the public, generally. It becomes decisive for some who might otherwise benefit from it. If you are older and suffer from really low energy, I think you are a solid candidate for NMN. If you are younger and suffer from low energy, I recommend giving it a try. But if no benefit, please get in to see a doctor; some of these energy related illnesses have other causes. Get your endocrine system checked out thoroughly. If your numbers are low, get tested for food allergies and antibodies. Don't assume anything; use test results.

    Hey Physionic. Thanks for the video! Can you maybe answer me this: Isn't the story that NMN could help protect your DNA? If telomere attrition and overall quality of DNA is the goal, would you see it in the markers from your video? With my limited knowledge, I would think it's hard to test without measuring lifespan.

    Thanks for this vidéo. Please note that I take 1g of nmn per day since 3 month. I alternate one month with, and one without.
    I therefore take 1 gram of MMN and 500 mg of resveratrol every day, alternating every other month. After a few weeks, my vitamin D metabolism was 100% restored. For 20 years, I took vitamin D supplements every day to compensate for the fact that, despite sun exposure, my blood level always remained too low. Since I started taking MMN with resveratrol, this metabolism has been restored to 100%. It should be noted that I wasn't taking MMN for vitamin D, as I didn't know it could affect this metabolism. It was a big surprise! Just wanted to share this info ;-)
    I'm 58, tall, healphy, and I run 50km every week.

    Very interesting. But I'm a bit confused by th conclusion. NMN has shown to increase NAD+. Let me try to analyze myself what the studies now really showed: Some alternatives that are possible ...
    1. Possibility: Rising NAD+ has no significant effect or low NAD+ levels are compensated by something else for olders
    2. Possibility: Rising NAD+ has a long term effect, not measurable in the studies
    3. Possibility: You need to rise NAD+ "really" much to come to a measurable effect
    4. Possibility: Rising NAD+ has an effect on parameters that were not measured (like cell-repair), and the measured parameters have no causal relation.
    So, as I understand, we are talking not about NMN but about compensating NAD+ reduction for old people, and if it helps?
    Is there any data available, that helps uns to chose one of the explenantions above?

    My 58 old Mother health dropped dramatically since she's got injected in 2020, especially her memory .. almost lost all hopes she will ever recover.
    After giving her NMN at 175 mg a day for just over a month now, she is literally BACK TO HER FORMER SELF.
    I don't care about studies and yadayada that you say, I look at FACTS my eyes can see, this stuff WORK!
    I ordered more today and plan to start using it my self as well.

    Thank you for the excellent video on NMN.
    In your next NMN video, I am especially interested in hearing details any energy benefit of NMN.
    Don’t care about extending life span past the normal 80-90 years. Just want to have energy to do things while alive. Because lately, my energy level has become worryingly low.
    (My comment got way too long. I understand if you do not read it)
    Why I am so focused on energy level:
    Age 64
    Until recently, I believe I have been in better shape and more energetic than ~75% of my cohort.
    Lifelong runner, mostly chip trail running, have ran marathons, Off and on weight training. Physically active job.
    Constant seasonal recreational activities: Snowboarding, snowshoeing, windsurfing, kayaking, canoeing, biking, hiking…
    Not that I did something every day, but pretty active.
    Took Chemistry in university, including Biochemistry of Nutrition, and have tried to stay informed in order to eat well.
    Diet is varied, but most of the time: Eat not too much, mostly vegetables. Some eggs & fish. Red meat is rare, but perhaps a little more of it recently. Have done occasional fasting, but seems too difficult lately.
    Blood work: Don’t have the numbers, but doctor says everything is very good. Thoracic CT scan says my lungs are good.
    I realize I am aging, but still disturbed about my energy level dropping so significantly, lately. So I am very interested in any supplement that may help boost my energy level; enough that I can continue to do some sort of interesting physical activity.
    I am not very social, so most life satisfaction, even if with the friends i do have, involves some sort of physical activity.
    Without that, I don’t care if lifespan is reduced rather than extended.

    I bought NMN a few times because I was hoping for better energy, but my final conclusion is that the effect was placebo. It has a really fresh lemon taste which I liked, but I think the effect from NMN will be the same as eating lime or lemon water, they also offer reduced bloodsugar by the way.
    Instead of NMN for NAD, I feel that B3 Nicotinic acid (Niacin) has way more positive effect and short term energising.

    My observation at 60 is a hammer toe where I had a injury healed giving. Flexibility rotator cuff injury healed knew injury don't hurt and jbee is stringer and after 4 months taking 1500 mg ( with bit d plus k multiple vitamin quercetin. ) generally feel younger

    I have had dry and scaly skin at the hind foot. No matter how much water I drank and cream I applied, it’s not improving. 6 months after taking nmm, I noticed a side effect - scaly skin disappeared! I don’t care what’s in your chart, my skin knows the difference.

      The problem with that, much like with faith healing, stuff changes over six months. Perhaps if you had the condition for five years prior? The only real evidence is speedy recovery, and the condition returning if you stop taking the product. This is how I discovered glynac helps my lungs. It's not why I took it either, but if I stop taking it for a few day, even two days, I start coughing up phlegm like crazy. I don't like depending on it, but it's unlikely to be placebo. Another example is vit K2. I took too much originally, and my varicose veins started leaking in my leg. Horrible condition. After strong steroids etc it came and went, till I realised I was OD'ing on K2 cos it was in two supplement I took. Cut back, condition cleared. Accidentally took too much, condition returned. Man I'm so relieved to have spotted that one, it was horrible. Couldn't wear long pants for a year. Anyhoo, you get my point?

    I don't think You should draw conclusions that fast. Increased muscle strength and physical endurance are not placebo-effects. You should consider this signals: 1. Higher NAD+-Levels in Cells through precursors like NMN or when fasting through katabolism and 2. Higher Oleic Acid-Levels through fat-katabolism (when fasting) or by consuming Resveratrol, Pterostilbene that lead to improved fat-katabolism - or throught eating Oleics. Those signals (and more) might lead to Sirt1-Activation and that is the thesis of slower aging or reverse aging. Does those studies evaluated this - no!? Or when taking NMN your cells might have more energy to produce things and speed up their production. But like in a factory, when production runs fast, the supply-lines of materials for that production have to keep up with the new speed or the new speed-up will be slowed down again by material-shortage, or even worse when materials (maybe Ca²⁺) get raw and critical for other functions and lead to sideeffects. If You want to draw conclusions on NMN You should look at studies that consider those effects. Did those studies here evaluated this - no!? What a waste of time - no, just a joke.

    Started taking 1g of NMN and TMG from Do Not Age just over 3 weeks ago for £235 (6 months worth) and felt no effect, so I then purchased 1 months supply of Apigenin for £38, thinking it must be a problem with CD38 gobbling up the NAD and the apigenin would inhibit that. Been taking that for a few days now and still felt no effect. Now thinking that I'm chasing Unicorns. I am 66 years old with life limiting angina, so not too healthy to be unaffected by this stuff...if it works. I wish this video had been made a couple of months ago.

      You could try niacin which is much cheaper alot cheaper and TMG as well. But careful niacin causes flushing.

      You are supposed to take it with an Oleic Acid... {Olive Oil] Originally Sinclair said Reservatrol should be taken with it but that proved to have side effects. Oleic Acid serves as a precursor/activator.

      @@fatboydim.7037 I tried Niacin for 6 months before this lot and I didn't mind the flush, but it didn't improve my condition in any way. Having said that, I did notice that while I had the flushing effect, I could walk marginally faster, but it didn't improve after months of taking it when I didn't have the flush.

      @@billbimson2408 I watched loads of his podcasts and i bought his book. In the book, he said to take it with Reservatrol. But later on a podcast, he said there were some downsides to reservatrol [ i can't remember what they were] and he took it with Olive Oil approx 10ml..Probably if you google it Sinclair, Olive oil it will come up. According to Sinclair, you need this activator to make the NMN work? It's all very confusing. But i hope Mr Pysionic reads this had has a look at it. Also, i watch Modern Health span from time to time and a Chinese Scientist guest said that after you have taken NMN for a few months and the NAD levels are raised you can take a break for a few months without them falling.???

    I’m 63 was born with a genetic inherited blood disorder called hereditary hemochromatosis. It went undiagnosed for decades until I was nearly dead. By the time it was diagnosed and treated I had damage to my heart damage to my liver, diabetes which is also related to it, Hashimoto’s thyroiditis and autoimmune phenomena. I have been experimenting With various supplements to see what would help my labs look better and me feel better with more energy. Over two years ago I started experimenting with MNM. I would stay on it for six months at a time and go off check my labs check my activity levels which are very low due to all the damage. Then back on it etc. etc. So I have come to the conclusion through years of experimenting with it that I do better on it. I wish I didn’t because it’s an expense but as a disabled widow I can’t afford. But anything to help me get out of the bed is important, it’s a quality of life issue. I also have spinal injuries and inflammation so a lot to deal with. Unrelated but it goes along with it I have also found the best anti-inflammatory anticancer diet that goes along with it. So I’m still alive with severe liver disease my liver works but not always. Sometimes it doesn’t work and I have to really figure out what I could do to help it. So anyway that’s just my personal experience with NMN. Apologies I use the voice to text and pretty shoddy language

      @@saliksayyar9793 Yes I’ve had over 100 units/pints of blood removed in 30 years. It doesn’t repair the damage, keeps some damage under control but not all. Tomorrow morning, I have labs to try to find cancer cells in my blood. We suspect the entire liver may be turning cancerous, like my uterus did.

    Well that was a bit disappointing, but thanks for the time you took to read the studies. I thought there would be a lot more. As a fairly healthy and active 70 yo I have found a positive effect. Even if it's mainly placebo I do have more energy. I'm more interested in Dr Nichola Conlons approach at Nuchido which is to restore the salvage pathways that enable the body to recycle the NMN from the cell back into circulation to be recycled rather than just adding a bucket load more. I'd be very interested in that since their studies show a massive increase. Well, I await with bated breath the Sinclair expose !

    You mention NMN a lot - Sinclair seems to suggest that it's only useful when taken with resveratrol, but i havent heard you discuss this. would you consider discussing / studying the benefits of taking both? Apologies if you have in fact discussed the idea of taking them in tandem. Keep up the good work!

    I used to love taking naps. I haven't taken a nap in three years, nor have I even felt inclined to do so. I've taken NMN for the past three years.
    Very little else has changed in my lifestyle.
    I'm not saying Sinclair isn't up to something selfish, but I just wanted to share this, for whatever it's worth.

    Thanks, this is extremely helpful. I was trying a high dose NMN (900mg) before working out, and it made my post-workout brain fog about 3 times worse. It took me a while to figure this out. I do think it has some sort of mild stimulatory effect, which could explain the walking result even though it is not making any blood markers improve.

    From what I understood, the increased NAD+ levels were theoretically supposed act as a coenzyme for SIRT 1. Improving longevity by reducing DNA damage over time. Is there a study that looks at NMN and the gene expression associated with DNA damage repair or epigenetic regulation?

    Thanks for the great video, I really enjoyed it. But, from what I recall, Sinclair's main claim about NMN and its potential for life extension is its effect on sirtuins, and PARP. Specifically boosting sirt6 and if I'm remembering correctly blunting PARP. Did any of the studies you read look at those effects?
