    This weekend will be the final setup of my home TC setup. 🎉 Pennsylvania winters are my biggest worry; last winter we had -40°F wind chill and everything in my house froze. 😕🥶 Pray for a warm winter, please! 🙏

    Take care while working with flame, cleaning and tools in alcohol.
    I put fire inside.
    Also the flame risk to melt the top of the plastic container.

    bathrooms are wet, wet is good for molds, molds are a contam risk. probably better not in the bathroom. kitchens have a lot of bacteria so also not the best. a good procedure with stil airbox etc however allows you to work in absolute filth :D the size of the space and thus draft matters a lot in this. watch out with stil air boxes SAB using open flames and cleaning products that are flammable ;), lol okay im still watching and kudos on that demonstration . i did see a procedure mistake here, when you use a S.A.B you want to organize it in such a way that you never have to reach over your work. i seen that hand hovering over the medium going for the burner, it would be better to set it in the center ( the burner also cleans the air btw by burning off any contam, bunsenburners are sometimes used to create a clean air bubble). this hovering over things can cause things to fall from your hands or arms. so always plan out the steps and set up things so you dont have to reach over things. work front to back instead of left to right. i always imagine the center being the place of work and the sides to be the hand moving area and the back corners are storage

    thanks for your great work. i have 2 grams of meta toplin and need to know how to make a stock concentration and the best solvent to use can meta topolin be autoclaved? Can meta topolin concentration be stored over time?

      Add meta topolin in the media after it's cooled down using the syringe filter technique.

      We didn't try orchids in our lab. But, you can easily find the protocol for the plant on the internet as it's one of the extensive tissue-cultured plants. @@Unkn0wn1133

      There are a bunch of vids on YT that teach acclimatization for TC. In my opinion, having a good humidity dome with vents is the best way to start.

    Is there a difference between 70% and 90% alcohol

      70% alcohol is more diluted having 30% of water, whereas the 90% is more concentrated with 10% of water.

      the higher % decreases the time it sits unevaporated which reduces the time its killing bacteria, it also helps getting in to the cells to have some water in it. be aware tho as alcohol isnt all that good at killing mold spores, unlike bleach solutions

    I'm going to try growing some Japanese red maple seeds in TC this winter. Is the protocol different for starting seeds? I read that tree TC usually uses 1/2 MS amount but no mention of seed protocol. 🫤

      Yes, generally protocols differ based on plant species and even the explant used in the process. We haven't tried the species yet in our lab. But, would love to get some update from you if you are going for it. Good luck!
