    Francisco, where do you get those containers? The ones we got from Karat melt in our autoclave.

    How important is it to make media and sterilize it right away? Would it affect the media at all to leave that jug in the fridge for a day or two (and hope nobody tries to drink it 😅)

    Hi. I have a quick question. Can i take a small tissue culture plant with roots puchhased from ebay and put it a biocoupler or would there be issues

    Giving this method a try now. Thank you for sharing. Any concern with using RO water? It comes out of my filter right around the target pH.
    EDIT - I re-watched the video, you mention reverse osmosis is OK around 0:55

    Can you please explain that the ms media that you used in the video is in the powder form, but I couldn't find it in powder form on your Plant Cell Technology's website. On the website, MS Basal medium is available in liquid state.
    So, my question is, is there any difference or just same

    Does plant cell technology’s ms media not have sucrose? Oh no my plants I just TC’d might be In Trouble 😢

      Uh oh! Yes, our media doesn't contain sucrose. It's mentioned on the website and will also be written in the manual. "MURASHIGE & SKOOG MEDIUM (W/ VITAMINS & CALCIUM CHLORIDE, AGAR)"

    I have a question. Do you use the skoog with agar? Or do you use it by itself?

      for initiation phase, it's always media with agar, but at multiplication and rooting phase, that we perform in Biocoupler, it's only media with hormones, without agar.

    But i've a qu...
    I made 100ml of ms media..use agar about 0.8gm .but it didn't make solid ...why is that..??

      You must not have mixed the agar well with the media. You need to ensure that you mix it well before sterilizing and then swirl the media in the bottle a bit before pouring in the vessels.

    Will my plants die if I did not add sugar to the PCT media?

      Yes, sugar is the source of carbon for plants. So, you need to add it to the tissue culture media.

      PGRs are added at the multiplication and rooting phase of plants. Generaly, the initiatin media doesn't involve PGR. But, it totally depends on the protocol you follow for your plant.

      Solid media results from adding agar or gellan gum (a solidifying agent) to the media.

      It's easier to buy pre-made media...
      Perspective, tools-on-hand, and budget are all factors that determine the meaning of "easy."
