You can kill the guide, but you need to get his voodoo doll first, which can only be obtained by digging into literal hell and killing a demon carrying one. If you equip it, you can hurt him. If you drop it in lava, it kills him instantly, but watch out. Alternatively, you can craft a flymeal, but you need 5 stinkbugs, a gold/platinum broadsword, and be near honey. Honestly one of the most trapping things about Terraria (which i'm sure you've experienced part of), is how so many things, progression and exploration too, tie together in a domino effect to become a potent greed and curiosity trap. A distant chunk of ore that leads to a underground house, a brightly-lit skeleton miner attracting you like a moth to then reveal from the corner of your screen a new chest and relics to find. Whole minecart tracks that can send you through several resource and even item/ biome-filled areas, which in turn can lead to MORE minecart tracks and biomes. And then as part of your glorious rewards for curiosity, the game flips you the bird and mulches you into a boulder-splattered paste or explodes you across the walls with the compacted force of a nuke in a bomb trap. Mmm, i like my spelunker dream with a side of agony and despair.
