What I really love about this somg is that it sounds like Curly and Quote having a discussion about what to do, how to proceed onwards with their endeavor and how uplifting it is.
The way this song is presented is pure genius. You hear the song for the menu throughout the entire game every time you boot it up, and then when you finally hear it at the plantation near the end of the game you get a feeling like "OH SHIT, THIS IS WHERE THE REAL GAME STARTS!" Everything that you've done, everything you've overcome, leads up to this moment, the final preparation before saving the island, and, consequently, the entire world. All your progress is embodied into these next few battles. Do NOT screw it up.
I love how Pixel placed this song in the game. You've just been in this very contemplative state on the outer wall, where you have the choice to stay and fight at impossible odds, or leave and abandon those of your friends that remain. Even if you choose to stay, the climb up the wall with the very peaceful Moonsong playing gives you plenty of time with your thoughts to consider if you're doing the right thing. And then, when you persevere and make it to the top, the game rewards you with the title theme, almost as some sign from Pixel that you made the right choice.
