  • Ever wondered if you can substitute one plant hormone for another due to availability issues?
    Let’s break it down:
🔬 In many countries, synthetic hormones like IBA or NAA are restricted, leading researchers to opt for natural alternatives like IAA.
Here’s how you can make the switch effectively:
✔️ Prepare Your Media: Sterilize your medium first, using a pressure cooker or autoclave.
✔️ Cool Down: Allow the medium to cool to 55°C.
✔️ Add Hormones: After cooling, add your plant growth regulators, such as IAA. Ensure these are added only after sterilization to maintain their efficacy and stability.
✔️ Sterilization of Hormones: Use a 0.2-micron filter to sterilize the IAA solution, ensuring it’s free from contaminants before adding to your media.
📌 Key Tip: Always use sterile techniques when adding hormones to prevent contamination.
🎥 Watch the full video to see these steps in action and learn more about hormone substitution in plant tissue culture.
This knowledge is essential for adapting to regional regulations and availability of materials.

Hi Francisco, have you ever induce flowering in tissue culture, especially with woody plant?

Pero si colocas auxinas cambia el pH, que recomiendas hacer para no volver a usar un potenciometro?

How unstable is IAA? Is it possible to compensate by lets say quadrupling the dosage? Or is it completely rendered useless under such high temperature?

Do you have a video where the filtration process is explained in detail?

We do not, but it’s a topic we will cover in this weekend’s master class 🎉

I got all the detail by watching the example with the .02 micron filter + syringe and his explanation. All in this video alone. . It is essentially filtration by reverse osmosis.

Can you tell me about tissue culture anubias nana petite what dosage of medium is needed
i am trying many times but all failed, the plant does not sprout and grow with:
    Ms: 2.4g
    Kinetine: 0.15g
    Agar: 4g
    sugar: 15g
    look forward to your feedback

1/2 Ms media for aquatic plants is okay. However, 0.15 g of kinetin does not make sense for the hormones. If you are actually adding 0.15 g of kinetin, that is way too much.
0.15 g of kinetin is enough to make 150 liters of media!
At the same time, Kinetin needs an auxin to work, something like IBA, NAA, or IAA.
