Thin layer chromatography (TLC) principle explained

  • This lecture explains about the thin layer chromatography technique. TLC principle is well explained with example. Thin layer chromatography is the easy method to separate different types of chemical molecules from each other with the help of thin layer chromatography plate also known as the TLC plate. Thin layer chromatography separates molecules based on the differential movement rate in the tlc stationary phase which is calculated with the retention factor or Rf value.

    if i were to compare polar and non polar solvent, and one of it has more spots than the other, what does it mean? how to determine if polar solvent is better than the non-polar and vice versa?

    You also needed to explain that some plates won't show up in UV light so they have to be sprayed with whatever spray is needed. Also, the Rf value isn't for quantitative analysis. It's used to ID the sample against a standard.

      Also in some cases IODINE is used. So that the compounds absorb iodine , which will show up as Brown spots.
      Also Ninhydrin solution for detecting Amino acids

    So the question is why does polar compounds interact with silicagel (also very polar) stronger than non-polar compounds?

      This is because silica gel is also a polar substance. Hydrogen bonding is responsible for the polarity.

    what is the effect of the similarity or difference polarity of the mobile phase and the solute we are computing; does it make a difference. i think i did not hear you saying about it except you said about the polarity of the stationary phase versus the solute.
