Making tissue culture media is the easiest part of the micropropagation process because it doesn't need to be done in sterile conditions, since we autoclave it. Be careful with the pressure cooker. In the future, I do plan to make a video specifically about using a pressure cooker as an autoclave.

The protocol that you follow will specify the optimal temperature! I don't have the AC in my home lab zoned separately from my home, so these plants stay around 72 - 75 degrees, but some protocols will call for a slightly higher or lower temp.

Very helpful! You just earn a sub. I have questions. For tc jar, is it necessary to have small ventilation hole? Ive seen some use air tight glass jar. Which one better? Do tc plants even need ventilation?
For the multiplication and rooting stages, you want the jar to be completely sealed with no ventilation hole to maintain 100% humidity. To prepare the plants for the final stage of tissue culture, acclimatization, you may change the lid to a vented lid, to gradually acclimate the plantlets to lower humidity before you take them out of tissue culture and plant them in soil.

Some plant growth regulators are slightly degraded when autoclaved, but it is ok. Antibiotics on the other hand, should be filter sterilized and added after autoclaving.

Yes. I have a special container for disposing scalpel blades but the pipettes get thrown in the normal trash

It's called VIVOSUN 3-in-1 Digital pH Meter with ATC, ±0.1 pH Accuracy on Amazon. I calibrate it every time I use it. This one doesn't get stored in kcl. I will probably replace it within the year though, they are known to become inaccurate after awhile.

So you are using 1:1 auxin and cytokinen? Does it has particular goal with the ratio? Thankyou.
I was just following a stage I african violet protocol that I found in a book. Different plants / stages of tissue culture will have different ratios though

Usually… yes. But I’m pretty sure she uses kinetin as her cytokinin of choice. While it’s not 10000% harmless it’s naturally derived and a lot less harmful than say BAP etc. I personally don’t even want to be in the same house as BAP.
@@andrewdarland8600Why? BAP isn’t radioactive. It’s not some deadly poison. It’s no worse than the other common PGRs.
