But with plant growth regulators. For mammalian , more hard, more growth factors & incubator (meatcubator if your the thought emporium) And you don't need sawdust (unless you want) But yeah, it's all very similar(sterile technique) ...till you get to 3d scaffolding and continuous process fermentation with industrial optimization and more big words.... Then some other smarter individuals have to describe that.🤪
Hi, my name is Dave and I'm an alcohol lab fire starter. Great tutorial! Thanks for putting that together. For me, straight surgical scissors do a great job (iris gum 12) I also use forceps but that's because I'm clumsy.
7:20 100% alc evaporate not just faster, its also dries what you want do kill like mushroom spores needs to get hydrated, befor killing in Mushroomcultivation you let the substrate sit for some houres to incubate what is in there and after this it goes in to the pressure canner, to make sure everythig dies
Ha, ha I totally use my insta pot all the time. It works great. I just set the time on the sterilize settings to 1/2 hour. I do media, foil packs, you name it. I did start off with some blank PDA to test how well it worked.
it sounds advanced they've been cloning for years with physical mediums but I like the idea of the safety of the tissue materials instead of the possibility but if theres seeds your only nuts away from losing them but they should always have those stored in more than a few vaults.
I’m loving your channel, thanks. It would be good to hear what your lab’s “business model” is. How do you go about choosing plants to propagate and selling them commercially? I assume that this isn’t just a serious hobby!
PSA: if you do start an alcohol fire the most important thing to do is remain calm and do not grab or move the burning alcohol. Have something nearby to smother the fire.
I look through your videos and your content really inspires me and everything looks pretty straightforward, but some of my friends keep telling the way you introduce the whole process of TC is "just for fun" and they mainly point out that having a "DIY" laminar cabinet will drastically lower the overall results in TC. I as a person who's just interested in this sphere, will definitelly have extremely miserable results. Could you please tell if having a "professional" laminar cabinet/autoclave increase the chances? If so how much? Will it require the person to go deeper and deeper into this area to get some visible results? Cause it feels like without having more profound knowledge about this issue, it will waste time and money. I understand that you have really big experience of many years studying and researching and doing TC, but hand on heart will the person "who just started" have sooner results or not, especially having a DIY tools? Thanks)
Hi! I actually just started doing tissue culture in 2022. I have researched the process a lot, but I don't have any formal training or education. Anyone can do tissue culture. Having a laminar flow hood decreases the likelihood of contamination but I have seen many people successfully do tissue culture using a still air box instead of a flow hood.
I couldn't find in your videos what kind of paper you use to make the micropropagation in the sterile cabinet. do you use normal kitchen paper or another kind? do you sterilize the paper in the autoclave? how is it done? thank you very much for your videos and your work!
Enjoying the videos! Curious if you (or anyone else reading this), knows where to get the supplies related to plant cell tech, in Europe? As paying over 100 dollars for shipping is not ideal. Thanks!
I’m a home mycologist, started in a carpet room with no SAB or flow hood and yes, I’ve set gloves on fire while culturing more than once, I just don’t use gloves no more, and I don’t have carpet now 🎉
For what it's worth I use an instant pot for sterilization of tools, making my own agar dishes, bulk media sterilization, etc. You just have to to run it a bit longer so the material has time to fully sterilize. It has never been a problem. (Literally ever) (I do mycology and now do some tissue culture with all the same equip)
Can I use petri dishes for culture vs. the plastic containers you use I have boxes of glass petri dishes? I'm wondering if there is enough room in the dishes.
Can you do a video that is the whole process from start to finish (initiation to acclimating)? I know it would take months to be ready, but it would be interesting to see the progress with date stamps, etc.
Do you have any recommendations on a specific desk to buy? The only one I have right now is from Ikea and it has grooves on it which makes it impossible to sterilize.
I used to have a stainless steel table designed to be used in a kitchen. It ended up rusting, so I'm not sure if I would recommend it. Now I use a lab bench with a heat resistant phenolic resin top. They are super expensive (definitely do not buy one new!) but I see them come up for sale from schools / research institutions sometimes for cheap.
My next vlog will have some begonia updates! I follow the protocol from Plants in Test Tubes for begonias (clean leaf sections in 10% bleach for 10 minutes and use 0.5mg/L BAP + 0.1mg/L NAA for the PGR's)
What is the largest size glass container Theoretically a tissue culture plant can be kept in? Is there limitations to size? Do you think a one gallon jar would be considered unreasonable? I want the plant to be able to get 8"-10" tall, That way I don't have to mess with them as much...
You could try a container that large but I think it would be much easier for it to get contaminated. I haven't seen a plant grow that large in TC before but could be a fun experiment :)
Thanks for sharing- I have to ask you, what do you do if you don’t have a gas stove? Pressure cookers don’t work on electric and it’s all I have- so I guess I’m going to have to spend over $200? 😊 please advise(I got the book and the rest, just no was to sterilize anything
Between us, I have a cheap electric pressure cooker (no instapot) on the high setting it gets over 15 psi. A large conventional pressure cooker has the benefit of being large.
With an instapot u adjust the time based on the lower pressure. When i started in mycology i would adjust at 1:1.7 now im almost 6 years beyond all that and using the proper equipment. Lol
If I had the space for this, I would blow hundreds in a heartbeat and then spend it again on weird plants. If you take requests / suggestions, Begonia lyallii is a rarely available (but not super rare) terrarium begonia from Madagascar that I'd totally buy so I can stick in my pygmy chameleon tank.
Can plants be GRAFTED at the tissue culture level?? Like, can u have pink and white variegated monstera in the same dish? What about unrelated plant species?
What about plants that are trademarked? How can you find out which plants you are allowed to tc and sell? I found one data base but it was extremely hard to navigate.
Do you still use fluvial statum and perlite as your substrate? I acclimated some Pluto Philodendrons and creatingthe holes for for their rooted booties was impossible
3:21 English isn't my native language, and I didn't get what she said about 'landlord' or something like that. Probably, is just a joke (I noticed her humor sense hehe), but I'm curious about this. Can someone explain it to me?
when you think about it on a microscopic level, scissors eventually just arent good enough simply because the blade is too wide and will ALWAYS make a cut that is too wide. As your stems get thinner it will be harder for you to expose significant surface area, Scisssors are not a replacement of a scalpel. I can understand that taking an injury like you did can make it harder to "hop back on the horse" but I can assure you that there is no replacement to getting more accurate and more exposed cuttings as you get deeper into the hobby. It sounds like you are pretty dedicated. Imagine how much surface area you can expose from day 0 on a new cutting if you choose the best angle. Such things just arent even worth thinking about with scissors because the best you're going to get is a cut. Imagine doing tissue cultures with a steak knife. Thats the difference between using scissors and a scapel vs a steak knife and scissors lol.
I have never started an alcohol fire but I have blown up a bunsen burner twice. short version the teacher didn't believe me when I told him what happened.
Fun fact: all of this gear is also appropriate for mycologists
But with plant growth regulators.

So true. I randomly came here searching agar.
This is how I found this channel
Yep pressure cooker 4 life 😊
exactly i have everything in my mycology lab already hah
This is by far the best tissue culture channel I have found, I am inspired and captivated by your content. Thank you for these videos!
You can also get infrared heaters for dental wax tools for a fraction of the cost of the bacizapper or any other purpose built infrared sanitizer
Didactic, motivating, and entertaining as always. You have no idea how much I appreciate your content. Thank you once again, L!
I cannot get enough of your uploads. Thank you
Thank you!!

Hi dave, this is a safe place and you're among family. I know a dude named faint who caught a whole glove on fire lol.
I hope someday to meet someone who is as passionate as you are about plants and science. I have so much passion for this.
The information on your channel is extremely useful and I admire your intelligence. You're teaching me a ton and it's much appreciated!
Your editing is perfect!! Constant knowledge and keeps you interested.
oh man, cutting the saran wrap with pvc pipe cutters is genius. i started buying parafilm because i just got sick of trying to cut wrap rolls
Love your stuff and your humor, keep it coming!
You are flowing source of knowledge, THANK YOU!!

This is fantastic, will consult as i try and build out my own stuff, thanks for making detailed videos like this!
Thank you! You have the best plant collection to TC
Ha, ha I totally use my insta pot all the time. It works great. I just set the time on the sterilize settings to 1/2 hour. I do media, foil packs, you name it. I did start off with some blank PDA to test how well it worked.
Not only fun and great vibes but funny
it sounds advanced they've been cloning for years with physical mediums but I like the idea of the safety of the tissue materials instead of the possibility but if theres seeds your only nuts away from losing them but they should always have those stored in more than a few vaults.
Your videos are awesome. I'm setting up for narcissus tissue culture, and appreciate your channel.
PVC Pipe Cutter! thats fantastic!!
I’m loving your channel, thanks.

How do you go about choosing plants to propagate and selling them commercially?
I assume that this isn’t just a serious hobby!
My next video is an interview but the video after that (late Feb) is actually really similar to the one you just described!
yes, I have the same question.
This was very helpful. Thank you so much.
Perfect! ... Thank u for the hope
Great video. I'm making notes so I've watched it a few times. I wish you'd do a livestream. I've got questions I'd like to ask you.

@@plantsinjars thanks!
How long do the HEPA filters last on your cabinet?
Great intro to getting started, 👏👏👏
The new instant pots have canning and sterelize options that go up to 15 psi
Following this! Has anyone used this with autoclave indicator tape?

Hi you are amazing, please make video on Banana tissue culture and Mango. Need step wise tissue culture video for these plants
I saw some people say you can us an insta pot for took autoclaving
Hello! Did you try temporary immersion bioreactor? I am looking for the components to diy.
Love yu videos!! I hav purchased about 75% of the stuff to get started. I have soooo any questions for you! Thanks for all you do!
Could you use a UV sterilizer? One designed for phones and toothbrushes and stuff?
You killed me “@hello gamers!”😂. I feel exposed! 🤣

Keep your blue lab in water or storage solution at all times. Submerged just past probe
Can I use petri dishes for culture vs. the plastic containers you use I have boxes of glass petri dishes? I'm wondering if there is enough room in the dishes.
Can you recommend particular devices required to start Home Tissue Culturing Indoor & Outdoor Exotic plants.
Can you make a plant from a piece of grapes from the store? Or only from the leaves?
If you found a protocol for grapes, it would definitely specify! I only work with houseplants
Can you do a video that is the whole process from start to finish (initiation to acclimating)? I know it would take months to be ready, but it would be interesting to see the progress with date stamps, etc.
I have a similar vid coming out next week :)
What volume range should I be looking at to buy a pipette?
How you auto clave those plastic container?
Do you have any recommendations on a specific desk to buy? The only one I have right now is from Ikea and it has grooves on it which makes it impossible to sterilize.

i'm mycology enthusiast. we use all the same materials. you could come to the Dark Side. 😎
Really glad you said you are a menace to probes lol
Also guilty, so made me feel better
Love the sense of mild shame/self recrimination you have over owning not two, but four pressure cookers 🤣
pressure cooked paper towels?
I just started attempting tissue culture with Begonias. Would love an update on how you do yours.

Really funny. Wasn't expecting that lol
What can be used to lift your racks in the PC so high?
I use balled up aluminum foil to lift up the bottom rack
Hi I feel lucky to find your channel I have some questions
I also had the problem of watching Naruto and not being able to finish. Where did you place the vpn to be able to watch it?
What is the largest size glass container Theoretically a tissue culture plant can be kept in? Is there limitations to size? Do you think a one gallon jar would be considered unreasonable? I want the plant to be able to get 8"-10" tall, That way I don't have to mess with them as much...

It's easier if the media is wet. But yes that is what I still use!
How would I find what PGR and Media I would need to do Euphorbia species?
I love your begonia shirt ❤ Where did you get it?
From a brand called Tropical narcotics in the UK :) I found them because they designed Good Growing's merch
2:57 hate it when that happens ;)
Ty for all the great tips. $50 000 sounds like a very expensive scalpel.
Where'd you get that shirt? I want it!
Hello we want tissue culture dates plant please help me
Is it possible to do tissue culture to any plant? Or only specific plants are fit for tissue culture.
Plz explain me.thanks
Any plant
Great information, thank you.
you are so amazinggg :)
Gldn content, cheers ❤
My both hands were blown inside the inoculation box. I was hospitalized and got 2nd and 3rd degree burns. Be careful in choosing the type of alcohol.
Do you store your MS and PGRs in a fridge or freezer?
Fridge
I thought pressure cooking psi determines where you live? You state 15 psi but what state do you live in?
I was under the impression that sterilizing at 10-12psi just required longer times?
I’m start to buy step by step.
Missing : laminar flux and a cold chamber at 25 *c celcius.
How much do you actually make on average per month?
how much do YOU make hmmmmm
In the cultivation of mushrooms, the community successfully sterilizes @ 12psi for 45 minutes for agar. I'm guessing it would also work here.
I too have started an alcohol fire, but from dabbing
3:21 English isn't my native language, and I didn't get what she said about 'landlord' or something like that. Probably, is just a joke (I noticed her humor sense hehe), but I'm curious about this. Can someone explain it to me?
Venusflytrap please. Tenks
You're not alone, my old girlfriend forgot she was holding a scalpel in biology class and jabbed it in her leg! Live and learn.
I think, i love that girl
Any idea why your videos look a little out of focus/blurry even on 1080p......? Great content never the less...
Tissue culture is easy but technology is hard :(
