This is a fantastic discussion! Two of my favorite UA-cam commentators in the field of longevity having an in depth discussion on a relevant topic! Wonderful! Very interesting information here with a lot to unpack. I have a couple comments. First, I like the focus on looking at male vs female effects. I'm a male, 49, and I started taking NMN (and later NR, NAD, and Trigonelline) several years ago. Within two weeks I noticed a number of changes, noticeably that I had much less fatigue and drowsiness, particularly during late night drives. It also changed my sleep, such that I could sleep less and feel more rested. So I surmise that NMN and the other boosters are doing something significant to improve my well being, even if ultimately they may not by themselves extend my lifespan - although I do a couple 24 hour fasts per week and also engage in regular moderate exercise which it seems from this video activates the salvage cycle. Second, I am curious about how liposomal delivery might change matters. If I understand correctly, the conversion of NMN to NAM occurs in the gut due to bacteria, and then this NAM suppresses the sirtuin activity. But liposomal encapsulation protects the NMN from digestion and allows it to go directly into the bloodstream and into cells. Perhaps this can help keep NMN supplementation from suppressing sirtuin activity? And combined with molecules like resveratrol we could get the double whammy of increased sirtuin activation along with increased cellular energy to facilitate the youthful actions of cells. Just a thought but I very much want to see research involving liposomes. For disclosure, I personally take the Reneu NAD+ Complete booster which has NMN, NR, NAD, and Trigonelline, all within liposomal capsules, so I'm fully in agreement with the idea of combining pathways, not just with NAD boosters but for all longevity supplements. Anyway, keep up the great work, Rimon and Nick! You're two shining lights of useful information in the longevity space!
Thanks for sharing Jason. The study suggests that a lot of what you see has to do with anti-aging effect on genes in your muscles. I haven't seen much data on liposomal to give you a response
I don't know if my input would be helpful or not, but I am a 61-year-old woman, and I have been taking NR or NMN regularly for about twelve years. It is not the only supplement I take. I feel like it helps my immune system -- after listening to this, maybe it's because it's helping my gut? Other than a couple of days of Covid years ago, which I brushed off quickly, I don't even get colds. Compared to other women my age, I do feel that I'm much healthier and look and feel younger. I don't have wrinkles or gray hair, no arthritis or aches and pains. I don't take any medications. Of course, all this is anecdotal, but I hope the information is at least a little helpful. Oh, I also give it to my 16-year-old dog, and he's doing fantastic.

I am female and 53 years old and I have post concussion syndrome. I have problems like brain fatigue and more. I have tried lots of supplements by now to increase energy and the one thing that has worked miracles is NMNH. After 3 days of NMNH I could feel the right side of my body again. Just an hour or so after taking my first NMNH I felt that something started to happen in the right side of my body and my brain. I felt pain in the right side of my body, which had been numb until then. Then the pain went away after a week or so and now I still have the sense of the right side of my body. NMNH has been amazing for me. I have tried NMN but not felt any different.
@@LesGaminuscules I don't know but NMNH is a more stable kind of NMN. I can really recommend NMNH. It's about 10 times more potent than NMN so you only need a very small dosage also. NMNH (Dihydronicotinamide mononucleotide), also known as reduced nicotinamide mononucleotid.

I just posted about this. I thought sirtuins were the ones for longevity. NMN is just the fuel for sirtuins to work. But this study was mainly for longevity for NMN specifically?
Just achieving a better health span makes it worth it. I and many experienced a revolution in brain function, oxygen uptake and resilience. It really has been a game changer for me personally.
@ this is the case where humans lead science because there’s a lack of clinical testing methodologies to substantiate or this is one of the biggest pyschops in biohacking domain. Another good example is my father in law had dementia for 7 yrs. 12 months ago I gave him NMN and now he shows no signs of dementia. I know NMN doesn’t solve these issues if given to other people but it worked for him. I think Sinclair has this problem. He sees the results but can’t prove it clinically. I know my life without NMN will be less so I continue to believe.
@@nutech1810 Niacin is even more powerful than NMN. Dr Michael LustgartenPhD in his channel "Conquer Aging Or Die Trying!" shows in his blood tests how smaller doses of Niacin raises your NAD more than NMN. Also, some of the NMN you take is transfored into Niacin by your gut (Dr. Matt Kaeberlein). Of course, the sheep follow the hype and buy expensive NMN or NR.
@@nutech1810 You are right. they are different. According to some results which I linked on a previous comment (but UA-cam deleted), Niacin is more efficient in raising NAD levels. Lower doses will raise your NAD more than NMN. The side effect is flushing though. Of course there's a conflict of interests, there's a lot of money involved in this business.
I know people who run one hour every second day in their 50s who started taking NMN. After two weeks they said that they do not get exhausted like before. Brainfog, tiredness seem to go away for many. But some people, seemingly adhd, fast brainers can get jitters from NMN so it seems like everyone just have to test. After 2-4 weeks the benefits seem to occur and if not, it is the wrong supplement.
I have ADHD. It does not give me jitters. Sounds like you bought a bad brand. I only take 500 mg a day, and other longevity supplements with it in a liposomal capsule, which works better than powder.
I have used the same brand even the same batch, and it does not seem to be the brand nore the batch. Some just say they get jitters or feel uncomfortable. Maybe 1/10 from my experience. Maybe they are too young, I cannot really tell what the common faactor could be. However, the older people are, the higher and faster the effect and older people often become faster in their thinking, get more resilience both physiologically and physically. For me it really was a positive lifechanger. I am almost 50.
I’m 81, been taking NR as Resveracel since 2018. I’m active, nothing hurts, feel good. No way to know if the NR is resonsible. Also it’s still early in the day.
Some anecdotes: I have been able to mitigate a beginning flu, by taking NMN multiple times a day. And I have been found it to be beneficial to insuffilaye it. It seems to have local effect on infection. Oral intake is much less effective. Now it is soon 2025 and I have not gotten even covid nor any other flu since I started NMN. Now I insuffilate immediately if feeling even slight onset if flu or such. My 90 years old mom has gotten significantly energy from oral NMN use. I have not experienced significant energy boosting effect from oral intake. But I get some acute effect from insuffilating it. It even boosts libido a little bit. It wears out rather quickly and definitely does not last a day. I have gotten rid of the flus by taking it about 5 times a day. And when insuffilating, only a little seems to be sufficient, as it is spread through the day. Then there are some combination effects, Qualia Senolytic plus NMN together, brings the acute effect of MMN even more concrete. But likewise the effect is short. I got energy boost to run a short run but after the run was subjectively back to baseline. But more NMN again re-energised it. We found that liposomal NR does have similar effect, but it is longer than NMN. Including the libido boost and energy boost. But that is with high dose of liposomal NR. It is not a dopaminergic boost, it is something else, feels different. My anecdotes would indicate IMO, that NMN (and also NR) needs to be spread through the day, and NMN orally is significantly less effective than other delivery methods such ss insuffilating it or sublingual intake. And my anecdote would indicate it has some local effect on infection, and combatting flu seems to be possible. To clarify: my baseline libido is very low, so I can easily observe this effect on NAD-boosters, because anything more than zero is more than zero. Not incredibly strong effect but can say it has an effect and it is a good/wanted effect when having low libido. Meaning that it could be unnoticeable to someone with high baseline libido to begin with. But for someone who struggless with low libido, NAD-boosting, on demand, seems to me one working way to combat the problem. But a NMN taken in the morning has no effect in evening. It needs to be taken with a lot shorter succession.
this is explained by the study - the effect on the microbiome which directly controls the immune system, inflammation.This is why I like this Sinclair study. - explains a lot of what people see and we didn't know why. and possible lifepsna extension benefits to you (female)
Moral of the story: until high quality human trials are conducted, NMN benefits are just conjecture. As this video points out, there too many confounding factors to draw any real conclusions
No.... people forget how long NMN has been studied..all forms of niacin. It has studies that predate Sinclair or any of the longevity movement. If you dig through research going back decades it supports longevity and definitely healthspan..it's not debatable.
@@paulharrisonadventuregearm5457It's only been available as a supplement since 2015, although it was discovered much earlier. As far as its longevity benefits are concerned, those are certainly debatable, and not at all settled science.
I take liposomal NMN 500mg, liposomal NR 250mg and liposomal NAD 250mg (huge improvement) when I take regular NMN it is noticeably not as effective. I also take NAC 600mg, milk thistle 1g, Glycine 1g, taurine 2 g, and liposomal astaxanthin, liposomal sulfurafane . I have been full carnivore for 14 months, lost a permanent 40lbs and am in the best shape of my life, even some of the gray in my beard has changed back to normal colors, my eyesight has improved, my sleep is improved also. Same here. I take NMN in a longevity supplement. I also did an N1 experiment and stopped taking it for 90 days... I could feel a significant difference. It also helps balance female hormones 40+. Skin, hair, hormones are great. I only cycle off it on Sundays to give my body a little break.

I'm switching to NMNH as apparently it remains for a longer period of time in the blood and in organs like heart, liver, muscles etc....I don't know if it's true and if it's not just an issue with NMN absorption ? When I take NMN or NR I truly feel a surge of energy and when stop it I feel a difference and i have been taking either NR or NMN (1g or more) for at least 7 years, probably more as I started this journey with NR from Niagen. I was taking more than 100 different supplements per day, now I have reduced a little the number of them , but the number is still very large. I try as much as possible to take as many supplements in the liposomal for or as cyclosomes or phytosomes to improve their absorption. My blood tests are excellent including my liver enzymes ; my kidney's filtration result is excellent and is usually find in people 20 years younger than me. I also look at least 20 younger than my age and the only thing I don't seem to be able to address is the greying of my hair ... no supplements i tried work for me .....
Here in Australia with the NSW cancer council they conducted a skin cancer experiment with NAM, it was found that NAM reduced the spread and inception of skin cell carcinoma. To quote "In this phase 3, double-blind, randomised controlled study, 386 patients at high risk of skin cancer took either nicotinamide 500mg twice daily or a placebo for 12 months. The results were striking: the rate of new non-melanoma skin cancer was 23% lower in the nicotinamide group than in the placebo group" . This of course means that supplementing B3 compounds such as NMN,NR and NAM are having an effect, I would speculate that B3 compounds have an anti-aging effect but as stated in your video the dose should be spread out over a 24 hour period, in the study aforementioned the dosage was given as 2 doses 12 hours apart over a 24 hour day.
I forgot to mention that since doing the NAM protocol myself (for many years) as I go to the beach regularly and get a lot of sun. I seem to have boundless energy for someone my age 60+ compared to middle aged adults and their children so it may be also increasing my NAD levels.

Prove to me in the context of the study I mention that it is not proof of anti aging in the context of results from none bolo dosing, it explains exactly the effect of dosage separation over time, sure they are just looking at the skin but it's happening in all cells in the body because it's oral dosing. I think your missing the forest for the trees, it's obviously stabilising the DNA structure in the skin so it's no reach to say it could be happening in all cells of the body. As "Sinclair" would say it's stopping the "scratching of the CD". Stabilisation is the first step, reversal is the next.
There are entire UA-cam channels based on anecdotal experience with NMN, so until human studies are more plentiful and conclusive, I'll stick with niacin as well.
Hi Rimon, I hope you see this - there's something very important to take into consideration when reviewing mice studies: laboratory mice are bread in an environment that encourages fast tissue repair over lifespan. Due to their fast commercial unnatural breeding, lab mice have extra elongated telomeres (the protein chain at the end of the chromosomes in the cells) which allow quick tissue repair of cells but causes tumor growth when they get old. Dr Bret Weinstein had made a fascinating work on the subject. The point is - any product that is aimed to show tissue repair in mice, is already has the cards stacked against it, since it will meet mice that are "turbo charged" for fast cell repair. As far as I know, there aren't any labs that suggest naturally bred mice that will not have this pre disposition.
@22:30 Absolutely, they needed to always have had two different cohorts, male and female - what a late awakening, by scientists, of all people! I too notIced that NMN improved my fur color 🙂 A small, but noticable, improvement in hair color. Still gray, but less so. I got this also from NR, not only NMN. That's from 1 g a day or a little less of NMN.
Yes exactly. this fur improvement correlates with what we've seen with people reversing white/gray hair! So NMN looks like "frailty" play, not longevity play!

So thing about nmn is I noticed improvements in my vision, all the hair on my body got thicker and longer, my eye lashes have gotten so long and thick that people have asked me if they were fake. I’ve noticed remembering dreams more but not much else. I’m a male btw.

I think we cannot conclude things and say NMN has less benefits based on small population clinical research. I know that NMN is good because myself I have seen many benefits on my overall health as I take it for maintenance and I am 51 years old. I also sell NMN and I have testimonies of my customers saying they are no longer taking their blood pressure medication. some tell me their vision improved. Some noticed that they no longer have prostate issues . Some mentioned their level of energy increased. So told me they have better skin and many people tell them they look younger now. Some reported they no longer have constipation issue. Another customer told me she no longer have pain.
Interesting! NMN fixed my friend’s constipation too. Others in my life have reported more energy and less pain. It helped with my perimenopause and gave me incredible energy and endurance. Lost about 15pounds and my friends have said I look younger too.
Yeah, I noticed that years ago, that women and men responded different to different stimuli. But ultimately, we're ALL different. NMN may help some individuals but not other individuals. Obviously age is a factor, as Rimon mentions later. What I would like to see more of is the analysis of the range of results of particular studies, rather than a focus on the mean (or median) results.
I’m in early 60s male, took NMN by Renue for about a year, felt nothing and quit taking it. Honestly tho I have taken supplements for 45 years and have never felt anything from any of them. Only two things I have ever felt a positive change from and that’s testosterone cream and exercise. I would try Niacin but hate the flush.

Yes the comments are are correct...of course, you can claim better movement and more fat burning are also "fitness" although they didn't view that this way in this study.
Great discussion, but looking into taking nicotinic acid rather than niacinamide: this 2017 study suggests niacinamide may *stimulate* sirtuin activity in cells (Nicotinamide is an inhibitor of SIRT1 in vitro, but can be a stimulator in cells), and other studies suggest nicotinic acid may reduce insulin sensitivity, raise blood glucose and increase risk of diabetes, whereas precursors generally have the opposite effect (glycation is probably one of the major factors in cellular ageing). So I'm not sold on taking regular niacin as an alternative yet.
This is only relevant to high dose niacin whilch works completely different. As you suggested, choronically hurting insulin sen. and blood sugar, and must be cycled - devil is in the details, as usual with every supplement I've researched. it's different supplement high doses vs low doses.
@@WellnessMessiah That seems to be the case for nicotinamide suppressing sirtuin activity as well (mentioned towards the end) - that those doses are in the range of grams (and may still have a net stimulatory effect). It seems the best way to counteract NAD decline is physical activity - which I understand completely ameliorates any age-related effect. And I'd wonder if the next best isn't CD38 inhibition with polyphenols?
I've been striving for better fur colour maintenance for years. Not sure less tail stiffening is an advantage, but then again that was females so who knows! Haven't tried burrowing yet - maybe 2025 could be the year 😂
Were any of the p-values adjusted for multiple hypothesis testing? Of course, if you look for many possible benefits (lifespan, frailty index, fitness, etc.) and do not adjust your p-values, you will end up with some significant (nominal) p-values by chance...
I think it may be because it does not replace exercise but only boosts it in terms of muscle building. People that do not exercise will not see much effect. Unless may be for females, due to additional microbiome effect… Does slight muscle mass improvement in itself improves longevity? Definitely healthspan, but lifespan I am not sure about. If you are not atrophied, slight variations won’t matter much, probably.
@@Nando_lifts2021Yes, most likely. But see, both groups exercised the same, as I understood. And in that case NMN only can do so much. Improve health, fur, muscle but not enough to increase longevity. Also, mice die mostly from cancer…so who knows how it would perform in humans long term. But spike in niacinamide by-product concerns me a bit - suppression of feedback recycling loop and inhibition of surtuins is not what we want at all…
If a company (or a person) doing the study has a financial interest in the results, well, it's not trustworthy. It is essential that independents do the research....Interesting video, I will try taking the niacin. I did take NMN for a while, I didn't notice any changes whatsoever in my body or brain...
My friend took Nichido time for 3 months and her liver enzymes were way high. Her doctor immediately told her to stop Nichido time as she was showing signs of liver damage. Pls be very very careful and don’t take Niacin without checking your liver enzymes. It’s so dangerous!
It should be seen from a different prospective, maybe. Not from the Longevity but from the state of health/quality. That is what Sinclair stated more than once, have a longer healthy old age life and a sudden natural death should be the goal. But here comes the one million dollar question: What has every slimming diet in common? Exercise! Now, we know today, exercise has a profound impact on your physical health, is exercise at first the key to everything, probably? Second, healthy food. That's it.
As a 60 year old otherwise healthy male, with T2D, I've been taking NMN for about three years now. With a consistent training routine over this time, I have observed significant performance increases in both recreational running and cycling races. This improvement was definitely not expected, especially owing to my T2D....Thanks for the thorough analysis....
My initial thought was the opposite in that I believe the study was designed to fail. Sinclair has been touting MIB-626 which is supposedly far more potent than NMN. And since the MIB company has tried to get NMN banned, why would they design a trial where NMN succeeded? My conclusion is that the trial is worthless.
@@lopezb Sinclair still takes 1g NMN in the evening. If NMN has no effect, why does Sinclair still take it? He specifically stated taking NMN not MIB. That fact and the fact the company has tried to get NMN banned indicates to me that they cannot be trusted to perform a study on NMN.
All the "bad stuff" were quite hidden in small lines or in the supplemental data. The publish the study highlighted the benefits...hidding the downsides. The study headline shold have been "NMN doesn't increase lifespan in male mice, not even with 3.5 grams".
I'm interested in MNM, NAD safety - and supplement quality. *I've never taken a supplement that has had such an incredibly obvious effect: (56 yr old male)* 300mg (Hpn brand) in the morning, I distinctly don't feel tired at the end of the day. As in, not "maybe" - but *definitely* felt better at the end of the day relative to a lonnnng time previously. Same observation the following days, accepted it as a new norm. Big life improvement, actually. I quit taking NAD because I saw a study that implied the anti cell death aspect could increase cancer risk? I again, distinctly miss the effect of it. I do not get this impression from any B-type vitamin supplement. So I do have a bias towards wanting to be convinced it is safe, because from my vantage point it definitely works.... I would also like to recommend it for my elderly father, but fear for a latent cancer possibility being exacerbated? My wife - but her mother had glioblastoma; risk?
I took NAD poster and the year before showed no tumour but a year after it showed a benign kidney tumour..I loved the effect of NMN but I stopped for a year now out of fear . NR gave not much energy compared to NR
Was NMN designed for longevity on its own? I thought it was just fuel for sirtuins which can from other molecules. We shouldn’t expect longevity from NMN but healthspan?
The body makes it naturally when you're under biological stress. I guess there was a desire to mimick this effect and see if you get the benefits (not quite, but some)
@ my question is more that longevity was based on information theory and that sirtuins are at the core of longevity not NMN. I understand they are studying it on its own but the root of Sinclair’s claims have always been about sirtuins. He used to think sirt1 was key. Now he has shifted to sirt6. Never have I seen him claimed NAD was the key to longevity but the gas for sirtuins to excel
The impact on the gut microbiome both in female and male suggest you actually boost their nad+ and allow them to stay alive as you grow older! They turn into niacinamide (not great)
Rimon said he takes Nicotinic Acid (Niacin) 50mg twice a day, thats the flushing kind. I haven't watched the entire video but I don't know if there is anything said against the other forms of B3.
You mention thatbif you are a man over 50, you should feel it or it doesn't work for you. And that you should test it? How much and how long before feeling it? What kind if test? I get so confused by these videos. What should we take specifically? What form? What dosage? How frequently? With without food? Sorry for so many questions. What about for women? Is there a cheap way to take it? I emailed you about Mexidol helping my brain after long, severe anemia. How is your wife? We are 57 Ah i see at the end the type. Still confused though. No they didn't take it with fat. They had quite low fat diet, so even if they drank water with meal (or went to bar...not really) it was without fat. Most of the NMN on empty stomach in this study.
@WellnessMessiah Thank you! But it would be better, to give the mices a food, which is mostly similar to the food for humans, otherwise they can't see the correct results. If humans would eat only water and NMN, they will not life longer too. :-)
Why didnt they design the study with different cohorts, each having different NMN protocols? Eg different doses of bolus (especially lower doses) , bolus at different times of the day, different distribution over the day etc etc Would Sinclair have a motive to make normal commodity NMN look bad so he could sell his super duper ""enhanced" NMN product? I wonder....
If we are getting only one cycle of NAD (and then niacinamide) wont it make sense to either have multiple small dosages (say rather than having 300mg at once have 100mg at interval of 4-5 hrs). Also wont delayed release capsules make more sense.
is funny that they run a study and do not understand the metabolism of NAD. Providing the equivalent of 3.5g per day heavily burdens the methylaion capaciy of the organism, gobbling up all available methyl groups. Such a high dose must be paired with both TMP/methylfolate and glycin, the latter to buffer a potentially ovvershooting metylaion capacity. Not dong that means that any positive effect of NMN would be reversed. Second, the pape does not explore the correct context. Boosting NAD is not the same as boosting the salvage pathway. Else, the TCA and the ETC must work as well too. Introducing too much NMN however corrupts at least the ETC. Sinclair takes Q10 and TMG, btw as well, as he probably understands the energy metabolism. But the researchers of that company: stupid.
It would be nice if those who talk about supplements (and studies etc ) would include "ALL" relevant information. Most will discuss dosage but, what about "very" important information such as take on empty stomach or with food, what time of day, "What form is best". I like many folks have a hard time swallowing pills and prefer to take supplements in powder form. However, I also realize that some forms are better absorbed than others and some aren't absorbed at all and are distroyed by stomach acid ( pills, shots, sublingual, liposomal etc.). Maybe you'd like to be the "FIRST" to give us "ALL" the information we need to understand a particular supplement and how they were used in a study (if known). Thanks Rick B.

The price of highly pure bulk NMN has dropped significantly-from $150 to just $33 per 100g. That means it now costs only $1 per day for a 3g dose. Capsules are still overpriced, but BULKsupplements offers an affordable alternative. Personally, I'm sticking to 2g of NMN daily, paired with 1g of TMG (trimethylglycine) to support methylation, which helps replenish methyl groups that NMN consumption can deplete. I also add apigenin and quercetin on a 3-weeks-on, 1-week-off cycle as inhibitors of the NAD+ase enzyme CD38.
The best approach is combining animal research + ancedotal evidence in humans + taking and tracking our own body. In 5 years we would have more data. Happy new year
It would be nice if they said what NMN was. I've taken a lot of vitamins over the years, but I'm cutting back because I've learned most are synthetic and that doesn't seem good. Reseveratrol helps . Total myth it doesn't do great things. It's supported in the literature going back seven decades. People like Dr Bradfield.. surface perused reseveratrol and found one study that supposedly undid any benefits. You sir didn't waste your money on reseveratrol..not at all. It's a very very powerful supplement for a whole host of longevity and healthspan issues. I suggest you look for yourself. It's a PDE4 inhibitor, it's a cox11 inhibitor, it reduces neuroinflammation gut/ intestinal inflammation, it specifically increases NAD+ in heart tissue, it reduces bad cholesterol, it reduces artery stiffness, mends the blood brain barrier..this has been found time and again in peer reviewed studies. It likely does switch surtuins on in an indirect manner
Sinclair has been using botox. Just look at his forehead. I saw him up close at an airport with my niece who is a dermatologist. She speculated that it was botox. She injects it to so many of her patients. She can spot a person with box injections a mile away. I would not be surprised if he colors his hair.LOL

But, if I am a 60 year old man, already doing some HIIT and IF and I can shift my mitochondria to more fat metabolism, even 3-5% more or whatever it was...I can either reduce calories, reduce carbohydrates, compensate for a slight "overconsumption" of nuts and coconut oil (Perplexity agrees, helps mobilize phytochemicals from spices)...I can get rid of garbage starch calories....I know when I did long distance cycling on The Zone...two consecutive 200 mile days consuming very little food...it could be that the paleo/keto guys aren't completely insane... I am thinking to dump white rice in favor of vegetables, almonds, walnuts, sunflower seeds, avocados in season, etc., because those are certainly better quality calories....does that make sense? I know you guys can do a better job on this line of thought. Also, if I think my bio age is more like that of a 50 year old :) maybe niacin is a good first cut?

This is a fantastic discussion! Two of my favorite UA-cam commentators in the field of longevity having an in depth discussion on a relevant topic! Wonderful!
Very interesting information here with a lot to unpack. I have a couple comments.
First, I like the focus on looking at male vs female effects. I'm a male, 49, and I started taking NMN (and later NR, NAD, and Trigonelline) several years ago. Within two weeks I noticed a number of changes, noticeably that I had much less fatigue and drowsiness, particularly during late night drives. It also changed my sleep, such that I could sleep less and feel more rested. So I surmise that NMN and the other boosters are doing something significant to improve my well being, even if ultimately they may not by themselves extend my lifespan - although I do a couple 24 hour fasts per week and also engage in regular moderate exercise which it seems from this video activates the salvage cycle.
Second, I am curious about how liposomal delivery might change matters. If I understand correctly, the conversion of NMN to NAM occurs in the gut due to bacteria, and then this NAM suppresses the sirtuin activity. But liposomal encapsulation protects the NMN from digestion and allows it to go directly into the bloodstream and into cells. Perhaps this can help keep NMN supplementation from suppressing sirtuin activity? And combined with molecules like resveratrol we could get the double whammy of increased sirtuin activation along with increased cellular energy to facilitate the youthful actions of cells. Just a thought but I very much want to see research involving liposomes.
For disclosure, I personally take the Reneu NAD+ Complete booster which has NMN, NR, NAD, and Trigonelline, all within liposomal capsules, so I'm fully in agreement with the idea of combining pathways, not just with NAD boosters but for all longevity supplements.

I don't know if my input would be helpful or not, but I am a 61-year-old woman, and I have been taking NR or NMN regularly for about twelve years. It is not the only supplement I take. I feel like it helps my immune system -- after listening to this, maybe it's because it's helping my gut? Other than a couple of days of Covid years ago, which I brushed off quickly, I don't even get colds. Compared to other women my age, I do feel that I'm much healthier and look and feel younger. I don't have wrinkles or gray hair, no arthritis or aches and pains. I don't take any medications. Of course, all this is anecdotal, but I hope the information is at least a little helpful. Oh, I also give it to my 16-year-old dog, and he's doing fantastic.

I am female and 53 years old and I have post concussion syndrome. I have problems like brain fatigue and more. I have tried lots of supplements by now to increase energy and the one thing that has worked miracles is NMNH. After 3 days of NMNH I could feel the right side of my body again. Just an hour or so after taking my first NMNH I felt that something started to happen in the right side of my body and my brain. I felt pain in the right side of my body, which had been numb until then. Then the pain went away after a week or so and now I still have the sense of the right side of my body. NMNH has been amazing for me. I have tried NMN but not felt any different.

@@LesGaminuscules I don't know but NMNH is a more stable kind of NMN. I can really recommend NMNH. It's about 10 times more potent than NMN so you only need a very small dosage also. NMNH (Dihydronicotinamide mononucleotide), also known as reduced nicotinamide mononucleotid.

When I started taking NMN 6.5yr ago, it was for "healthspan" and quality of life improvements. It has definitely delivered in that area for me.
I just posted about this. I thought sirtuins were the ones for longevity. NMN is just the fuel for sirtuins to work.

Just achieving a better health span makes it worth it. I and many experienced a revolution in brain function, oxygen uptake and resilience. It really has been a game changer for me personally.

Another good example is my father in law had dementia for 7 yrs. 12 months ago I gave him NMN and now he shows no signs of dementia. I know NMN doesn’t solve these issues if given to other people but it worked for him.
I think Sinclair has this problem. He sees the results but can’t prove it clinically. I know my life without NMN will be less so I continue to believe.

@@nutech1810 Niacin is even more powerful than NMN. Dr Michael LustgartenPhD in his channel "Conquer Aging Or Die Trying!" shows in his blood tests how smaller doses of Niacin raises your NAD more than NMN. Also, some of the NMN you take is transfored into Niacin by your gut (Dr. Matt Kaeberlein).
Of course, the sheep follow the hype and buy expensive NMN or NR.
@@nutech1810 You are right. they are different. According to some results which I linked on a previous comment (but UA-cam deleted), Niacin is more efficient in raising NAD levels. Lower doses will raise your NAD more than NMN. The side effect is flushing though.
Of course there's a conflict of interests, there's a lot of money involved in this business.
I know people who run one hour every second day in their 50s who started taking NMN. After two weeks they said that they do not get exhausted like before. Brainfog, tiredness seem to go away for many. But some people, seemingly adhd, fast brainers can get jitters from NMN so it seems like everyone just have to test. After 2-4 weeks the benefits seem to occur and if not, it is the wrong supplement.
Now we know why... it opposed age-related gene expression in the muscles!

I have ADHD. It does not give me jitters. Sounds like you bought a bad brand. I only take 500 mg a day, and other longevity supplements with it in a liposomal capsule, which works better than powder.
I have used the same brand even the same batch, and it does not seem to be the brand nore the batch. Some just say they get jitters or feel uncomfortable. Maybe 1/10 from my experience. Maybe they are too young, I cannot really tell what the common faactor could be. However, the older people are, the higher and faster the effect and older people often become faster in their thinking, get more resilience both physiologically and physically. For me it really was a positive lifechanger. I am almost 50.
Probably they feel better because they expect to feel better.
You can tell Rimon knows so much more on NAD. He understands the nuances and also experiments on himself.

Some anecdotes: I have been able to mitigate a beginning flu, by taking NMN multiple times a day. And I have been found it to be beneficial to insuffilaye it. It seems to have local effect on infection. Oral intake is much less effective. Now it is soon 2025 and I have not gotten even covid nor any other flu since I started NMN. Now I insuffilate immediately if feeling even slight onset if flu or such.
My 90 years old mom has gotten significantly energy from oral NMN use. I have not experienced significant energy boosting effect from oral intake. But I get some acute effect from insuffilating it. It even boosts libido a little bit. It wears out rather quickly and definitely does not last a day. I have gotten rid of the flus by taking it about 5 times a day. And when insuffilating, only a little seems to be sufficient, as it is spread through the day. Then there are some combination effects, Qualia Senolytic plus NMN together, brings the acute effect of MMN even more concrete. But likewise the effect is short. I got energy boost to run a short run but after the run was subjectively back to baseline. But more NMN again re-energised it.
We found that liposomal NR does have similar effect, but it is longer than NMN. Including the libido boost and energy boost. But that is with high dose of liposomal NR. It is not a dopaminergic boost, it is something else, feels different.
My anecdotes would indicate IMO, that NMN (and also NR) needs to be spread through the day, and NMN orally is significantly less effective than other delivery methods such ss insuffilating it or sublingual intake. And my anecdote would indicate it has some local effect on infection, and combatting flu seems to be possible.
To clarify: my baseline libido is very low, so I can easily observe this effect on NAD-boosters, because anything more than zero is more than zero. Not incredibly strong effect but can say it has an effect and it is a good/wanted effect when having low libido. Meaning that it could be unnoticeable to someone with high baseline libido to begin with. But for someone who struggless with low libido, NAD-boosting, on demand, seems to me one working way to combat the problem. But a NMN taken in the morning has no effect in evening. It needs to be taken with a lot shorter succession.
this is explained by the study - the effect on the microbiome which directly controls the immune system, inflammation.This is why I like this Sinclair study. - explains a lot of what people see and we didn't know why. and possible lifepsna extension benefits to you (female)
Moral of the story: until high quality human trials are conducted, NMN benefits are just conjecture. As this video points out, there too many confounding factors to draw any real conclusions
No.... people forget how long NMN has been studied..all forms of niacin. It has studies that predate Sinclair or any of the longevity movement. If you dig through research going back decades it supports longevity and definitely healthspan..it's not debatable.

I take liposomal NMN 500mg, liposomal NR 250mg and liposomal NAD 250mg (huge improvement) when I take regular NMN it is noticeably not as effective. I also take NAC 600mg, milk thistle 1g, Glycine 1g, taurine 2 g, and liposomal astaxanthin, liposomal sulfurafane . I have been full carnivore for 14 months, lost a permanent 40lbs and am in the best shape of my life, even some of the gray in my beard has changed back to normal colors, my eyesight has improved, my sleep is improved also. Same here. I take NMN in a longevity supplement. I also did an N1 experiment and stopped taking it for 90 days... I could feel a significant difference. It also helps balance female hormones 40+. Skin, hair, hormones are great. I only cycle off it on Sundays to give my body a little break.

I'm switching to NMNH as apparently it remains for a longer period of time in the blood and in organs like heart, liver, muscles etc....I don't know if it's true and if it's not just an issue with NMN absorption ? When I take NMN or NR I truly feel a surge of energy and when stop it I feel a difference and i have been taking either NR or NMN (1g or more) for at least 7 years, probably more as I started this journey with NR from Niagen. I was taking more than 100 different supplements per day, now I have reduced a little the number of them , but the number is still very large. I try as much as possible to take as many supplements in the liposomal for or as cyclosomes or phytosomes to improve their absorption. My blood tests are excellent including my liver enzymes ; my kidney's filtration result is excellent and is usually find in people 20 years younger than me. I also look at least 20 younger than my age and the only thing I don't seem to be able to address is the greying of my hair ... no supplements i tried work for me .....

Here in Australia with the NSW cancer council they conducted a skin cancer experiment with NAM, it was found that NAM reduced the spread and inception of skin cell carcinoma. To quote "In this phase 3, double-blind, randomised controlled study, 386 patients at high risk of skin cancer took either nicotinamide 500mg twice daily or a placebo for 12 months. The results were striking: the rate of new non-melanoma skin cancer was 23% lower in the nicotinamide group than in the placebo group" . This of course means that supplementing B3 compounds such as NMN,NR and NAM are having an effect, I would speculate that B3 compounds have an anti-aging effect but as stated in your video the dose should be spread out over a 24 hour period, in the study aforementioned the dosage was given as 2 doses 12 hours apart over a 24 hour day.
I forgot to mention that since doing the NAM protocol myself (for many years) as I go to the beach regularly and get a lot of sun. I seem to have boundless energy for someone my age 60+ compared to middle aged adults and their children so it may be also increasing my NAD levels.
Yes niacinamide is good for this specific purpose. Aging is a different thing.

Prove to me in the context of the study I mention that it is not proof of anti aging in the context of results from none bolo dosing, it explains exactly the effect of dosage separation over time, sure they are just looking at the skin but it's happening in all cells in the body because it's oral dosing. I think your missing the forest for the trees, it's obviously stabilising the DNA structure in the skin so it's no reach to say it could be happening in all cells of the body. As "Sinclair" would say it's stopping the "scratching of the CD". Stabilisation is the first step, reversal is the next.
There are entire UA-cam channels based on anecdotal experience with NMN, so until human studies are more plentiful and conclusive, I'll stick with niacin as well.
Hi Rimon, I hope you see this - there's something very important to take into consideration when reviewing mice studies: laboratory mice are bread in an environment that encourages fast tissue repair over lifespan. Due to their fast commercial unnatural breeding, lab mice have extra elongated telomeres (the protein chain at the end of the chromosomes in the cells) which allow quick tissue repair of cells but causes tumor growth when they get old.
Dr Bret Weinstein had made a fascinating work on the subject.
The point is - any product that is aimed to show tissue repair in mice, is already has the cards stacked against it, since it will meet mice that are "turbo charged" for fast cell repair. As far as I know, there aren't any labs that suggest naturally bred mice that will not have this pre disposition.
It is also my experience: a niacin flush doesn’t appear consistently ( 50mg dosage) and I now have a possible explanation by Rimon…

So thing about nmn is I noticed improvements in my vision, all the hair on my body got thicker and longer, my eye lashes have gotten so long and thick that people have asked me if they were fake. I’ve noticed remembering dreams more but not much else. I’m a male btw.

I think we cannot conclude things and say NMN has less benefits based on small population clinical research. I know that NMN is good because myself I have seen many benefits on my overall health as I take it for maintenance and I am 51 years old. I also sell NMN and I have testimonies of my customers saying they are no longer taking their blood pressure medication. some tell me their vision improved. Some noticed that they no longer have prostate issues . Some mentioned their level of energy increased. So told me they have better skin and many people tell them they look younger now. Some reported they no longer have constipation issue. Another customer told me she no longer have pain.

Interesting! NMN fixed my friend’s constipation too. Others in my life have reported more energy and less pain. It helped with my perimenopause and gave me incredible energy and endurance. Lost about 15pounds and my friends have said I look younger too.
Yeah, I noticed that years ago, that women and men responded different to different stimuli. But ultimately, we're ALL different. NMN may help some individuals but not other individuals. Obviously age is a factor, as Rimon mentions later. What I would like to see more of is the analysis of the range of results of particular studies, rather than a focus on the mean (or median) results.

I’m in early 60s male, took NMN by Renue for about a year, felt nothing and quit taking it. Honestly tho I have taken supplements for 45 years and have never felt anything from any of them. Only two things I have ever felt a positive change from and that’s testosterone cream and exercise. I would try Niacin but hate the flush.

Great discussion, but looking into taking nicotinic acid rather than niacinamide: this 2017 study suggests niacinamide may *stimulate* sirtuin activity in cells (Nicotinamide is an inhibitor of SIRT1 in vitro, but can be a stimulator in cells), and other studies suggest nicotinic acid may reduce insulin sensitivity, raise blood glucose and increase risk of diabetes, whereas precursors generally have the opposite effect (glycation is probably one of the major factors in cellular ageing). So I'm not sold on taking regular niacin as an alternative yet.
This is only relevant to high dose niacin whilch works completely different. As you suggested, choronically hurting insulin sen. and blood sugar, and must be cycled - devil is in the details, as usual with every supplement I've researched. it's different supplement high doses vs low doses.
@@WellnessMessiah That seems to be the case for nicotinamide suppressing sirtuin activity as well (mentioned towards the end) - that those doses are in the range of grams (and may still have a net stimulatory effect). It seems the best way to counteract NAD decline is physical activity - which I understand completely ameliorates any age-related effect. And I'd wonder if the next best isn't CD38 inhibition with polyphenols?
I've been striving for better fur colour maintenance for years. Not sure less tail stiffening is an advantage, but then again that was females so who knows! Haven't tried burrowing yet - maybe 2025 could be the year 😂

Were any of the p-values adjusted for multiple hypothesis testing? Of course, if you look for many possible benefits (lifespan, frailty index, fitness, etc.) and do not adjust your p-values, you will end up with some significant (nominal) p-values by chance...

I think it may be because it does not replace exercise but only boosts it in terms of muscle building. People that do not exercise will not see much effect. Unless may be for females, due to additional microbiome effect…
Does slight muscle mass improvement in itself improves longevity? Definitely healthspan, but lifespan I am not sure about. If you are not atrophied, slight variations won’t matter much, probably.

But spike in niacinamide by-product concerns me a bit - suppression of feedback recycling loop and inhibition of surtuins is not what we want at all…
If a company (or a person) doing the study has a financial interest in the results, well, it's not trustworthy. It is essential that independents do the research....Interesting video, I will try taking the niacin. I did take NMN for a while, I didn't notice any changes whatsoever in my body or brain...

My friend took Nichido time for 3 months and her liver enzymes were way high. Her doctor immediately told her to stop Nichido time as she was showing signs of liver damage. Pls be very very careful and don’t take Niacin without checking your liver enzymes. It’s so dangerous!

With a consistent training routine over this time, I have observed significant performance increases in both recreational running and cycling races. My initial thought was the opposite in that I believe the study was designed to fail. Sinclair has been touting MIB-626 which is supposedly far more potent than NMN. And since the MIB company has tried to get NMN banned, why would they design a trial where NMN succeeded? My conclusion is that the trial is worthless.

@@lopezb Sinclair still takes 1g NMN in the evening. If NMN has no effect, why does Sinclair still take it? He specifically stated taking NMN not MIB. That fact and the fact the company has tried to get NMN banned indicates to me that they cannot be trusted to perform a study on NMN.
All the "bad stuff" were quite hidden in small lines or in the supplemental data. The publish the study highlighted the benefits...hidding the downsides. The study headline shold have been "NMN doesn't increase lifespan in male mice, not even with 3.5 grams".
I'm interested in MNM, NAD safety - and supplement quality. *I've never taken a supplement that has had such an incredibly obvious effect: (56 yr old male)* 300mg (Hpn brand) in the morning, I distinctly don't feel tired at the end of the day. As in, not "maybe" - but *definitely* felt better at the end of the day relative to a lonnnng time previously. Same observation the following days, accepted it as a new norm. Big life improvement, actually. I quit taking NAD because I saw a study that implied the anti cell death aspect could increase cancer risk? I again, distinctly miss the effect of it.
I do not get this impression from any B-type vitamin supplement. So I do have a bias towards wanting to be convinced it is safe, because from my vantage point it definitely works....
I would also like to recommend it for my elderly father, but fear for a latent cancer possibility being exacerbated?

I took NAD poster and the year before showed no tumour but a year after it showed a benign kidney tumour..I loved the effect of NMN but I stopped for a year now out of fear . NR gave not much energy compared to NR

The body makes it naturally when you're under biological stress. I guess there was a desire to mimick this effect and see if you get the benefits (not quite, but some)

No they didn't take it with fat. They had quite low fat diet, so even if they drank water with meal (or went to bar...not really) it was without fat. Most of the NMN on empty stomach in this study.
@WellnessMessiah Thank you! But it would be better, to give the mices a food, which is mostly similar to the food for humans, otherwise they can't see the correct results. If humans would eat only water and NMN, they will not life longer too. :-)

If we are getting only one cycle of NAD (and then niacinamide) wont it make sense to either have multiple small dosages (say rather than having 300mg at once have 100mg at interval of 4-5 hrs). Also wont delayed release capsules make more sense.

is funny that they run a study and do not understand the metabolism of NAD. Providing the equivalent of 3.5g per day heavily burdens the methylaion capaciy of the organism, gobbling up all available methyl groups. Such a high dose must be paired with both TMP/methylfolate and glycin, the latter to buffer a potentially ovvershooting metylaion capacity.
Not dong that means that any positive effect of NMN would be reversed.
Second, the pape does not explore the correct context. Boosting NAD is not the same as boosting the salvage pathway. Else, the TCA and the ETC must work as well too. Introducing too much NMN however corrupts at least the ETC.
Sinclair takes Q10 and TMG, btw as well, as he probably understands the energy metabolism. But the researchers of that company: stupid.

It would be nice if those who talk about supplements (and studies etc ) would include "ALL" relevant information. Most will discuss dosage but, what about "very" important information such as take on empty stomach or with food, what time of day, "What form is best". I like many folks have a hard time swallowing pills and prefer to take supplements in powder form. However, I also realize that some forms are better absorbed than others and some aren't absorbed at all and are distroyed by stomach acid ( pills, shots, sublingual, liposomal etc.). Maybe you'd like to be the "FIRST" to give us "ALL" the information we need to understand a particular supplement and how they were used in a study (if known). Thanks Rick B.

It would be nice if they said what NMN was. I've taken a lot of vitamins over the years, but I'm cutting back because I've learned most are synthetic and that doesn't seem good. However, they've apparently helped me a lot. I'm almost 70, healthy, dont get sick, I don't take meds, and though I didn’t take it often, I even stopped Tylenol in 2000. I barely have any gray hair, just a few wisps, or wrinkles. Bottom line, I've done well so I'm going to stop listening about something since I don't know what they're talking about.

Reseveratrol helps . Total myth it doesn't do great things. It's supported in the literature going back seven decades. People like Dr Bradfield.. surface perused reseveratrol and found one study that supposedly undid any benefits. You sir didn't waste your money on reseveratrol..not at all. It's a very very powerful supplement for a whole host of longevity and healthspan issues. I suggest you look for yourself. It's a PDE4 inhibitor, it's a cox11 inhibitor, it reduces neuroinflammation gut/ intestinal inflammation, it specifically increases NAD+ in heart tissue, it reduces bad cholesterol, it reduces artery stiffness, mends the blood brain barrier..this has been found time and again in peer reviewed studies. Hey Guys, Please consider that you might make more connections on this. I think Rimon's perspective is awesome. But, if I am a 60 year old man, already doing some HIIT and IF and I can shift my mitochondria to more fat metabolism, even 3-5% more or whatever it was...I can either reduce calories, reduce carbohydrates, compensate for a slight "overconsumption" of nuts and coconut oil (Perplexity agrees, helps mobilize phytochemicals from spices)...I can get rid of garbage starch calories....I know when I did long distance cycling on The Zone...two consecutive 200 mile days consuming very little food...it could be that the paleo/keto guys aren't completely insane... I am thinking to dump white rice in favor of vegetables, almonds, walnuts, sunflower seeds, avocados in season, etc., because those are certainly better quality calories....does that make sense? I know you guys can do a better job on this line of thought.
Also, if I think my bio age is more like that of a 50 year old :) maybe niacin is a good first cut?
