I have two questions. 1) Should I put 0.05 mL of cell in a new flask in order to set 1 × 10⁵ cells/mL without media according to the number 2 problem? And did you mean the flask is a T-75? 2) Should I consider the dilution factor, which is 2, after knowing the live cell concentration by using a cell counter? This is because a person might put 10 uL of cell and 10 uL of Trypan blue in a C-slide before using the cell counter. I appreciate your video, and it is extremely useful. Thanks for sharing!!!
Hi Seulki, I'm glad that you found the video useful. In answer to your questions: 1) That question is talking about a NUMBER of cells (1 × 10⁵ cells) NOT a concentration (1 × 10⁵ cells/mL) this is because often when you subculture adherent cells, you work with the total number of cells, not with concentrations. The flask you're putting it into doesn't matter for this question. The question only asked you to work out what volume you would need to get a specific number of cells. If you were putting these into a really big flask (T-125 for example) then you might need to put more cells in e.g. 5 × 10⁵ cells - but again, this is a number, not a concentration. 2) The dilution factor will need to be considered when you are first calculating the concentration of what you have. Automated cell counters usually do this automatically before they spit out the number, but that is something that you will have to check. If you are doing it by hand, then yes you will have to consider your trypan blue dilution to calculate your CURRENT concentration, before you do any of the calculations in this video. Hi! Thanks for your question! The calculation to get a certain number of cells is at about 2 minutes into this video. The number you will need depends on 1) the size of the well (e.g. 96-well plates compared to 6-well plates), 2) How long you're going to be growing/incubating them for, and 3) the type of cell e.g. you will need fewer big cells compared to if you're using a small cell type.
