Notice the lack of white sclera around Mario's eyes. Also, the crystals indicating health are absent.
You can briefly see parts of the health crystals at 1:12, right before the person playing uses the prototype's "extra health" feature. It looks like the crystals were either cut off-screen while the footage was being recorded, or the glitchy sprites they turn into when using that health cheat blended in with the background

When she’s talking, the movements look too smooth to be stop motion, so I want to say she’s a rubber puppet, but the movements seem too precise to be a puppet. So while I’m pretty sure she’s some kind of puppet, I’m still not 100%. I think when she spits out the egg it’s stop-motion, but once again I’m not 100% on that.
very late reply but im 100% sure thats a rubber puppet , with one person controlling her full left arm and head , while potentially someone else puppets her right arm and tail , and then the second bit is just a simpler (probably smaller) puppet
