When did sex and gender come to mean different things? It's been historically understood that both terms, sex and gender, referred to male and female, but gender was specifically applied to grammar. In this video, Dr. Michelle Cretella and Dr. Ryan Anderson talk about the shift in how these words have been and are now used in society.

Actually grammatical gender is more complicated than that. Many language have neuter genders and non-Indo-European language have other gender categories like state of mater(liquid) or animacy(sentient).
More to the point, while language can be used to shape perception it is more often used to reveal what was already there.
Before the invention of the word “homosexual” homosexual people existed but the concept had not been solidified.
The use of the term gender is not a plot, nor is it being used to make some sort of spiritual claim about trans people.
A good way of thinking about it that gender is to sex what psychology or sociology is to neurology.
The two are connected but distinct.
Many gendered expectations might have have roots in overarching trends of biological sex but much of what is considered manly or feminine is factually and knowingly arbitrary.
That doesn’t mean it’s necessarily wrong, most culture is arbitrary.
But the fact that our gender expectations are being challenged shows that our culture is not compatible with the culture that preceded it.
As far as trans people are concerned, how you act, behave and are viewed has a lot to do with gendered experiences.
People don’t check DNA when they open the door for a woman they just open the door.
The reasons for gender transition are complex and diverse but to pretend as if nothing changes is to ignore the reality of how gender works in our culture as well as most others.
