you have to kill all of the amanecidas and then kill the final one (located in the top room in the ferrous tree) and then once you kill her, i think you are awarded with the prayer in a nearby room. you probably already researched this but just in case there you go

How difficult is the game compared to Bloodstained?

pretty easy since you can block,dash and parry but the boss mechanic are quite difficult
