How to Make Petri Dishes for Mushrooms (Instant Pot Method)

  • I uploaded a video a couple years ago about making petri dishes with no pressure cooker. I like that video and while it is one of my most viewed videos, I do have to admit it is a little overly complicated for what it is. In this video I make enough agar to fill 15, 100mm petri dishes using an instant pot. I'm currently in the process of trying to put together a method of making petri dishes from entirely grocery store sourced materials, but for now, at the very least, all the components used in this video are easily purchased on amazon and this formula can be infinitely scaled up or down, and customized to your specific needs.

    400ml water
    10 grams nutrient Powdered agar
    10 grams Light Malt extract powder
    Less vital nutrition additives
    1 gram yeast extract
    2 grams potatoe starch

      yours has an even higher nutrient profile than mine (only due to less water)
      500ml H2O
      9g agar
      8g ELME (Extra Light Malt Extract)
      2g raw sugar

    Exactly what I was looking for, thanks! Finaly just got some 250ml media bottles that fit my Instant Pot. Needed info on cap tightening and was looking for a potato agar mix....I got both in one. :) A little tip about agar powder I got from a Japanese chef I knew...if you hydrate agar with cold water for 15-20 minutes before adding hot water/ingredients you will greatly decrease clumping, sediment, and cloudiness. I just take about half of my water, cool or even cold, toss in the agar power stirring ocassionally while I get my other stuff together 10-15 minutes. Then I warm up the 2nd half of the water in the microwave and stir in my sugars/nutrients to let them dissolve. Then, you temper the liquids by slowly adding the hot to the cold and either toss in on a stir plate or pop it back in the microwave (being careful to not overboil.). Sounds more complicated than it is, but I went from cloudy with sediment to clear and fully dissolved instantly. Thanks again for the vid!

    You are a godsend. Your videos are very informative. You helped me jump the gun on making my own agar especially with this instant pot method I can’t afford the expensive sterilization machines . God bless you 🙏🏾

    I'm in York county. I did 11.5-23 months in York county for growing mushrooms. This video is relevant to my former interests.

      Hahaha you do with this information as you please. No judgment here. I never trusted my brain enough to try any mushroom other than the kind that taste delicious and don't make the walls melt lol.

      @@randomergy683 That is the beauty of mycology. Most of the tools, knowledge and technology are interchangeable between all fruiting bodies. Agar plates are great for isolating particularly nice strains of mycelium and are generally interesting to study.

      @@TechGorilla1987 Watching mycelium grow on agar is one of my favorite parts of mycology. I don't know why but I've always been fascinated with watching things grow on petri dishes. I want to do some stuff with slime molds this summer if possible

    Just curious why you chose the trifurcated plates, are you triple inoculating them?
    Also, I have those same media bottles with blue caps, they work great!

      I wish I could say I had a better reason but to be honest, I found these ones on ebay which were cheaper in bulk compared to the standard plates lol. Plus I like the way they colonize, essentially having three smaller petri dishes in one.
      I really like the bottles too. I broke one last month and I was really sad haha. But I'm going to buy a bunch more here soon so it wasn't the end of the world.

    Jokes aside, thank you for the video! I'm foraying into mushroom growing and this is very helpful, since premade "sterile" agar plates are ridiculously expensive (and possible not sterile).

      Hahaha the cheapskate version of an instant pot I guess would be more accurate. I found it at my local surplus store for 30 dollars.
      But thank you. I'm glad my video was able to help you. I made it for that exact reason. Pre-poured plates are way too expensive for the amount I go through. So I've always just made my own. Plus I like being able to control what I put into the agar solution. (:

      Thank you for the comment. (: I have an older video where I make petri dishes just by boiling the agar solution in a glass flask. I'll link to it in another comment if you're interested. I think I'm going to redo it though to make it more accessible to people who don't have borosilicate glassware and the like. (:

    interesting that you pour your agar without using a flow hood OR a still air box. what percentage of them would you say get contam'd this way?

      Before I set up my workshop the way I did I did everything in my dining room. Even then I still had a pretty good success rate. Maybe 10 percent would get contaminated. Now with the set up I have and the methods I use I have over 99% success (:

    Was the pressure cooker at 12 psi or was it higher?

      I can only go by what I've read on electric pressure cookers online but when you put this specific pressure cooker on sterilize it goes just above 12psi. Which seems to be more than adequate for my needs (:
      I wish I could give a more precise answer but this cheap electric pressure cooker I bought doesn't really give any details about the psi levels they use heh

    This should be done in a still air box or laminar flow. Petri dishes and agar bottle shouldn't be opened in open air like this, airborne contaminants can easily be introduced

      Ideally yes but not everyone, especially in the home setting is going to have access to those. So I wanted to show how you can still do this in an amateur setting. I also set my shed up to be sealed up specifically for doing this. 3 HEPA air filters running 24/7, except for when I pour agar or do culture transfers/spore inoculations/etc, as well as treating the air with ozone every 8 hours. Along with doing the hot agar pour. Its not a perfect method but its good enough for the home setting. Theres enough stuff people who want to do petri dishes need to buy when first starting out. This method is more accessible to the amateur and I generally have a 99% success rates as far as air born contaminations go. It's a little more tedious constantly making sure your space is clean but it's much more doable for the average person. All the petri dishes I use in my videos are made using this method.

      @randomergy683 3 HEPA filters running 24/7 and treating the air with ozone sounds way more expensive and less accessible than a 5 dollar tote with 2 holes in for SAB 😅

      @@famouspigeon I used to use a still air box but it's much easier to do it that way and I can record things easier lol. But yeah, a tote is way more accessible.
