Sometimes if you have a leaky valve you can tap it with a key or a screwdriver really fast and it blows crap out of the seal and it reseats and stops leaking.

Hi sir today i just check the refrigerant level in my car by using a gauge and it was good. Car is 5 years old. I doubted if the schrader may got damaged by the gauge fitting. And after driving my car for a while i checked the port cap it was too tight now i had just hand tight it but i had to use pliers to open it. When i opened i heared a hiss sound.
So my question is can the port cap which has a o ring inside prevent the refrigerant leak if there is a leak.

No. Your hiss sound is due to a leaky Schrader valve which needs to be replaced.

"just check the refrigerant level in my car by using a gauge and it was good" - Sorry, a gauge cannot really determine refrigerant level; only real way is to have special equipment recover and weigh it.
