Is it better for fast colonixing to use 5g of ME or to use 10g, which would develop a more ryzo growth?
Depends on the volume you're looking to produce. My research suggests 5g of ME per 250ml of water.
Hey I have a question. My pre mixed MEA agar mixture from Amazon is 56 grams. It says on directions to dump it all into a liter of water. It just seems over kill compared to what most people mix in their ratios.. but I’m thinking about fractionalizing the amount while still keeping the same ratio. I’m just worried it either won’t set good or it will be too jelly. Thoughts? Maybe do like 28 grams per half liter? I hate to use the whole bag at once when I really only need 10 plates or so
Most pre-mixed media has an agar content of 1-1.5%, which is about the minimum concentration for solid plates. I would use the mix at the suggested concentration unless the agar percentage is specified, in which case use at least 1% agar.
@@julianliber4633 okay perfect. Can I store any excess leftovers back in the fridge with the caps covered so no moisture escapes,until I need again then I can just re sterilize and use again? Thanks
@@dressedanddepressed2301 The powder is stable for a while, as is the sterilized liquid/gel if kept in a sealed container.

Hi i Have a question: can I use maltodextrin instead of malt extract?
This would likely depend on what types of carbon sources can support your organism's metabolism. Most fungi will happily grow on dextrose, so it will probably work just fine.

Yes, that will work.
Hi, how do I know the percentage/concentration of the MEA+YA?
You can calculate it based on how much medium you make. These percentages are expressed as weight per volume, which means 10 g per 1000 mL is 1%.
Which kind of malt extract use
Diastatic or non diastatic
It's just being used for sugars and will be autoclaved, so it doesn't matter. However, I use dry bacteriological malt extract which is presumably non-diastatic due to heat treatment.
Is it okay to also add antibiotics with this media?
Yes, filter sterilize then add after the agar cools to 65C or less.
It is useful for plate total count?
You can use these plates for counting colony forming units, usually with fungi.
Do you have the tutorial video how to make Yeast agar media for total plate count?
Can you explain the benefit of yeast extract? Mycelium seems to grow just fine in a plain MEA or Dextrose Agar, but does yeast extract provide a benefit to colonization speeds? Been wondering this about peptone as well...
Peptone and yeast extract add phosphorus, nitrogen, sulfur and other nutrients required for growth. Lab-grade malt extract and potato dextrose are really pure, but cruder grades might have more of these nutrients and thus don't require supplemental nutrition.
Just curious over the year since that question, what is or was your go to agar.

That means that the process likely did not stay sterile. Were your hands washed and sterilized? Were your Petri dishes in sealed, sterile bags? Did your medium get heated sufficiently in an autoclave or pressure cooker? Did you pour the plates in a sterile environment? Did you wear a facemask to prevent contamination?
@@julianliber4633 i never touch the petri dish.., i just simply pour the agar mix and done, then after a day..., they spoil...
i mean, i pour some alcohol and let it dry
before pouring the agar mix...
How do you heat your agar mix? You need to autoclave or pressure cook it at 1 bar = 15 PSI.
@@julianliber4633 stove,......., should i cooked it more THAN 30mins?? agar agar + wheat powder... and cooked it on stove for 10mins.... thats what i do
@@marjhuncantago9476 stove top is not hot enough. You need to use a pressure cooker to increase the boiling temperature.
You’re only using 5 grams of agar?? I’m using 10
1% agar is perfectly fine for mushroom culture. It's also easy to cut for transferring plates.
