    From age 14 to almost 59 now, I've just continued to lift heavy, working around injuries etc and staying on point with recovery and nutirtion. If continued lifting is possible for you, just do it.

      The concept here is about a once a week supplement that potentially could prevent injury as well as other potential benefits including reduction of misfolded proteins. If the targeted 24 hours of catabolic state doesn’t throw off the 144 hours of mostly anabolic state, it’s the best of all worlds.

      @@DrBradStanfield But that's not how rapamycin works (24/144h).
      First, the half-life of rapamycin is about 60 hours, which means it won't be fully out of your system for almost two weeks.
      Second, the duration of rapamycin's biological activity, particularly in terms of mTOR inhibition, is not strictly tied to its half-life. mTOR activity is typically suppressed as long as rapamycin is bound to its target, which can extend beyond the plasma half-life due to the drug's binding affinity and tissue distribution.
      Generally, significant mTOR inhibition is observed for about 24 to 72 hours after a single dose of rapamycin, depending on the dose and individual response. After this period, mTOR activity likely begins to recover, returning to more than 90% of its baseline activity about 4 to 5 days after a single dose for most individuals.
      For this reason, dosing every week may not be the optimal strategy for balancing mTOR activity and muscle preservation. Do we really need a "spring cleaning" every week, Brad? I'd consider having a trial group doing dosing every fortnight or even monthly for comparison. Happy to participate. 😉

    We need to see a meta analysis on human after 2-3 years. Understanding the cancerous effects etc is vital. Guys do not jump in every hype and do not buy the fomo. Good luck !

      This drug has absolutely horrible side effects. Would be interesting to see how this would be any better than fasting. If I'm not mistaken, fasting had the longest life extension in rats ever recorded.

      @@CommanderRiker0 Only if started at a very early age, otherwise the lifespan extension effects are marginal at best. Fasting by itself induces muscle wasting while rapamycin once a week wouldn't have thaf effect.

      @@CommanderRiker0 They generally use a low dose for life extension investigation. Nothing like they use for organ transplants. Some people get mouth sores, that is about it. Typically.
      And no, it was not fasting, it was calorie restriction (CR). This is a very unpleasant lifestyle. Some people are doing this. Bowl of frozen berries in the morning and a small lunch with some beans...starving the rest of the time.
      You are never going to get millions of people onboard with eating 30% fewer calories than their weight and height suggest they need. And that can be dangerous as you have to get all the nutrients your body requires. That means fairly close doctor supervision.
      That said, there are other choices that have been very successful in rodents: Acarbose did great, and Berberine did great. Probably doing the same thing. Rapamycin is likely a different mechanism, but perhaps not. I hope he measures body temperature, as, I suspect, body temp is really how the vast majority of things work in rodents.
      Could fail in humans because humans will just wear more clothes, put more covers on the bed or turn up the heat and such to stay warm.

      @@volos_olympus They have gone back in forth on the mechanism of this drug between mtor and other pathways. Most still believe its mtor, which fasting directly acts on via calorie restriction. If that's true I would rather fast since fasting has other benefits as well. I am very doubtful this will beat fasting in any meaningful way. Hopeful, but I don't see it.

    Rapamycin is synonymous with sirolimus, a drug mainly used in organ transplant as it suppresses (I should probably say “erases”) immunity. It’s also known for causing hypertension, non reversible vascular kidney damage, and for its heavy metabolic side-effects (frequently even causing diabetes mellitus).
    This safety study should be interesting, provided the patients are followed for long enough to see if the dose difference keeps them free from those side-effects, as the molecule is meant here to be used for longevity.

    At point, this is a hypothesis, only. Because it's a self-report format, it's not clear whether there's a verification of compliance. Most folks 65 and older also have polypharmacy regimes, which is another variable. Diet, gender, ethnicity, race, pre-existing conditions, are all variables that have to be assessed and accounted for. We haven't even seen the pre-test, post-test protocols, yet. If and when the results are published in a reputable peer-reviewed journal, I'd like to hear Dr.Nicholas Verhoeven's analysis over at Physionic. Analyzing the strengths and weaknesses of published research is his thing, and he's superb in that realm. Until then, Dr. Stanfield has an interesting hypothesis, and perhaps an interesting pilot study, but too much is unknown and unknowable, at this point, for some of accolades in the comments. As is often said, "The Devil is in the details."

    My concern is that Mtor may not be reactivated once you stop using rapamycin. Like the testicles shutting down with TRT and not potentially reactivating once you stop TRT.

    Yeah, but it doesn’t matter if it works-- no doctor is going to prescribe rapamycin to a healthy adult. Also we need to know if it works without exercise because it actually may be the exercise which is making the difference.

    well I am 69 and i am stronger than most people half my age, pity the study wasn't here in Spain.
