Great video! In the case of testing using direct IF for a disease such as Pemphigus Vulgaris, what is the antigen & what is the antibody? Which one is taken from the human sample?
It is an autoimmune disease. Antigen- Desmosome (desmoglein 3 protein) Antibody- IgG in serum(suspected). Confirm by looking for fishnet/ chicken wire pattern

Why to use blocking solution before the primary antibodies? Is there not a chance you will be blocking the protein you want to find? Or it is just worth it, because even if you block some of the target proteins, the primary antibody will just do better in finding the not blocked target proteins, than finding inespecific conections?

Sir.. We students are in serious trouble... Mh set exam conducted by SPPU university some days before for asst. Prof. In Maharashtra and goa... They published intrim answer key two days before... And there are 5-6 question are such that we can challenge them... So we want u to make video on that 6 question... Bcz when we challenge a question we have to give correct explaination from std books.Plzz sir help us..give me ur whtspp nmbr or email id so i can send u those question
