Hi, Thank you for this great lesson. Can you give me help? I'm doing my presentation about HIV-1, and I don't know to detect p24 antigens, we would use Sandwich indirect or direct Elisa?
Sandwitch Elisa would be most appropriate, othet wise direct Elisa may be used

Sir well plates is used in so many experiment are reusable or it requires new plate for ech experiment?
The ELISA 96-well microplates allow performing the common laboratory testing procedure that can be carried out on multiple samples at once.
Sir ELISA test or EIA test boths are same test

Hi sir I am form karnataka,25 years old i have one doubt.
While performing the hiv test with test kit i toughed the area where sample is collected with pricked finger and give the blood. At the time i toughed sampale collection area accidently. I don't use pippet(dropper) I heared live antigen of hiv in the test kit.
Now I worried about this.
Can Hiv is spread by test kit in such method?
I am at risk of getting hiv.
Please sir answer this🙏.
HIV can only occur if there is a transfusion of a positive blood with negative one. If that happened in your case then get it checked out.
Awesome explanation sir 🥰🥰🥰

Thanks for your honest opinion....in future videos I would try to simply the material even further
@@animatedbiologywitharpan thank you for answering me.
Hello what is the substrate used after secondary enzyme linked monoclonal body.?
Some commonly used substrates used in ELISA applications are hydrogen peroxide, luminol, and P-Nitrophenyl-phosphate (pNPP substrate
Plzzzzzzz send the more videos
To detect the presence of hormone we use sandwich elisa but can we use competitive elisa for that?
Not really recommended

I have one doubt.. In elisa test y we will prefer only serum.. If plasma will possible for test.. in case plasma will we prefer what reaction will happen.. Can you explain please
In order to prevent blood clotting during the preparation of plasma anticoagulants commonly are used (like EDTA or heparin).
In most cases, anticoagulants affect the interaction of antibodies with antigens, so most test systems designed to work with serum, but not with plasm
Which test is best for hiv diagnosis?? Among all hiv tests ...
ElISA is the best way

Ok I will make a better video on competitive elisa
I agree, I was so excited to have found the perfect video until we reached the competitive ELISA part, still thanks alot sir, it was very well organized and broken down into easy bits of information.
I don’t like how HIV is confused with AIDS … or refered to as aids in this video
It’s [HIV] not at all being confused with AIDS, HIV is essentially a pre-cursor to AIDS, AIDS is the final stages when the HIV infection becomes most prominent; there are associated decreases to the CD4+ cells, greatly reducing one’s immunity, allowing them to be susceptible to opportunistic infections that the normal person wouldn’t be. Thats how it was discovered; young people having cancers that weren’t typically seen in their age demographic, I.e more of those in their 80s… LONG STORY SHORT, AIDS is HIV, HIV is not CATEGORIZED as AIDS until there is final progression characterized by low CD4+ (T-cell) counts, typically around 200/uL (vs ~1000/uL) , so it’s again the final stage of the infection from the virus, therefore the same thing. It’s a 5-minute summary video, in which he did a great job. 🎉😊
Illiterate 🥱
whats the difference between direct elisa
Direct ELISA directly uses labeled antibodies for antigen detection, while indirect ELISA employs unlabeled primary antibodies and labeled secondary antibodies, amplifying the signal for increased sensitivity. Direct ELISA is quicker but less sensitive, while indirect ELISA offers enhanced sensitivity at the cost of additional steps.

What is the substrate in this case of ELISA? Is it the patient's serum?
ELISA is done on the patient's sample which has the antigen or antibody. If you are asking about the substrate for color reaction then its different
Yes that's what I mean. What is the substrate for color reaction?

the washing process isn't needed?
I agree to your point. Acute this video was an old video , the wash step and the necessity of it should have been highlighted.
Why you can't explain direct and Mac Elisa
you should practice it first so the explanation is fluent.
Yes my friend, thanks for your suggestion. I will incorporate
Good, but don't explain the direct method
Thanks for your detailed feedback
Why only serum is used
Serum, the clear liquid portion of blood after clotting, is typically used for diagnostic purposes instead of whole blood because it lacks clotting factors, which could interfere with certain tests. Serum provides a clearer sample for accurate analysis of various biomarkers, such as proteins, enzymes, and antibodies, aiding in diagnostics.
