I know this an older video but in the future if you're going to rescue a contam plate you could try and drip some paraffin wax from a tea light candle over the contam BEFORE opening and closing the petri dish ...just as a precaution so as not to disturb/spread the contams. No guarantee but it's worth the effort if you're trying save good genetics.
Wondering if by opening the lid so many times it kinda fluffed the mold a bit on the last few plates? I'm thinking if one was to cut out the mold first then attack the air with rubbing alcohol then work the transfers if that would work cleaner? Just my 2 cents I'm still a wee little rook in the game of spores....
Or of you were to let the mycelium over run the mold if that would keep the mold spores down while you transferred a clean spot of mycelium off the plate...
Great comment! I encourage you to try that and let me know how it goes. My thoughts on that are... If you cut out a sporulating mold you are at much higher risk of spreading spores. think about where you would put the mold when you take it out... any method of dumping the mold will probably dislodge thousands of spores. the best option is to trash the plate without opening. the next best option is to take a smooth cut from the cleanest section and move it to a clean plate.
This can happen for sure and its fun to watch. It is best to do the rescue subculture as early as possible to reduce the number of spores in the plate and reduce the time they have to spread in the plate. i have another update coming where i was able to rescue before spores formed on the mold. thanks for the comment. glad we could stay in touch.
@@DavidJones-il5tl I've had strong mycelium overrun contam. It seems to get denser and try to "capture" the contamination. It certainly would reduce the chances of cross contamination if one waited until the contamination was covered. However, I've also seen cases where the mold beats out the mycelium so I try to grab the mycelium as soon as I notice contamination in the dish.
I would be interested to see how throwing a cap such as a plastic bottle cap or something similar first thing over the contam then transfer to reduce any spores that can float on you, might be trying this myself from contamd spores I'm going to try to isolate soon

Not sure why you would have had plates 1 & 2 and your original in your way while doing this.
It is to show the progress. This is purely for demonstration. I would normally use a SAB for this work.
