    Metformin, Rapamycin Studies |TAME Project : This is the part 2 of our interview series with Dr. Nir Brazilai from Albert Einstein College of Medicine.
    Dr. Barzilai is the founding director of the Institute for Aging Research at the Albert Einstein College of Medicine and the Director of the Paul F. Glenn Center for the Biology of Human Aging Research and of the National Institutes of Health’s (NIH) Nathan Shock Centers of Excellence in the Basic Biology of Aging.
    Dr. Barzilai discovered the first longevity gene in humans and has since discovered several others. In addition to his “longevity gene” research, Dr. Barzilai studies key mechanisms involved in the biology of aging, including how nutrients and genetics influence lifespan. He is also investigating the physical and mental declines associated with aging and how they affect longevity.
    He is the author of over 270 peer-reviewed papers and a recipient of numerous prestigious awards,including the recipient of the 2010 Irving S. Wright Award of Distinction in Aging Research and is the 2018 recipient of the IPSEN Longevity award.

    I take both metformin and weekly rapamycin. The metformin has a side benefit of lowering blood glucose that the rapamycin tends to raise. It seems that rapamycin has helped lower my fasting glucose level but raised my post prandial glucose levels. I use a glucose meter to carefully monitor glucose after meals and I just don’t eat the foods that raise the glucose higher than I desire.

    I make smoothies with blueberries, banana, strawberries, dry apricots, avocado, and brussel sprouts, or broccoli sprouts. I add Japanese matcha, spirulina and chlorella, and chia seeds powders also I put a tablespoon of coconut sugar.

    Thank you so much for bringing this vital information to us. I've seen a couple of your other videos and progress. I don't want to discourage the use of metformin but, I read that if the kidneys are not functioning properly, metformin can build up in your system and cause Lactic acidosis. I was just given my first Metformin prescription, and will start my dosing very soon.

      Drinking too much alcohol is the main cause for build-up of lactic acidosis.

    Dr Barzilai said the billionaire who funded him was starting a foundation to fund anti aging at $1 billion per year and would be announced in January 2021. Anyone hear anything yet?

    I'm 82 and started taking metformin before it was legal in the US. I recently took my first dose (6mg) of rapamycin and intend to repeat that dose every 10 days. Am in good health except for asthma and am mega-dosing on vit D3 in hope of controlling my asthma

      Iggie, I would like to do the same. I’m 84, quite slim and fit but take Metformin 500mg.. I’ve seen the results with mice when rapamycin is added just a few times a month.. Hopefully my doc cooperates.

    Two questions: How does evidence the Metformin make the cells younger? Do you have a standard test to get results, do you found that individual genetic variants affect the results??

    I suspect the main risk factor is not aging but the food that you have eaten age 35-60. Smoking , alcohol, carbohydrate consumption, insulin resistance etc are all likely more important risk factors for the diseases at the start of this video.

      I concur with your comment regarding food. I base my belief on the fact that my mother lived until 107 years. She lived most of her life without gmos and pesticides. She did not eat processed foods. She prepared salads with all dinners served with meat and cooked fresh vegetables when raising her family. She never ate fast or junk food. She never required prescription drugs and did not have dementia-just had major short-term memory issues. She walked for her exercise daily.

    Ashwagandha, Triphala, Haridra. These are prescribed in Ayurveda for longevity. There are cases of humans living more than 200 years in India taking these. But there's a way to take them - time of day, in what form they are taken (in alcohol / with pepper and other herbs etc).

      @@ModernHealthspan Yes, both are prescribed. Primary motivation is anti-aging. A few Drs in USA give that prescription (e.g. Dr Alan Green from NYC,
      www.rapamycintherapy.com) but more and more Drs are interested. My Dr thought Rapamycin should help with things like fatty liver, metabolic syndrome (obesity etc), insulin resistance etc.
      * While on Rapa, my liver markers (ALT, AST) improved and they're the best they're been. HOMA-IR (Insulin resistance) score improved from 5.something (severely insulin resistant) to 2.something (just insulin resistant). Btw, you should get your insulin tested if you aren't doing that already. HOMA-IR is calculated from fasting blood glucose and insulin.

      (There are 10x more papers, but these are the most comprehensive analyses).

      * Another supplement I think very highly of is Glucosamine. Extends life of nematodes, mice and people (observational).

    I tried to get Metformin from my doc but he refused because he says is bad for your kidneys...

      Unless you have a specific kidney issue, get a new doctor. As a 7 year diabetic, and several years before that pre-diabetic, I have taken metformin for 10 years, no issues, smaller doses in the early years but now 2000mg xr daily, excellent energy levels for a 69 year old, hba1c 5.7. My doctor tests half yearly for kidney issues.

    Another "risk" to the government is a public policy issue, i.e. if we increase average lifespan in the US population, the Social Security program would quickly go bankrupt. This is also true of corporate and union pension plans.

    Unfortunately I’m in my 40s in the US, and I don’t have diabetes, and a internist MD / doctor who asked me this Summer what a microbiome is. So, I have to take Berberine instead. I really wish doctors were keeping up on these studies, especially since drugs like metformin have been around already for 60 years. Does anyone know of any doctors in the US who are willing to have remote patients and give metformin? I’ll even pay out of pocket. Or a reliable manufacturer who will sell and ship internationally?

      I don't want to mention the company outright but just do an internet search on aging and metformin. You'll see a company/practice based in South Carolina that is a wonderful source.
      You'll fill out a medical questionnaire and a practitioner (doctor or NP) will review and prescribe metformin for you if it is indicated.
      Furthermore, it is really inexpensive.

    This all great. Thank you for doing it. We know though regulators love to regulate. If we had the cure to whatever ails you, regulators would want to regulate it. So how are you going to get around this nonsense and broken academic medicine? Please let make suggestions as follows: 1) For each center, publish the cost per patient. 2) For each IRB, publish the time it takes from protocol submission with consent to time of approval. 3) List the IRBs that refuse a single common IRB (so they can get the IRB smarty pants award and run up bills).

    I was on Metformin 500 mg for 9 months and my creatinine levels increased to abnormal levels. Not good for my kidneys.

      You are taking Metformin because of diabetes? Diabetes is bad for the kidneys. Need to get diabetes under control. May not be the Metformin. Since you have stop the Metformin has the creatinine decreased?

      @@ellenlee1 I do not have diabetes. Thank God. I was taking it as it us being purported to be an anti-aging drug. And yes, my last blood test showed by creatinine levels are normal. I wish Metformin didn't increase my creatinine levels. Thank you

      @@barbarawalker4292 In the United States you need a prescription from a Doctor to get Metformin. He went off label and gave it to you? They need to study this for non diabetic people, I am guessing the drug has been out 60 years but only diabetics have been using it. Life expectancy is a lot less for diabetics, if those diabetics that took Metformin live longer than those diabetics that did not take Metformin than anecdotoly I would assume it's does something. There are millions of diabetics on Metformin. Get some statistic from this. Medicines are all statistics. Maybe for non-diabetics it increases creatinine, others it has no effect on the kidneys. Good you had your blood tested by the doctor. Most people Metformin effects the Liver.

      @@ellenlee1 I live in the U.S.A. and she did give it to me off-label. Many doctors do that here. I have had Botox injected in my skull for migraines and it worked. (That is off-label). No more migraines. I don't know what you mean by "Get some statistic from this." The introduction to this UA-cam video alludes to Metformin used as an anti-aging drug. The TAME project "Targeting Aging with Metformin" is interesting and I had hoped it would have helped me with anti-aging, but I cannot take it. It is well-known here in the U.S.A., that Metformin MAY increase creatinine levels in SOME people.

    I took metformin for 2 years prediabetes I hate it. I had stomach issues, and affected my Testosterone I threw it in the garbage. I prefer to take berberine, and 100% organic extra virgin olive oil before going to bed.

    You know it is not possible to get metformin without doctors prescription. Can I consider a berberine as 100% metformin's substitution?

      I get mine from agelessRx.com. In about 6 months I'll ask my regular dr. if they'll prescribe it for me since I'll have already been on it, but if they say no, then I'll stick with these guys.

    It's such a shame metformin is prescription only. I'm taking berberine now but feel it's a bit of a poor man's cousin in comparison

      I would get metformin. Nobody knows if berberine will be life extending. Evidence for Metf. is fairly strong. You can always look for an open minded doc (aka anti-aging Drs).

      As long as it's doing its job of lowering blood glucose, it doesn't really matter. It will stave off diabetes which is the main culprit of all these diseases.
