I don't understand the question completely. This was done using an emulator, since it's a Tool Assisted Speedrun. The runner most likely used the original ROM, and not a hacked one. It would make this run kinda pointless, I imagine.
There's a right order to do everything, but you're never punished for going out of order. You just get a checkpoint you can return to when you come back tot hat level, so there's never any backtracking.
