  • WHAT IS THE mTOR PATHWAY? And what effect does it have on aging? And how to balance or inhibit the mTOR pathway. These are some of the subjects we’ll be discussing in this video.
    The mTOR pathway plays a major role in aging, and in slowing aging. We’ll go into the science of the mTOR pathway, as well as strategies for balancing mTOR and AMPK…another aspect of the nutrient sensing pathway that is closely associated with mTOR. Both of these pathways need to be balanced to achieve the best results.
    We’ll also be taking a look a autophagy and the role that it plays. So join us for this enlightening discussion.

    Just found you. Super lucid explanation. I'm 82, need no meds, do 15-9 and love and indulge a passion for red meat frequently along with otherwise whole foods, Science does not yet know timing but if I'm the test rat my system seems to work. However, if important, know that I am not a body builder but a regular walker and swimmer. cheers and well done; that's a compliment from a crusty old skeptic.

    Are there studies about protein cycling over a week?: no to low to normal to very high - repeat; thus keeping the average ratio constant.

    how to measure the relative activation of mtor or ampk, well, Ii guess, that's the billion dollar question. ...
    I guess, a couple of things need to be tracked, yet I would start investigating the relative proportion of reduced and oxidised glutathione in a dense monitoring, almost like glucose in CGM. A second proxy for autophagy would perhaps be ketones.
    personally, yet i am 62 now, i experienced much stronger muscle growth if i dedicatedly take more protein between 30..50 h after workout. the third day then i fast and do the workout fasted, once a week then... at least that's my goal.

    We fiddle around with our natural biochemistry at our own peril, especially something as complex and poorly understood as the mtor pathway. Everything in life is about balance. There are times of feasting and times of famine. We are built to endure both.
    I think bodybuilders have the right idea with cutting and bulking. It's not only a great way to build muscle while staying lean, it balances activation anabolic and catabolic pathways. I think eating a caloric surplus a few months a year, then fasting and eating a caloric deficit a few months a year is a good way to strike a balance.
    It also follows a natural seasonal cycle (winter/summer, wet/dry season) we probably evolved to endure as seasonal food availability fluctuated throughout the year.

      I think you're on to something here. I think the same. Like everything else in life, it's all about balance. It's when you take things to extremes that you get in trouble.

    Likewise, cycling between feeding microbiome and the mycobiome in balance, keeping each in balance is another area to explore further. An occasional purge and regrowth of each biome may be beneficial too.

    Do cardio and weight training on alternate days.
    Weights in the morning and cardio in the evening.
    Wait till noon to have the first meal. Only proteins + fats.
    Eat the second meal after the evening cardio session.
    Carbs + protectins.
    Than there may be a balance between mTor and AMPK.

    Thank you for unpacking the definition of mTOR so brilliantly.
    I am curious about how a human body that has never taken Rapamycin exogenously (and does not make it endogenously) can have a pathway for said Rapamycin...

      The pathway was unknown until it was discovered that it was targeted by rapamycin. So, the pathway was always there, we just didn't know about it.

    Hello Lance. Siim Land's book Metabolic Autophagy has some interesting page's about this subject.
    My personal strategy. On days that i workout i want to stimulate mTor and i eat protein with carbs about one hour after the workout ( i workout fasted). The second meal of the day i eat protein with fats. With this meal i take Berberine, ALA, ECGC & Curcumin to activate AMPK again. On days that i not workout i fast 18 hours and have two keto meals. I take the same supplements as mentioned before to enhance AMPK.

      I'm a big fan of Siim. But I'm beginning to think that cycling on a daily or even weekly cycle might not be long enough. I'm thinking about a plan that has cycles within cycles. A really big cycle that takes about 3 months, and then shorter cycles that are much less extreme. Kinda hard to explain here, but I'll be doing a video.

      @@LanceHitchings 3 weeks is my intuition. 3 month, there's a lot of junk, lot of junk. Try stay drunk for 3 months and then recover does seem like overkill

    I’m going to have to listen to this many times. I really appreciate this presentation. I’d prefer not to be distracted by the background music sound tho. Again the content is invaluable, Thank you so much. I’m taking Sirolimus ( rapamycin). 4 mg. ( 4 pills , 1 mg ea. ). Once a week for 4 weeks, then skip 4 weeks. What do you think ? I’m 76 . ❤

    Thanks for the detailed explanation. I'm in my 50s and have gone from Vegetarian to IF to Carnivore (last 1.5 mths) + light exercise to heal my severe eczema and bad skin. Not sure if I should focus on Autophagy (fast longer) or stimulate MTor? Any advice is appreciated.

    Brilliant video by the way Yes!!! it’s a bit like breathing in and breathing out with your cells -If you think about your breathing too much it becomes too complicated - sometimes you just have to breath deep and let it happen - if you only breathe out and eliminate toxins through your lungs you’re gonna end up in strife - because you need to breath in to sustain your life - so being a bodily rhythm it seems that it would be adaptogenic ie you set up the rhythm of exercise and fasting and this cycle will optimally adapt to your rhythm - also my question with regard to rapamacin - the reason why I watched this video in the first place - is that it’s pretty obvious that rapamacin is devastating to your body’s Microbiome if taken in large enough quantities - my mind wonders whether or not a micro dose at certain points in our cycle might be beneficial ???

      It's the way of science, I think. Something new is discovered, smart people make guesses to fill in what they don't know...and then, as they learn more, some of those guesses turn out to be right, and some of them turn out to be wrong.

    Thanks for getting this info out there - a service of public health in fighting decrepitude that some people will hear. I've cycled metformin (&berberine) along with rapamycin a few different ways. I'll probably be more intermittent with the rapamycin cycle and take better measurement of my athletic performance. The biomarker I'd like to measure most is klotho. Anybody got a credible method/source?

      Balancing mTOR and AMPK is something that I've been struggling with for a while now. I've got a video on Autophagy coming up in about a month that I think will resolve this a bit.

    Hi Lance, informative video and great presentation style. Had a few points to discuss/debate
    Curious to ask though what evidence base that we need to get involved in these deep body processes at all, or what actual evidence we have that this is the reason we’re experiencing pre-mature aging…
    Is this the hubris of humans to tinker with things we don’t deeply understand and perhaps cannot even control if we wanted to?
    Easily we can debate we should be having more mTOR not less because of the prevalence of muscle loss and organ decline in our elders… unless there are valid long term studies of statistical significance you know of?
    Also as you said, mTOR is less when fasting, balance achieved perhaps just through intermittant fasting, just as our ancestors did.

      Longevity science is in its infancy and there's a lot more questions than answers at this point. We could debate the topic, but never arrive at a definitive answer. But I think we're on the right track and I definitely believe we'll eventually find an answer.
      With regard to mTOR, I believe balance is the key. I'm coming out with a video very soon on balancing mTOR and AMPK that I think might answer some of your questions, so stay tuned.

    Great video thank you! I know this is an old thread but I was curious how TRT might factor in to MTOR since it is a huge part of protein synthesis and muscle growth…along with IGF1. I’m assuming a lot of your audience may be in the HRT camp as well, like me. Any thoughts on whether TRT can over stimulate mtor? Thank you!

      Don't know anything, really, about the connection between TRT and mTOR. At 74, my T level is around 850. I used to be around 350, but I raised my T level using natural methods, not TRT. I'm not a big fan of TRT if you can raise your levels naturally, but I understand that TRT is the only solution for some men. If you look at my early videos, I did a few on raising T naturally. Don't know if this helps much. Good luck.

    bro after a heavy workout then eating good ammount of protein in noon, then intermittent fasting throughout the night, then - if i wake with sore muscles or until it gets recovered fully, will fasting cause ampk activation?

    I’m curious if a young person in their mid 20s could focus on activating MTOR pathway most of the year to grow big and strong while young. Maximize their short term potential. And then take breaks where they activate autophagy throughout the year through AMPK and Sirtuins
    For example
    5 month growth phase , 1 month autophagy reset
    5 month growth phase, 1 month autophagy reset and cycle that every single year…
    That way they build their physique like a bodybuilder and then cleanse the cells throughout the seasons.
    Any thoughts?

      I'd make the cycles more like 2 or 3 months. But a good plan, otherwise.

    I intended to explore a mix of both high protein after fasting and small amounts throughout an eating window and cycle between periods of peak protein and periods of dispersed protein in intervals. And at least once every two months of minimal protein for about a week, perhaps after a 3 day water fast. I think that this may help with targeted waste removal and activation of metabolic pathways seldom used. The idea is similar to working every muscle in your body with focus on the weakest muscle like Arnold Swartznegger used to do during his body building days but with a twist of focusing on exercising the metabolism.

    8:44 Just checking, but you mentioned that you want to activate the AMPK pathway more frequently; don't you mean mTOR? Rapamycin being an mTOR suppressant?

      No, I meant what I said. I try to activate AMPK more frequently, and to suppress mTOR. That's why I take rapamycin. With all the food we constantly eat, mTOR is almost always activated.

    Sort of a good news / bad new situation. With the resiliency of the body, I'm thinking it might be okay to work on a longer cycle. Take a couple months with mTOR as the primary focus and build muscle, then focus on AMPK for as many months or more while just maintaining as much muscle as reasonable.
    What do you think of that strategy?

      As you may know, my theory is constantly undergoing revision as I learn more. I'm working on a video about autophagy, and I'm hoping this will clear things up a bit. It will, by necessity, be also about balancing mTOR and AMPK.

    What is the half life of an amino? Doesn't the body store them from taking apart the protein that we ate? I've heard that the body is either in a state of destruction or repair, so in the night the liver is tearing down old proteins and storing amino acid chains to use in the rebuilding of proteins that our body needs. So figure out what amino you are most interested in and see if you can find out how long the body stores it. Maybe?

    I thought it interesting that people with LAM disease are prescribed mtor inhibitors. Smooth muscle tissue that grow in the lungs and lymphatics. Trying to determine the value of that. Thoughts?

      In the body, proteins are built from amino acids. The source of the aminos is irrelevant because at the molecular level, all aminos of a certain type are identical. For example, leucine is the same amino acid, regardless of whether it's sourced from animal or plant sources. That said, because of other factors, I believe that eating a diet that's composed primarily of plant-based foods is superior than one composed of animal-based foods. Hope that helps.

      @@LanceHitchings but surely the point is that the relative proportions of the amino acids within different proteins are different. So some amino acids may be entirely absent from a certain protein, and this is of course the case with plant proteins.

      @@Katedebates perhaps it depends on whether you’re fat-adapted or not. If your body is efficient at using fat for fuel perhaps it won’t resort to gluconeogenesis. This is just a guess though...
