Hi, Karen! The kit only knocks out the lacZ gene in the bacterial chromosome. It is for educational use to teach CRISPR gene editing and isn’t able to be adapted to knock out other genes.
Hi,@Pi Pony! A different guide RNA sequence would be needed to target a gene other than lacZ. The Out of the Blue CRISPR Kit only targets the bacterial lacZ gene, as the guide RNA sequence is for that of lacZ.
Hi, @Pi Pony! Yes: each guide RNA is specific for the DNA sequence of the target. Thus, if you wanted to target a different gene you would use a different guide RNA, and that guide RNA would have a specific sequence that matches the desired target gene. You can learn more about the bacterial transformation and the role of transformation solution from our family of pGLO bacterial transformation kits: visit bio-rad.com/pGLO and bio-rad.com/teachpGLO for more info. The manuals discuss the role of transformation solution in plasmid transformation which, for the Out of the Blue CRISPR Kit, is the method in which the plasmids pD and pDG are introduced into the cells.
