I just love how MIB has watched people for thousands of years and thinks he knows them so well. So evil, so corrupt, so self-serving. That's how MIB views people. He doesn't understand Desmond and I think that scares him just a little bit.
He wonders if it's already destined for him to push Desmond down the well, and if it would hurt his plans to get off the island. MIB's biggest fear is that everything is designed to always be one step ahead of him, because that's exactly how Jacob arranged everything. He pushed him down the well anyways out of anger, because if he can't trust himself to do what he wants, then he'll be frozen in fear about what to do and then definitely won't ever be able to leave the island....but that too is part of Jacob's plan of setting everything up to be one step ahead of him. Jacob simply knew MIB too well, to the point that no matter what MIB tried to do, Jacob was always one step ahead of him. MIB knew it too, but he was hoping that eventually Jacob would make a mistake. Except Jacob didn't make mistakes, because he was more powerful than MIB, able to travel through time and exist after death as long as his ashes were burning. When MIB kicked Jacob in the fire, he unwittingly did exactly what Jacob wanted him to do. That's why the fire was set to begin with. That allowed Jacob to transcend time and basically had all the time in the world to go visit people and execute his plans. When you see Jacob visiting people off the island, from Jacob's perspective, this is after his death, because physically Jacob was never actually there, he was just a manifestation. It was MIB's rash anger that was ultimately his undoing. It's what blinded him to moments where it should have been obvious Jacob set everything up. He should have known that he wanted Ben to kill him, and that he set that fire in that room specifically because he knew MIB would kick him in it out of anger and finally being free of Jacob.
Fear is probably the main weapon MiB used too, trying to use people's fear to get them to do what he wants. For the first time he has encountered someone with no fear and can't understand it.
Exactly, it makes mib seem so petty that Desmond has let go of all stake in the world because he knows about the afterlife but mib is still so emotionally invested in the world. And when he asks Desmond why he's not afraid he might think someone told him that mib cant kill him, which is why mib as always so cocky cause he knew he couldn't die

Jacob was always one step ahead, and MiB thought he finally had a victory when he killed Jacob with Ben, but thats why Jacob spoke to MiB not to show respect but to tell him he hasn't won anything, the candidates are coming, and you still can't leave. I think Jacob could've come back as a ghost even if his body didn't burn, that was just MiB being impatient and sadistic. And he never left the island when he was dead he confirmed that with Hurley. He thought he could win because he knew Jacob would follow his rules and his philosophy on not forcing anyone to do whats right, so MiB was right to think he could win but he always feared that Jacob would have an ultimate plan which is why he was genuinely afraid when Jack said he would kill him after he proved he just spoke with Jacob who gave him critical information. MiB always depended on Jacob leaving people in the dark.

"God knows how long it took them..." subtly narsassistic as obviously he knows how long it took as he was there when it was dug. His dialogue was amazing, always subtly presenting his evil or hidden intentions/views.

Well...there's more to it than that. Desmond felt the peace of knowing whatever THEY are, even Smokey can't touch there. He felt the peace that they all felt as they awakened in the pre-Afterlife. Desmond has been touched by the light to such a degree that he no longer has any fear and just goes with it. Jacob had that same peace. Jack does too. Locke actually got there as well, only he felt he failed and embraced the notion that he's supposed to die (as fed to him by Smokey through Richard & Christian).
Thing I like about this scene is that it neatly correlates to the outrage Ben had towards Jacob saying "What about you?". But instead of God saying it to Man, it's like Man saying it back to God.
Desmond isn't afraid because he knows about the alternate reality and what he has to look forward to. The type of information is what Flocke is using to mislead his followers to helping him escape.

All that foresight Desmond has, and yet it didn't register that the man (who BTW dosen't act at all like Locke used to) had him tied to a tree, and was staring him down like a piece of bad meat earlier, might not be the kinda guy you wanna stand next two a well with? Its kind of like the creepy guy inviting you into his scary looking van for some "candy" Its always a bad idea.

Desmond conversely is a man who is filled with love, faith, kinship, belonging, purpose, peace, trust etc. and has been touched, perhaps more than any other living human being, by the energy from the Light. Desmond is the counterpoint to Smokey...imbued in some sense with the Light's power, but with goodness in his heart. He visited the alternative reality where the Oceanic and Island's visitors went and perhaps even saw their journey into the Light itself.
