Yep, we are allowed to play with any character in the game, including the unlockable characters "TT" and "Drumstick." The progression for unlocking them are saved onto the cartridge itself and not onto any one game file, so you could delete all your adventure files and they will still be there. It's also not too uncommon in other games to use unlockable content in speedrunning, like in the later Mario Kart games like 8 Deluxe.
