@@cyberbid not my video, but I use it to culture a LSS into mycelium, and then I put a sample on an agar plate to test for contam. If it grows clean, I go back to the liquid culture and set the mycelium to grain spawn. You could use a piece of a fresh cap, but might have a higher risk of contamination. Good luck!!

@@PhillyGoldenTeacherquick question about the 15 psi. I live at lower elevation and my pressure cooker for all recipes is 5 psi due to elevation. Should that also apply to using in making LC?
I had oyster mushroom culture on a petri dish for longer than 2 years in the fridge abd it still worjed perfectly. So in liquid culture it should last forever basically.
Instead of mixing all the ingredients with a spoon at the end, drop the mixing bar in first and leave it on the mixing plate, spinning, while you add ingredients to make it a little easier on yourself.
I see that we put the foil over the lid to protect the filter, from water right? Does the water in the jar not create steam from the inside while pressure cooking, and wouldn't that get the filter wet either way?
Thanks a lot Sensei!! I used your technique from this video and have got amazing results. I have super healthy liquid culture!! Keep up the great work 👍
This lc recipe was the first time I ever got a lc to work. Not saying that others don’t work. But the way you explain and the order you do it, really just everything is awesome 🤙🏽🤙🏽🤙🏽🤙🏽🤙🏽
Thanks a ton for your videos. You helped me tremendously when I was starting out and I still check in to compare notes. What’s the deal with peptone? I’ve been incorporating hydrogen peroxide a bit to reduce contam, but what’s your experience with peptone?
I quickly grabbed a marker and wrote your recipe down.Thank you.Twas short and precise.I'll follow it to the letter.I dont have a pressure cooker yet so i tried with rain water and some honey and I used my favorite sugar substitute, organic Agave.Any comments on that?I believe it should work. Thanks again for your video.I liked and subscribed.PEACE.
I've recently seen a few videos where they REDUCE the nutrition in the agar mix, and claim it increases chance of rhyzomatic mycellium. The idea is reduced nutrition causes the mycellium to have to stretch out to find nutrients. Too nutritional of an agar mix might cause nothing but the fluffy pomentous (sp?) Mycellium to grow in your plate...
I got hiveg peptone which is recommended over the stuff used here but every attempt I’ve made has turned sour, cloudy and stunk bad. Leave the peptone out and it’s fine.
I seem to have done almost the same research you did and went about things very similarly. However I used dry corn sugar, the four pound bag, and made a pre-mix with the dry malt extract. I also cooked for 40 minutes because I figured a darker solution would be good for the mycelium, but it certainly is harder to see what is going on.
Has anyone tried preparing grains with peptone? I’d be wary to add it to substrate for contamination risk, but seems like it could help grains colonize faster and potentially promote more vigorous growth?
I use 5g per 5lb dry in my simmer, also found that brown rice is a banger for nutes, so when I make 1lb bags (basically a full quart) I throw in 1 tbs of dry brown rice before I pc, I will never go back
Or a marble in there or something.. Worst case you can draw some.of the LC up and squirt it out and draw it up, it'll help break up the myc...I tried a few times. 1 didn't stir or shake enough it ended up being a huge golf ball. The other jar I did with that 1.i shook enough. It looked like a sow globe!!

I'm looking into modified plastic lids for my first time making LC, but kind of confused about those red syringe filters. Are they used to suck out the LC into the syringes?
I’m doing it on my instapot and every time the glass is breaking at the bottom I think the glass was too cold even tho I used warm water to do it . I also let it cool overnight and still . This time I warmed up the jar on a pot for like an hour at 110* and used the same hot water to do my LC I’m waiting for the pc to finish now and see what happens. Any suggestions? ( I’m using the tray to the pc too)
Hello Bro. Hope you're doing well. What are your thoughts on just adding peptone to like your regular corn syrup LC. For example -i got premix LC stuff. Agar powder. Peptone. Shit ton of honey ect. Is peptone just a good gram to add to it all? Take care man🍄🙏🤘
I have a serious question..if anyone can please answer: why would it matter if there were a little bit if water left at the bottom of a grain spawn jar but at the same time you can go ahead and breed mycelium directly in water????
Would this recipe work if inoculated with tissue instead of agar? I keep getting very murky LC after about a day or two and was wondering if the peptone made it more prone to contam with tissue vs agar sample.
Can you use an instant pot instead of a pressure cooker? I’m guessing it does around 11 psi, so then assuming if so, to add apx 20% to the sterilisation time.
600 ml H2O
18 g corn syrup
1.2 g light malt extract
1 g peptone

How do you inoculate this mixture with mycelium? Can you use part of a fresh mushroom?

I wish they’d use cups, teaspoons, 1/4 teaspoon etc. instead of weight measurements. 12 ml, 18 grams…, How much is that?

Hey PGT, what are your thoughts on liquid innoculant vs liquid culture?
Do you mean spore syringes vs liquid culture?
Because liquid culture IS “liquid innoculant“
Pgt taught me how! I’ve had much success thanks to this man. Cheers bro.
Thank you for your time, should incubation process be in the dark , or doesn’t really matter?

Can you use glucose syrup instead? It’s difficult to find corn syrup in Denmark.
Same thing, I believe. From everything I know, you want a glucose source. Hence, why the dextrose in potato dextrose agar.
Philly, I picked up the olive stuffer from your Amazon links for agar punching - how do you sterilize it?
put 10cc of the thick solution as a jump-start and then put constant stirring on for 24-48 hours, it will look almost entirely colonised
