- Reading medical studies and boning up on longevity makes for some dry reading. Not so with the discovery story of rapamycin and the mTOR pathway.
Today's video is a 2-parter. The first part is the history of rapamycin and it's a tale with exotic locales and not one resurrection, but two; one from the last place you'd think. The second part of this video takes a good look at several studies of rapamycin, both past and present. And we'll examine rapamycin's role in the age-related decline in immunity and loss of muscle mass.
I'll admit it. I've never been a big fan of taking rapamycin to inhibit the mTOR pathway and possibly extend lifespans. But these studies have reset my thinking, and I may be changing my mind.

Rapamycin seems to be working for me - I've been on it for over two years now, minimal side effects (small skin rash, once a canker sore), but feel like I'm 15 years younger (in fact, my Levine phenotypic age is now 15 years lower right now - but i did not do the calculation pre rapamycin - so my generally healthy diet / active lifestyle may be the key factor). Stiffness and pains have largely gone away. Lost and kept off weight (for the first time in 20 years) - back to highschool weight and waist measurement, active as I was when I was 20 years younger.

Rapamycin is available from compounding pharmacies at a huge cost savings vs CVS or other chain pharmacies.
There seem to be bioavailability issues with the compounding pharmacy product. Generic rapamycin / sirolimus seems to be best option.
Dr Peter Atelia In a recent podcast said that some of the compounding pharmacies do not include factors that make rapamycin more biologically available. Some people are buying from Indian Mart For less than two dollars per milligram shipped, This is not Recommendation

Like you I regarded rapamycin as an immunosuppressant, as such I didn’t want to suppress my aging immune system for an intangible benefit. Thanks for bringing these papers to my attention. Maybe I will change my mind as well.

Always great information. I'm curious if you looked at the studies done with topical Rapamycin for skin aging? The two I saw showed significant improvement in tone, texture, elasticity, reduced wrinkles and increased collagen of the areas applied. Really interesting.
I made an ointment myself. Aquaphor 99gr mixed with 40 mg sirolimus. I haven't seen much progress after a month until now.
Yes, I did see the studies on rapamycin for skin aging. But I didn't want to do a 20 or 30 minute video.

I take Rapamycin in form of Sirolimus ever 6 days 5mg. I can hardly wait until the 6 days are over and I can take the next intermittent dose of 5mg. I didn't feel any side effects. I order my semi-annual ration in India so it costs me 1mg $ 1 including shipping. In addition to other medications, I also take 1.5 grams of metformin daily in the evening.

Great video Lance, really valuable information and your concerns echo my own about the potential for mTOR inhibitors to prevent building muscle. I’ve stopped taking metformin daily for the same reason and am struggling to get the information to guide the best regimen to cycle mTOR activation (to build muscle) and inhibition (for longevity benefits). Keep bringing videos on these topics and hopefully they can get out to more people.

Very good information. Thank you. I have question: How old is too old to fast for longevity?
I've heard that mice do not have as much benefit after the mouse equivalent of 65 years for people. But that's mice. I have noticed that as a middle aged woman I don't feel as well after about 3 days of fasting as I used to, but the main thing is it affects my sleeping. I have also read that the stress of fasting causes one to put out more cortisol, which would explain the insomnia. I have started taking hydrocortisone, about 10 mg (I have a prescription) and that completely takes care of it. I'm guessing it's an individual thing. The only problem I could really think of with age is a slight loss of muscle mass, like maybe a pound of muscle after a month of fasting. The loss of weight is almost all from fat and the autophagy of senescent cells. By the way, the body does replace lost tissue with brand new tissue quickly. These new replacement cells are tantamount to neonatal cells.

I don't know about metformin.
Dr. Eric Berg on his youtube videos says that Growth Harmons can grow muscles even your mtor is slowed down due to atophogy (via long fast.)
How about the side effects of it? Do they relate to the mTor inhibition?

Not sure, but I think it was first discovered in yeast. So, mammalian refers to the same complex in mammals.
if rapamycin has such a large half life then why take it more than oce a week? shouldn't it inhibit mtor as long as it is in your system?
Great question, there a study going on right now, called the PEARL trial, to determine that. They'll be studying a few different doses. I'll be doing a video on the PEARL trial in the future.

Interview with Dr. Kaufmann on Health span saying Rapamycin studies pointing to shrinking hippocampus and Sarcopenia. She wouldn't ever recommend it to her patients.
Interesting. I haven't heard about the hippocampus, but I heard that it's supposed to help prevent sarcopenia...and this is backed up in a couple of studies.

I'm not really up-to-speed on Alzheimer's.
Seems to me lowering carbohydrates to 20 gms per day or lower.

I asked one scientist, who works with rapamycin, whether he would recommend taking it for anti-aging. His reply follows: "Blocking mTORC1 by rapamycin could have both beneficial and harmful events. mTORC1 is a major cellular hub and influencing its activity in various tissues can lead to many adverse effects. In my personal opinion, I wouldn't recommend taking rapamycin."
Unfortunately, you didn't name the scientist and the harmful side effects.
The story of rapamycin was very unfortunate and its real benefits were misunderstood. If used properly, rapamycin is not much more dangerous than ordinary aspirin or metformin. Both are also anti aging drugs and harmful but the benefits predominate by far. Especially in a cocktail of Medicine including a PDE5-Inhibitor ( I take Tadalafil 5mg). I'm 71 years old and i can't wait until there is a ready alternative, it would be certainly to late. Side effects of rapamycin are known and reversible. And my favorite scientists are Dr. Green and Mikhail Blagosklonny. He said " "Not taking rapamycin may be as dangerous as smoking","The time is now unless it’s too late"
@@Harosie His name is Yossi Tam. You can google his name & rapamycin. He did not mention the harmful side effects, but I rely on his opinion. I am 77 years old and do not want to experiment with anything with potential harmful side effects. As far as I know, rapamycin is not part of the daily regimen of David Sinclair, which says something.

I respect the science of David Sinclair and I read his book. But science is not black and white. He takes NMN and even Lance Hitchings at least questions that ua-cam.com/video/HiKTaCQaP78/v-deo.html. His recommendation of resveratrol is not confirmed in known studies, and even considered harmful in some studies. ua-cam.com/video/cVXTVb-0KkQ/v-deo.html. Nevertheless, I continue to take both supplements (in addition to others such as niacin, apeginin, TMG) even if there are still few statements from studies on people, precisely because I'm 71 and can't wait years. By the way my daily expenses for my supplements are about $ 4 to 5, less than a packet of cigarettes and $ 150 a month. Including my two most expensive medicin Dasatinib und Rapamycin. I switched my VPN to Vietnam, got onto Baidu, and found a Chinese chemical manufacturer who sells rapamycin for $70 per kilogram. That will last you forever.
