Could I send in a shed to isolate a project gene I’m working?
We would be happy to! But you will need to produce like 3-4 generations in order to get numbers large enough for us to be able to identify and develop a test for a new morph. We usually need 30-40 animals that are not all from the same sire and dam. There needs to be some outbreeding. So, it does take some time, but can certainly be done after you have worked a new project for a few years!
Am I right in remembering that the mutation in DNA codes itself that is not the issue. It is when the mutation changes the codon to code for a different amino acid that the mutation changes the shape and folding of the protein. Also I believe as humans there is a fair amount of historical viral RNA and then converted to DNA in our genome..
Yes, you are correct about both! Silent mutations don’t change the protein at all, but mutations that do code for a different amino acid will change the composition of the protein, and that commonly also changes the shape and function of the protein! But, not always. There are exceptions to most processes, but what you are talking about is what generally happens!!! And yes, it is believed that we can inherit DNA from different organisms in rare cases 👍
I am late to this video - could a viable andro hatchling actually be YY? Because in most species wouldn’t YY be incompatible with life that would die before hatching? Or does the Y chromosome in snakes contain more genetic material the loss of the X isn’t as much of a problem?
The Y chromosome in ball pythons does contain a similar amount of genetic material as the X chromosome. So it does seem like it would at least have a chance to live…but we shall see!

Where does a gene like Fire or Disco fall regarding hypomelanism since they lighten the animal? How are they working in comparison? And isn't banana a form of albinism as well? Is it the only one that isn't recessive?
I believe both your points about fire/disco and banana are on the right path! We will have more info about those morphs here soon!

So incubation temps doesn’t have an impact on the sex of the clutch?
In ball pythons temperature does not affect the sex of the offspring. But in some other reptile species, like some crocodilians, turtles/tortises, and lizards, some of those do have sex that is determined by incubation temperature!

Oh yeah, sounds like you do need a Mamdarin test alright! We are working on it! Will take a little while still because we have sheds out getting sequencing done on them, that usually takes at least 2 months. But we are working on it!!!

I hit on a strange snake myself, and that is a head scratcher. First time pairing each, never breed. YB Batman het VPI Axanthic x Fire Spider VPI Axanthic het Clown female. Result 2 white snakes, one died in egg 2nd runt 12g with a wobble and assit feed first 5 meals before took of on own, and turned out to be a male. Still alive, but with a wobble.
Sounds like you made Spider Spotnose animals. I believe those are white, and usually don’t live for long. So sounds like you have done well being able to keep one alive for awhile!

This makes a lot of sense to disprove the people I have seen post negative comments that expected guaranteed heterozygous snakes they had tested turning up not het meant the error was in the testing and those running them. Especially to hear that partial partho clutches are more common than we have realized.

Great information guys. Kinda brings up a question we had here at Royal X Reptiles. Maybe you can tell us what percentage of males from a male maker female maker banana produce one of opposite sex that is expected. Also for the male created by female maker, is this a copy of father and be a female maker.
Good question! You can reference our Back to School episodes for more detail, but here is some info: I believe it is about 6% of the time that a male maker male will produce a female banana. Similarly, about 6% of the time a female maker male will make a male banana. In the second case where a female maker male produces a male banana, he will be a male maker because the banana mutation he will have will be on his Y chromosome, which will in turn pass to 96% of his male offspring from then on. I hope that helps!!!
Soooo I’m no geneticist but would the Banana/Coral Glow be an example of the phenomena? Since males are “male makers.”
Banana/Coral Glow is a fun genetics story! We did do some episodes about that, they are our Back to School Episodes with Dr Travis Wyman. Androgenesis is a different phenomenon. It is more similar to parthenogenesis. Partho is when a baby is produced just from the mothers DNA, and Androgenesis is when the baby is produced just from the fathers DNA!

Im confused. How is a 30 gene panel 130$, while only a 4 gene panel is 110$? Thats doesn't make sense. Personally the speed of test results is not nearly as important as getting as many answers as possible for as cheap as possible.
The biggest reason why there is a difference in price for a 30 gene panel and a 4 gene custom create your own bundle is speed. The reason why we can do the full panels as cheap as we do is because we wait until we have several full panel samples in to run before we do a run, and then the run we do is a long and complicated type of run, but it does make it so that we can run many samples and many tests in one run for a much cheaper price. So the turnaround for a full panel is usually 3-5 weeks. For the custom create your own 4 gene panel those are being done as separate FASSST tests, so you would have those results in 1-2 business days the vast majority of the time. So in your case where time is not a big issue and you want to get more tests at lower pricing, then the panels are a no brainer winner for you!!!
@raregeneticsinc Ok, can one choose to get a 4 gene panel done the slow way for cheaper then? If speed makes that big of a difference? Because apparently its a difference of paying 4$ a test vs about 28$ a test just to have fast results. I feel like many would choose to wait if they could save that much money.
@@acaciaballpythons Unfortunately we aren't able to do that. The big savings in cost comes from taking one DNA sample and running 30 tests on that one sample through a slower process. We can't get that kind of savings if we are only running one DNA sample for four tests. The cost to us would be the same if we run 4 tests or all 30 on that one DNA sample, so it would have to be $130 for four tests if we run it the slow way. So it is still a little cheaper to run it the FASSST way at 4 tests per sample, but once you get to 6 or more tests per sample, then it is the same or cheaper to just do a 30 test panel as long as you don't mind the wait time. Over time as technology improves hopefully we will be able to change tests so that we can do them cheaper and faster, but for now we are constrained by the technology that is available and we are doing our best to get costs down as low as we can for customers that prefer that, and come up with options that are as fast as possible for customers that hold that as a high priority. But this feedback does help us to know what to watch for in the future as more options come available for testing. We appreciate it!!!

Thank you for the comments on our oldest ball python. We hatched him in 1991 and he actually has a name "Tiger" since his pattern was so different back then. We also have another male that hatched in 1992 on our oldest son's birth day. They are both still here and hopefully will be for a few more years!

I would think it in your best interest to segregate the answers by current customers and again by pet owners that may order a few tests in their lifetime versus breeders that order dozens of tests every year.

If it's feasible, I think having the *option* to upgrade a full panel to expedited status would be the best course. As you wonderful folks have stated in this video, some people care about faster turnaround times while some don't; this option would allow for more time sensitive results to come in faster for the people who want to pay the premium.

I don’t mind paying more if I needed the panel test faster but for the most parts I don’t mind waiting. So giving the option to pay and get it fast would be great but if you can save some money and wait that’s always good too.

I prefer the tests as cheap as possible to be able to open up the accessibility to smaller breeders. It’s part of the problem with the top 30, as that directs HEAVILY to what’s effectively the most saturated parts of the market. It would be actively less helpful for people exploring other genes. I think a subtle gene panel would be awesome too, like yb complex, fire complex, OD, Etc. helpful for the “what the heck is kicking in this”

In order, #1, i definitely want a full panel to include all possible tests, because thats kind of the point of a panel is to discover possible unknown genes. Then additional options, #3, a full recessive, and a full inc.dom panel would be useful too. Then #4 , just as a custom maybe 6 morph panel, either a complex or buyers choice.
3 followed by 2. If you order both test with #3 you would be getting the equivalent of #1. Maybe run a deal where you get a slight discount if you order both recessive and dominate panel with #3?
