Having party members in general means you're not playing on Hardmode, which is required for Enki's (and anyone else's) S ending. If Nosramus doesn't wanna talk to you, that means you opened the chest in their first laboratory and pissed them off.
Due to the rules we agreed on Speedrun.com, it's 1 second faster to start through New Game. The reason for this is because when you start the timer when selecting a save file, the game has to fade out from the menu and then fade into the world. While when starting from character creation, the prompt disappears and the game immediately fades in.
