    First time I actually find a use for that "playback speed" option.
    0.75x makes it perfect.

    I'm actually thankful she speaks fast cause in loads of these types of vids they speak soooo slow and have to speed it up to 1.5x or even 2x to not fall asleep.
    Also, great explanation!

      ***** This was made for a university assignment and we had to keep it within a 3 min +/- 10 second time limit or we would lose 10%, and qPCR data analysis took longer to explain than we thought, so unfortunately we had to speed up the narration. I can post the script if you would like.

      +TARDIStennant It may be unintended, but speaking quickly made this a really good revision tool. Repeating what I had already learnt in the lab slowly made other videos excruciatingly boring. This video is great for those who just need a crash course.

      It may be two years too late but now you have the option to slow down video. For instance putting this at 0.75X speed renders it in "normal" speech speed.

    I usually get very bored listening to instruction videos when the speaker is talking at a slow or normal pace, so this was actually great for me and encouraged me to stay focused and not fast-forward! Thanks so much, it helped a lot.

      +Alexus02 I completely agree that this is too fast. It was made for a uni assignment and unfortunately we had to keep the video at 3 min +/- 15 seconds or we would loose 10% of our mark :( The original narration was about 4min so unfortunately we had to speed up the whole video to get in in the required time, so it doesn't sound as good.
      Sadly I don't have a copy of the normal speed video but I have included the video script in the description :)

    Just had a quick look at a paper about this double delta CT method and understood nothing. Watched this video for three minutes and i have to say i understand the concept of the analysis now. Why do people always write stuffs more complicated than they actually are. Thanks man for the vid!

    The work may be good and clear but oh my god is this narration annoying as hell. Good job on your project but the narration speed + the pitch of the voice = not a good combo.

    Oh my god so many people just complaining about how speed she talk... The content is AWESOME can't you just be thankfull for that?     at 3:00 don't you mean the expression is 1/3 in the dark compared to the light, instead of 3x in the dark?

      10 months too late:
      The expression is higher in the dark because we got a lower Ct, meaning we needed less cycles to reach a threshold amount.

    I disagree with her assessment concerning relative quantification. Although there is more gene expression of the target gene in dark treatments the ratio seems to be incorrect. Using the Livak method or the deltaCt method you can see that the target gene is expressed at a 32 fold change if the reference gene Ct number is ( for explanation purposes) 2 and the target gene is 10 in light treatments. The reference gene Ct number is 2 and the target gene is 5.

    PhD student in Belfast, Ireland here. I have to demonstrate to Undergrads the basics of qPCR tomorrow and thought I'd watch a video or 2 in case I missed anything.     I have a question, if the mean of my ct value is 27.6. When I plug this number into my standard curve equation to generate DNA concentration, the concentration is like 660, which doesn't line up with my standard curve plot.

    I'm pretty sure people can adjust the playback speed manually. Don't worry about speaking too fast, this video was helpful and informative.

    0 min: ct = 34, stimulated 30 min: ct = 35, stimulated 60 min: ct = 32, RT- = 33 how do I calculate ?
