That trunk line is made of fiberglass. It 100% has microbial growth in it. Either caused by duct leakage or from the air conditioning. You can cut into these the same as you would a pumpkin. Pull out the plug, take a picture looking down in each side. Then take your tape lift off the plug you just cut. The closer you get towards the air handler the more contaminated it will be. Replace your plug and mastic to seal shut
