Great recording. Love this demo track. After several trials, I found out that the max ouput level from the monitor output of my Focusrite 2i2 gen2 is actually lower than the level of my guitar (passive but hot humbucker). So the trick is to use the headphone out at max volume which actually corresponds to my guitar output volume (input track volume trimmed to stay in the 'green' zone). I went direct from headphone out (left) to tube preamp input (Synergy) and back to input2. Did not need a transformer actually. Finally I did a comparison of a direct recording vs reamped in this way and it was nearly indistinguishable. By the way Reainsert seem a cool method to reamp but I would probably need a bigger interface..

If I want to use my reamp box, the the process is the same?

yes, exactly the same. Just put the reamp box between the line out of your audio interface and the amp. However, check what the output gain of your reamp box is, a lot of passive boxes are -10dB, so your signal strength will be significantly reduced, and you may need to compensate for that by increasing the gain of the channel in your DAW (and hopefully you won't clip). It's actually a drawback of most passive reamp boxes that's yet another reason why I don't like using them.
