Actually the procedure itself wrong .
This type of injection called as implantation
The Implantation always should be done either Right side or left side flank subcutaneous region of mice, which is easy to take tumor volume.

Well done! You are an amazing vet/researcher thanks a lot for doing it ethically and humanely and not scaring or torturing the mouse

Your mice are so calm when you grabbed them and injected. In our lab, we shoud have had anesthetized the mice then wait them for sleeping, or those energetic mice wuold twist themselves and try to run away. After that, we have to wait them recovering from anesthesia. So it always takes a long time to finish the xenograft. Do you train your mice to get used to injection or what to do if your mouse are too energetic to inject ?

According to my experience, the nude mice is calmer than hairy mice.
So they will obediently until I finished the experiment without any treatment. :)
But the hairy mice may not be the same. QQ
However, subcutaneous injection is a simple experiment.
For the experienced operators who can through the good restraint method without anesthesia.
Except some very painful experiments, such as tongue injection,
must be use appropriate anesthesia to reduce their pain.

    Hi,
How long does your whole xenograft experiment take? from doing the experiment to phenotypic observation. just curious. Thanks!

    dear michael,
did you inject two different cell lines or cell line conditions in this mouse?
because i want to do something similar and wanted to ask if this were different cells for each flanks.
and maybe how well the tumor grows in such conditions.
    best regards
    pablo

Hi, Michael, How do you control the variance of tumor in these mice? Do you use 1/2", 0.5ml, 29 gauge syringes? Do you prepare 0.2ml cell solution then use the syringe to measure 100ul? Will 29 gauge shred the cells? Thank you!

    • @michaelchou0806

to investigate the effect of X gene in tumor formation
thus, we induce (overexpression) or inhibit (knock down) X gene in cancer cell
then the cell inject into mice to observe tumor growth
if it a cancer promoting gene (oncogene), the tumor of mice might be larger than control
this is my first time doing this animal experiment with not a good restraint manner.... QQ

i have nothing against experiments in rodents, because they are unfortunately crucial for clinical research. But it annoys me when people say things against it but still take pharmaceutical products which were tested using animal models

    • @jairnetto
      @jairnetto 6 років тому +14

Not just pharmaceuticals. The pigments on the screen of your monitor or cell phone were tested on rodents for toxicity. Short and long term. In science, we do our best to use them rationally and painless. A video like this help us to learn the technique without using tons of mice.

@daniel gabriel because half knowledge is fatal. Immune cells behave different to cancer cells. Cancer cells can grow only in immune compromised mice. And it's better to experiment on an animal than on a human, unless you are a moron to hate your own species

@@shaktiprasadmishra1720 Actually, not entirely true, if you try to xenograft or allograft tumor cells, then yes, you need to have immunocompromised mice, but if you try syngeneic grafting of tumor cells from one strain to an immunocompetent host of the same strain, tumor cells won't be rejected.

Human nature, nothing you can do about it that will change, we will all only cease to be a part and only a part of society, nothing more, and it's also human nature to feel empathy, sorry for the rats.
