5 most promising anti-aging compounds (updated 2022)

  • Due to last years success, I am back with an updated list of my 5 most promising anti-aging compounds.
    There are more than 200 different geroprotective compounds. Some of these are already available as supplements, some are in clinical trials and some have FDA approval. Which are my top 5 that I think i most promising in terms of their anti-aging effects (so focusing more on healthspan than lifespan). And note the use of "promising" - there is still much work to be done to better investigate the long-term safety, dose and timings of different supplements which you should always bear in mind.
    Anyway, I made this video more for a bit of fun and to make a longer summary video to pull ideas together from multiple previous videos with some new info too! So hope you like it! And disclaimer - this is not recommendation or health advise - i am not a medical doctor!!

      In the last year I become more aware of the bio-availability issue and this is why I'm pairing things. So, this is my latest top 5:
      a) krill oil + astaxanthin,
      b) NR+quercetin+resvertrol+TMG,
      c) CoQ10+shilajit,
      d) berberine + aspirin,
      e) arginine-AKG + carnitine,
      as this are available for me and I only have to take 1 or 2 pils for each.
      The second pairing is a) + b) + c) from which an element might be absent sometimes, and separately d) - each combination for one or two days per week,
      and in yet other days: e) two or three days per week (this are workout days when generally I also take other vitamins and minerals).
      As an honorable mention: hyaluronic acid and collagen at least three days per week.
      I can't wait to see the Christmas and New Year posts!

      My list of the top favorite anti-aging and related supplements wouldn't include nutrients that can be obtained from food or produced from other food-based nutrients: fat-soluble vitamins, B vitamins, choline, omega-3s, phosopholipids, astaxanthin, spermidine, N-acetylcysteine, D-ribose, BCAAs (leucine, isoleucine, and valine), glycine, creatine, carnosine, carnitine, taurine, citruline, arginine, glucosamine, alpha-ketoglutarate, lipoic acid, hyaluronic acid, magnesium, glutathione, etc.
      Still, I do emphasize them in eating a diet of whole foods that is nutrient-dense, animal-based, and nose-to-tail. There is no essential, semi-essential, or conditionally essential nutrient that isn't found in one animal food or another. Nonetheless, I do supplement of these for proven or potential improved healthspan, lifespan, physical performance, neurocognitive functioning, and such. It's just I'd keep them separate from those kinds of anti-aging supplements that can't be obtained from food at all.
      Some of my top 5 have changed, but some have remained the same. At the moment, these are: MitoQ (bioavailable CoQ10), AliveByScience Lipo NAD+ Complete (NAD+ and NMN), Cerule StemEnhance Ultra, Cerule PlasmaFlo, and Peak ATP. But I also like nitric oxide boosters, if I don't know their benefit for longevity. Specifically about anti-aging, I'm thinking about a Sirt6 activator, specifically the product from DoNotAge. And I'm interested in Timeline Nutrition Mitopure, a new supplement involved in mitochondrial health.

    I will investigate Flucoidan -which targets Sirt-6… whereas many target Sirt-1…

      I take all the fat-soluble supplements together with yogurt that has a high fat content and with my omega-3.
      This way my body can absorb it all more efficiently. Then I eat breakfast.

    Lipo Berberine is awesome. My arthritis symptoms are greatly reduced and I can walk without a limp.

      @Fepper it’s well known in studied to help people with rheumatoid arthritis. Within a few days of taking liposomal Berberine my stiffness and pain was reduced and symptoms came back when I stopped taking it for a week. More studies should be happening including it’s blood sugar lowering effects on people with type 2 diabetes.

    Thanks for this great and concise video. Metformin lowers insulin resistance. So is supposed to do berberine. Would you rank them at the same level?
    All these supplements act on epigenetic. What about supplements to make telomeres longer? Ashwaranga, astragalus...

    You stated that you didn't include vitamins because they are essential and a "given", but you kinda did include them by listing NMN, NR and Niacin as they are basically different forms of B3. In that spirit, I would include Methylfolate.

    I stumbled upon your show just a few weeks ago and becoming a fan 😊. You mentioned about Rapamycin being anti-aging drug but needs to understand the dosage and frequency for anti aging purpose which could be different from clinical condition treatment. I have read mixed comments with some recommended taking 5mg/week and some 10mg/week once a week and not daily intake.. And what about Calcium AKG?

    My top 5 in no order would be:
    1. Fisetin and quercetin
    2. NAD+ (including the precursors) with a good quality oil or liposomal for better bioavailbility
    3. Spermidine
    4. NAC
    5. Berberine instead of metformin since it's OTC

      @@ET.... I think there was a study done and they used regular fisetin which isn't bioavailable by itself. I wonder if it would have made a difference if they used liposomal fisetin or something. I felt like liposomal fisetin for me made me get into autophagy stage much faster. But maybe it's all placebo, who knows.

    Hello, I found this UA-cam chanel a year ago. I have become a fan 😊. You mentioned last year and I reviewed that last year list. It seems a few revisions for this year and I have a few questions. I currently take daily NMN 700mg, 500 mg Trans Resveratrol, Quersitine, Fisetin, and vitamin supplements. But disregarding the research (as there is so much out there). My main influences come from science people as yourself and "What do you take?" as this is the ultimate determination in considering the science?

    Video suggestion - a list of compounds/studies that have extended maximum lifespan in mice/humans. Even amongst this list, the compounds are not setting a new maximum lifespan in mice. I'm curious if that is because all the pieces of aging have to be solved in order to achieve a new maximum lifespan, or none of those pieces that would extend maximum lifespan have been discovered. Great video as always.

      What do you mean "setting a new maximum". The record for mice longevity is probably on genetically modified mice. This is about external factors.

    One thing others have not mentioned already are anthocyanidin-containing foods (blueberries, acai, hibiscus tea, etc.) - I phrase it that way since it isn't absolutely certain that it is the anthocyanidins themselves that are doing the heavy lifting in these foods.

      Quercetin and fisetin had little effect in humans but did not seem to cause any harm either. Fasting is likely the more effective option for the same benefit.

      I’m curious, why would you recommend flaxseed over fish oil? The conversion of alpha linolenic acid, found in flaxseed, to the pharmacologically active EPA and DHA are so low that it takes very large amounts of flaxseed oil to raise serum levels. Do you think flaxseed oil should be included for its other pharmacologically active constituents?

      Intermittent fasting made my health conditions worse, particularly diabetes.
      So did keto.
      But people the cult of fasting and keto don’t like hearing this.

    Thx for this interesting video. mTOR activity can be inhibited by fasting and by reducing calorie intake but also by coffee intake.

      No it didn't, and that was quite surprising. GlyNAC is tested in humans where they measured improvements in gait, grip strength, cognition and lowered inflammation. It was quite clear results and not just some DNA methylation clock which we still don't know has any real impact on aging.

      @@boklugenkopf8170 Agreed, I believe the GlyNAC study that came out earlier this year to be a huge breakthrough and pushed NAC (and maybe NACET, if the price comes down) to the #2 slot behind NR/NMN/TMG. Since glutathione drops as we age and it's critical to keeping down inflammation and cleaning out the system, this treatment's profound effect should be at the top of everyone's list who's over 50 years old.

      @@boklugenkopf8170 Also the methylation clock is just an average. So when someone has a “biological age” of 40 when they’ve been on the planet for 50 years. That’s probably how old they’re supposed to be biologically anyway, it’s just with over half the Western world following the McDonald’s diet, relatively speaking one is “biological younger” and beating the clock... But not actually younger than they should be.

    My Top 5:
    1: Whey Protein: for Leucine and mTOR activation Yes, I'm contradicting your video! I am doing weight training to increase Lean Mass and lower body fat and mTOR is essential for Muscle Protein Synthesis;
    2. Creatine monohydrate: to increase intracellular water and increase intensity during weight training. Increase ATP production;
    3. Apigenin: to increase intracellular NAD by downregulating CD38;
    4. R-Alpha Lipoic Acid: make sure to only get the R enantiomer and not the common racemic R/S or (DL) mixture. General antioxidant;
    5. Betaine anhydrous also known as Trimethylglycine (TMG): methyl donor. Help keep Homocysteine low. May help Muscle Protein Synthesis; and
    6. Sunscreen with UVA-UVB protection. To protect against premature ageing of the skin;
    7. Curcumin: to help lower general inflammation. Also, it helped me in preventing Delay On-set Muscle Soreness (DMOS) that occurs after doing eccentric movements (weight training).

      Sunscreen actually blocks vitamin D absorption. I think vitamin D should be everyone’s #1 anti-aging compound.

      Looks more like you want to extend your healthspan, not your longevity ;) I guess fasting would be a good addition. Dr. Stanfield takes a similar approach (with added metformin).

    Affordability is also a key concern when it comes to longevity substances for general public, currently I'm only taking metformin and nuts/olive oil(for oleic acid) because they're cheap and easy to get.

    Very good. Rapid pace............ and you gave the highlights. SOME of these compounds, and health benefits all come from a KETOGENIC DIET ....... and healthy eating. LOW CARBS, GREEN VEGGIES, and NO junk food.
    I'm old........ eat very strictly, with literally Zero junk food, and low carb KETO diet, and could say that I weigh the same as I did in high school, and feel fine. ** I always want to learn more, especially about BRAIN FOOD.

    Wheatgerm is sweet and works well to sweeten cereal without sugar. Add 5-10 grams (1xTablesoon) will really sweeten up your Müsli. I want to stir it into melted unsweetened chocolate to see if it makes good candy bars, probably not sweet enough and not a cheap alternative to commercial ones but I suspect it has no fructose. Needs refrigeration or will go rancid.

    5. Good social life (surround myself with positive environment, and cut off toxic people).

    I think you will find it AAKG. Pyrroloquinoline Quinone,N-acetyl cysteine plus Collagen should be added IMO.

    I more or less agree, I do take NMN, AKG, and lots (40g) of wheat germ every day. I have no access to rapalogs or rapamycin and I am unsure that metformin helps nondiabetics. Instead of those I take the NAC/Glycine combo and I am thinking about taking Acarbose though I have heard it causes flatulence. Apigenin seems to go hand and hand with NMN - I take that. Astaxanthin seems likely to be beneficial based upon ITP discussion - I am taking that. Genistein seems likely to be beneficial - I take that. I would really love to hear more about ISRIB, but have heard too little to consider taking it. I think Locamidazole has great potential for the tiny bit I've heard about it. I am holding off on "senolytics" like quercetin, fisetin, and grape seed extract, but I do think these will likely be beneficial though not necessary the best ones to take.

    As a 61 yr old personal trainer, I am interested in aging and how it can be slowed. I am not a scientist so looking for snippets that I can learn about without being too "scientific". I can understand this so have to say great video.

    Given how some previous compounds have now shown to have the potential to fuel cancer (scary) I think the safest way to slow aging is intermitent fasting, exercise (both cardio and resistance), avoid bad things like ciggaretes and excessive alcohol, good nutrition and good sleep.

      Everything causes cancer at this point.
      Do you want to know greatest cause of cancer? Inorganic food and a poor microbiome.
      In the West, where the average diet consumes only 12 species of plants and 6 types of protein for the average person, our cancer rates are approaching 50%. Not to mention the pesticides, steroids and micro-plastics that accumulate from inorganic foods and processing.
      Compared to a rural African diet? One that consists of roughly 600 species of plants and 300 different protein sources? Their cancer rates are next to 0%. Yes, zero.
      Yet the FDA bans every supplement with promise for our health put approves supplements like Lipitor that have been shown to actually cause heart failure early.
      I agree with you that diet, exercise, intermittent fasting, additionally sleep and mental stress reduction are the keys to longevity. But do be wary of reports regarding ‘cancer’. It’s very easy to feed a lab rat a certain medication and then prove it causes cancer if you feed that rat a high-fat, high refined and processed carb diet (like the West).

      All you mentioned are not enough to slow aging. Do all you mentioned then include food supplements. There's a reason they are called FOOD supplements; you CAN NOT get all the body needs even if you eat balanced diet.

      Yeah I'm with you on that!!! I feel best when I eat healthy whole food diet high in fats and protein and low in processed carbs. I feel even better when I incorporate intermittent fasting! I also do high intensity work outs for 20 minutes a day. Recently I have started doing red light/near infrared light therapy with great results!! 😁👍

    Off the top of my head, I'd be tempted to include soil based organisms to my personal list. Unlike Metformin, these products actually promote digestion and have no nutritional negatives to speak of.     I noticed that when I took metformin that I had noticeable muscle atrophy, presumably, due to the inhibition of mTor. Obviously given the multiple independent pharmacological
    mechanisms of actions of metformin, other pathways could have been at play there, but given the role of mTor in muscle protein synthesis, it seems to be likely causally related in my situation. Now, I definitely had superfluous lean mass (I train for an aesthetic, bodybuilder look) so it may have been easier for my body to lose muscle mass. But it does make me wonder if long term mTor inhibition may increase sarcopenia and increase morbidity in elderly populations.

      @@bredenis5 Most people take between 3mg to 9mg (in a single dose, all at once, once per week). But people seem to get the best results by starting low (e.g. 1mg) and then slowing going up. everyone is different.

      Longevity is deeper than just telomere lenghtening. I would focus on metabolic robustness, like mitocondrial health, ROS supresion and protein folding, which are things that you can change.

      You can't.. It is used as an immunosuppressant in transplant cases. I dont know how long you would live with a suppressed immune system. You would be vulnerable to microbes and cancer.

I feel like I did when I was 20 years younger now, and can do the same type of physical things that I could do then (in terms of weight lifting, rock climbing, etc.). It really seeems to be working. My latest blood test suggests my biological age is about 15 years younger than my chronological age.

      That is interesting. If I could get it, I would chance to try it. However, I would be quite cautious and probably would not take it on a regular daily basis until more data are in. Perhaps the immunosuppressive effects are reducing inflammation and that could also happen with anti-inflammatory steroids, which have long term dangerous side effects. Good luck and keep us posted.

      @@scottpreston5074 For anti-aging, the dosing protocol is once a week (not daily). Immunosupressive effects are caused by high daily doses over a long time. On low doses, pulsed dosing schedule you see immune system upregulation. Its easily available from dozens of online pharmacies, inexpensively. We have a list on our website that people have used and like.

Dr. Peter Attia is of a similar view. He wants himself and his clients to win the "centennarian Olympics" and not beat some academic DNA clock.

      My list:
      Things to avoid for aging itself:
      1. glucosepane (the most dangerous advanced glycation end product, made by high temperature cooking, high speed blending, and fermentation). This cannot be cleared from the body, accumulates and leads to tissue stiffening and inflammation. Hard to simulate with animals, for scientific research, as this generally requires time to accumulate.
      2. hydrogenated soybean/corn/palm oil (mostly deduction on my part, as gamma-Tocopherol is associated with high methylation aging. But that makes no sense until you consider the top sources of gamma-Tocopherol in the typical American diet are these oils.)
      3. Trans Fat
      4. synthetic vitamin E
      5. high dose testosterone or other high dose hormones.
      Stuff that is just plain toxic, ubiquitous and can easily cut your life short:
      1. Lead (far more deadly than most people realize. A study in 2018 showed that low level lead is responsible for 18% of all deaths in the US...mostly heart disease related.
      2. Tobacco smoke
      3. Alcohol
      4. Polybrominated diphenyl ethers (PBDEs)
      5. Organophosphates
      Obscure stuff:
      1. Glycyrrhizin, natural chemical in black licorice
      2. Coumarin a chemical in normal Cassia Cinnamon. Better is the less strong Ceylon Cinnamon. The dangerous one is the one in virtually any product in the grocery.
      3. Pine nuts. These have high levels of advanced glycation end products naturally, so leave those to the birds.
      4. Kava Kava (can damage the liver)

      @@rhyothemisprinceps1617 It can starve neurons of potassium. It is especially dangerous in pregnancy, where researchers found lowered IQ in the children of mothers who ate black licorice. I tend to think that the things that cause brain issues during pregnancy also cause similar issues in adults, just at a reduced rate of damage, but over time... You see this with tobacco, alcohol, hypoxia (in adults due to sleep apnea, in babies during labor), and lead. See no reason adults would not suffer brain damage from anything that damages fetal brains. It can also raise blood pressure, but lots of things can do that, I suppose. But, most of those other things people have been warned about. I suspect it could cause a stroke in someone who already has very high blood pressure, or cause major renal damage.

    Glynac certainly deserves a spot on this list. Just like nad+, the body needs healthy levels of glutathione and studies so far conducted show an increase in longevity in mice, improvement of various blood markers in humans as well as an effect on abilities such as walking speed in aged individuals. Antioxidants got a lot of bad press the last years, but the main conclusion should not be antioxidants are bad. Instead the conclusion should be that antioxidants need to be in balance, which is clearly not the case as aging continues. Glynac seems to give the cells control over how mach glutathione they want to produce with it, restoring balance.

    Is it not possible to get more AKG by revving up the krebs cycle / demanding more of our mitochondria (e.g. Maximize aerobic activity?) When considering geroprotective agents, I usually try to go with as natural way to obtain the protection as possible. If we can exercise more vs. taking AKG, would that seem to be the better option?

      @@amperage8032 I would love to hear Eleanor's opinion, but my intuition would be to maximize mitochondrial output, meaning the upper limit of aerobic output before lactate starts to accumulate (I.e. Anaerobic output) so to exercise as vigorously as possible before the first lactate threshold. Peter Attia calls this zone two training, but that seems slightly arbitrarily named to me.

      @@rapamycinnews Atherosclerosis kills more humans than anything else (and even if someone doesn't die from atherosclerosis, they'll die with atherosclerosis). Culprit is definitively LDL particle count, exasperated in some people by high Lp(a), and functions as an "area under the curve" problem. PCKSK9 inhibitors (like evolocumab) reduce both LDL particle count *and* Lp(a) in a way that's more effective/targeted than statins (and don't carry same side effect profile). There are also companies looking into base editing in humans to inhibit PCSK9 permanently, but aren't yet FDA approved (e.g. Verve Therapeutics). Stands to reason that if you can significantly delay atherosclerosis (or eliminate it), human lifespan would be extended as well. I also think things like cyclodextrins are really interesting (e.g. Underdog Pharma), but we're still in the early days.

    I would have thought the precursor to the mother of all antioxidants, Glutathione would be in a top 5. I haven't looked into Rapamycin too well, just saw a couple videos by Dr. Stanfield on it and another channel but what I don't understand and if someone can explain, if Rapamycin is prescribed to immuno-supress someone, then isn't that making their immune system suppressed and therefore weak?

      Rapamycin is an immunosuppressant. It is prescribed for those who have received an organ transplant so as to reduce the likelihood of rejection of the transplanted organ. It moderates the body’s immune response and thus is precarious for anyone who is already immuno-compromised.

    Is there a possibility of biological dependence forming to any of these compounds - eg if you constantly take NAD+ precursors such as NR/NMN, would enzymes involved in steps further down the metabolic pathway be downregulated? If this happened the natural biosynthesis pathway for NAD from tryptophan would become incredibly slow and you would essentially be dependent on the precursor molecules to feel normal ie without them baseline NAD biosynthesis would be even slower than if you had never started taking them in the first place.

    My current top 5 are Resveratrol, NMN, TMG, Quercetin and Fisetin. I take all of these except Fisetin so far. I would be interested in learning your Top-5 WITHOUT prescription drugs (Metformin and Rapamycin).
    I do have a question though: Rapamycin is - as you also mention in the video - used as an immune supressant and supressing my immune system certainly isn't something I want to do, since I doubt that that could be beneficial for a longer healthspan. What is your understanding of this?

      Hi @David The effect of rapamycin depends on the dose. According to some studies, a lower effect does not seem to suppress the immune system. Unfortunately, there are currently no studies that determine an ideal dose for anti aging. The natural alternative to metformin is berberine. Berberine appears to have a similar effect, but has not been as well researched as metformin.The primary effect of rapamycin is to inhibit mTOR. The same effect can be achieved through fasting or a fasting mimicking diet.

      I got Rapamycin from India and take 5 mg once a week. I follow that 8 weeks, then take 4 weeks off, then 8 weeks on again. Taking only once a week allows my immune system to benefit from a quick suppression then bounce back. Like Hormesis. Next week I start on my "bye" period. That is the protocol I learned from Peter Attia MD; if you are interested in longevity, you should be following him. Are you fasting regularly? Exercising? You can combine fisiten and quercetin for senescent cells, but Rapamycin does some of that also.
