    Not a student, just trying to educate myself on antipsychotics after being prescribed for a few years, I had psychosis. Currently weaning off of olanzapine.

      I'm on risperidone after psychosis in 2018 and 2019. I lowered my dose from 4 to 1 mg over the past 1.5 years. How's it going with you so far weaning off the olanzapine?

      Awesome that you are weaning off, I currently have minor form of PTSD, self diagnosed, and have been prescribed olanzapine, methex od etc.

    my life was for a long time ruined by antipsychotics. was prescribed them and although I suffer from major depressive disorder, I do not have schizophrenia, bipolar, or any sort of mood disorder. Because I already lack serotonin and dopamime due to my depression, this medication sedated me/turned me into a zombie, made me gain 15 lbs in a matter of 2 weeks, stripped me of my personality, confidence and creativity, & even after I went off of them, I could not get back to the girl I was before. I no longer was responding to any antidepressants that had worked in earlier years or stimulant medication that had helped in the past. I looked awful and became a different person. All I wanted to do was sleep. I was so severely depressed i could not function at work (yes, I was trying every class of antidepressants at this point). I was no longer myself and had become completely overcome by my mental illness. It got so bad that my therapist insisted I seek more comprehensive medical treatment. I admitted myself to the hospital and after many more trials of medications, I had to resort to electroconvulsive therapy. I am 23 years old. Although it has a terrible rep, this treatment saved my life. the reason I am writing this here is because I think antipsychotic medications are necessary for people who naturally go into manic states, suffer from schizophrenia or psychosis. however, I am writing this as an informative warning that these medications WILL change you, and WILL sedate you. for some this is necessary. however, if you are prescribed these because antidepressants alone are failing, I really hope you will do the research on how they work because I believe that for a depressed person, these meds can be extremely detrimental . I hope that those who need these meds find them helpful. However, for me the damage they did could only be repaired by literally shocking my brain out of it. Good luck to all and please know YOU ARE NOT ALONE.

      This "meds" are cell poisons (study from 2023) and nerve poisons (many studies). The people you call ill are not ill they are a different species (study in human genetics 2024). By talking people into taking this drugs you make yourself guilty of a crime against humanity called "extermination".

    thanks for this but just to note a MAJOR correction: psychosis is associated with increased dopamine presynaptic function in the nigrostriatal pathway (particularly the part that projects to the associative subdivision of the striatum) it is NOT the mesolimbic system

    Quetiapine saved me from major depression. Came off them for afew days and cried all day, very low. Back on them and well again.

    I was misdiagnosed with schizophrenia. Been drug free for a long time and I am still suffering the disabilities caused by these medications. I cant express emotion and have slurred speach. The eay I walk changed. My hormones are all over the place. I exercise and hope for a better future. I feel like my a bird shose wings been clipped.

    Most people will never experience extrapyramidal symptoms (EPS) so they have no idea how horrible they can be. There are many factors contributing to EPS but mainly a dysfunctional dopaminergic and acetylcholinergic neurological system play a role.
    There are so many hyperkinetic movement disorders like dystonia, dyskinesia, akinesia, akathisia, athetosis, ataxia, chorea, myoclonus, ballismus, tics, tremors, Tourette’s, spasms, RLS,etc.. it can make it very hard to distinguish.
    I sympathize for schizophrenics and people who need certain antipsychotics and other medications that exacerbate these movement disorders. Most these medications drastically deplete Dopamine which is one of the main reasons for these extrapyramidal side effects.
    Science has had more than enough time AND MZoNEY to develop a novel medication that should not cause this. Many schizophrenics need glutamate and dopamine regulation (not always “reduction”). There are several dopaminergic receptors and binding sites that can be antagonized and agonized therefore there is ZERO reason why there isn’t a proper neuroleptic to control psychiatric side effects while not exacerbating extrapyramidal side effects OR anhedonia!
    Dopamine agonists can do wonders for EPS and science was able to develop non-ergot agonists which are much much safer yet can’t develop an agonist that will not cause debilitating Dopamine withdrawl Syndrome? Come on that is ridiculousness there is no excuse for no development of one. Or how about an MAOI with no withdrawal? Nope, doesn’t exist 🤦‍♂️
    9-Methyl-β-carboline should be further examined as it appears to have incredible dopaminergic restoration properties. A study was done on rats that proved it. In layman’s terms scientists injected rats with synthetic neurotoxic compounds like 2,4,5-trihydroxyphenethylamine (6-OHDA) and 1-methyl-4-phenyl-pyridinium ion (MPP+) that selectively destroy tyrosine hydroxylase and dopaminergic neurons. So they essentially gave these rats Parkinson’s like symptoms.
    After a few weeks of 9-Methyl-β-carboline administration around 80% of the damaged neurons returned to normal determined by measurements of nicotinamide adenine dinucleotide dehydrogenase activity.
    Also the adamantane derivative called Bromantane and Pentadecapeptide BPC-157 play important roles.
    Yet no ongoing research is being done. 🤦‍♂️

    Remember 'Antipsychotics' can be 'hyperbolic' - which means that withdrawal needs to be approached more carefully at reduced doses.
    And remember that abrupt 'antipsychotic' drug withdrawal can result in 'Withdrawal Psychosis'.

    I think the APs causing onset or worsening of negative symptoms is called “Neuroleptic-induced deficit syndrome”. I feel like this is something that needs to be talked more about!

    Antipsychotics cause erectile dysfunction and loss of libido, emotions and motivation. Risperidal for me caused decreased quantity of sperm and no orgasm, premature ejaculation and erectile dysfunction. Now, after 4 years, Im on path to healing myself but strugle is so big.

    just to note that in 2010 it was found that there is no increase in the levels of dopamine in the nucleus acumbes, but rather in the nigrostriatal region that projects ventrally in both the striatum and the cutaneous nucleus, and this region is related to the positive symptoms of schizophrenia.

    Good video. Except, it is stated and implied from the neuronal diagram of serotonergic neurons synapsing with dopaminergic neurons that 5-HT binds to 5-HT2A receptors on dopaminergic neurons to decrease dopamine release. And that antagonism of these 5-HT2A receptors results in an increase in dopamine release.
    The problem with this theory is that 5-HT2A receptors are coupled to Gq/11 proteins, that when activated, stimulates phospholipase C activity, thus enhancing intracellular Ca2+ levels. This is a trigger for vesicular release of neurotransmitter.
    My question for you is, how do you explain activation of a Gq/11 coupled 5-HT2A receptors produces a decrease in dopamine release?

      5HT2A antagonism increases dopamine rather than decreasing dopamine. It is believed that the antagonism mainly affects in the nigristriatal pathway without affecting the mesolimbic pathway. Thus.. the EPS side effects are relieved without exacerbating psychosis.. hope this clears your doubt.

    After they all sat and said we are putting the quetiapine up again and with that I
    have to give Lorazepam to .All this time myself and carers see what all these medications are doing .I lost I wanted to hurt myself because I feel like I am talking to a brick wall but they still refuse to listen now they are talking about having an adult protection meeting .This has got so bad with these people they do not care obviously its not their child whether grown or not when you think how people with health issues are treated and drugged it is so sad

    I think a great direction medical science could take would be to try to see if lifestyle modification could have the intended effects on these biological mechanisms, rather than trying to precisely target them with drugs. While these drugs work they come with many undesirable side effects, which is natural since they are so precise. It would be better to increase or decrease the activity of dopamine pathways as a result of activities that reform the whole brain-body system instead of just target isolated areas of the brain or body. I'm not sure if this is possible with something so damaging as psychosis but if it is then it would far more desirable.

    What I find interesting about the newer class of (so called atypical) neuroleptics is their loose binding to D-receptors in contrast to the old typical ones. In exchange they have a very high affinity to 5HT (Serotonin) and guess where the body has the most of these receptors which are then antagonized? In the gut! So basically you loose your gut feeling...

      What u just described can it be said about Olanzapine? Also what do u mean u lose gut feeling, like, that you cant feel anxiety in belly or? Also i heard that serotonin in gut its like a myth, its in brain.

      @@RudisBoss no one claims seretonin is in the gut. the claim is that an unhealthy gut is linked to these mental disorders and seretonin and dopemeine have little to do with anything. its been disproven a long time ago that chemical imbalance is a myth. its nothing more then a lie to convince people they have an incurable disease and the only treatment is these drugs, they help very few people and cause worse side effects then the disorder. its mass genocide

      (tmi but it’s medical talk so i’m sharing) i was put on olanzapine during an involuntary (and unnecessary) commitment in december, i was only on it for about a month total. i swear i SWEAR my bowel movements smell different sometimes. it’s very specific and didn’t happen until i was on the meds, it has continued despite being off of them for a few months. idk if my body is releasing certain molecules from the meds of if they changed my gut chemistry and now that’s just how that goes. either way i believe it to be a result of the medications

    Smoking marijuana as a teenager caused me to have this problem. I was desperate for friends and they did it around me and I didn’t know any better. Deeply regret hanging around them. Would have been better off hanging out by myself all the time. Which is what I do now.
    Don’t. Smoke. At such a young age. And warn others!
    It will create a lifetime of issues and struggles that can be avoided by NOT smoking marijuana at a young age when your brain is still developing.

    I am on a really low dose of aripiprazole and have been on high dose of risperidone from psychosis they both make me feel shit(currently on aripiprazole), lack of confidence I can get happy but is not a true happiness, also makes me lazy is this common?

      You might want to add in your assignment how bad these drugs are to the brain long term. I suffer every day because of these awful drugs and these doctors that I put my trust in as a kid have totally fucked it up!!!!

    Can someone answer me.
    If D2 antagonist are used, won't it increase dopamine levels b/c D2 receptors are auto-receptors, and their inhibition will lead to increased levels of dopamine????
