Sure, because when the piece of bamboo will dry, your tuning (holes you made) will be modified, and your bansuri will be "flat". ;-)

Hey! So I tired to make one exactly how you demonstrated on the video, however I can't figure out how to get it to sound like how it does on you video. I made one and all it sound like is someone blowing air through a hollow tube. Is there a trick that I'm missing? Or am I using it wrong?

Green bamboo will not give a good sound. The bamboo has to be completely dry with all the moisture taken out.
