Allele-specific polymerase chain reaction (AS-PCR), also known as amplification refractory mutation system (ARMS) or PCR amplification of specific alleles (PASA) is a PCR-based method which can be employed to detect the specific variations or mutations in DNA. Since the PCR amplification is targeting Single nucleotide polymorphism or mutations in the alleles, this PCR technique is called as allele-specific PCR.
In the case of allele-specific PCR, the primers are designed to specifically recognize and amplify a particular allele or variation of a gene. For example, if a gene has two known alleles, one normal and one mutated, two sets of primers can be designed, one for each allele. The PCR reaction is then performed, and the specific amplification of one allele over the other can indicate the presence of the corresponding mutation.
The concept of AS-PCR was initiated by Newton et al.
