How does an ELISpot work?

Explain what problem the ELISpot solves.
Explain how an ELISpot assay works.

Adhesive plastic plate.
Coat the plate in a capture antibody for the cytokine of interest.
Block the plate with bovine albumin.
Incubate cells with stimuli onto the capture antibodies in the plate.

Wash step to remove all the cells.
Detection antibody with HRP conjugation
Substrate peroxidation through HRP
Explain why the product of the reaction is insoluble.
