- Topoisomerase 1 and 2 - This lecture explains about the topoisomerase 1 and 2 mechanism of action. Topoisomerase 1 enzyme cleaves single strand of the DNA and can change the DNA linking number while the topoisomerase 2 can relax the DNA by creating negative supercoiling by cleaving both the DNA strands at the same time. This video lecture explains about the topoisomerase 1 mechanism and the topoisomerase 2 mechanism to relax the DNA by changing the DNA super coiling.
Type II topoisomerase cuts both strands of one DNA double helix, passes another unbroken DNA strand through it, and then reanneals the cut strand.
Both type 1 and type 2 topoisomerase are of 2 types.

Type I topoisomerase has 2 subfamilies typeIA and typeIB
Type II topo has 2 subfamilies typeIIA and type IIB
I don't think that is correct... in the second part of the video the primer in the loop…when you imagine you straighten the loop out... than the primers in both chains will be in the same direction….or…leading strand is 5‘ (on the left) - 3´(on the right) so lagging strand is 3‘ (on the left) than on the end of the loop it should be 5‘ ....but primer on it starts also with 5‘…Still thanks for your videos, your teaching helps me a lot!

What is the difference between DNA gyrase and topoisomerase IV? In bacteria
DNA gyrase is Topoisomerase II acting as a swivel to induce negative supercoiling from positive. And topoisomerase IV is responsible for termination of DNA replication
