The jokes are nearly all sexual innuendo, apart from 2:10, where Elton and the audience are just laughing at Matt Lucas's OTT Welsh accent, especially how he pronounces "Billy Elliot".

It seems to me that Americans don't get innuendo's (that's why they invented the dreadful "that's what she said" to clarify), while the Brits thrive in them. My favourite one is still Stephen Fry in his documentary about his trip in the US "where I helped my uncle Jack off a horse".

Alan Partridge: Well, what are "watersports"?
    Producer: You don’t know?
    Alan: No I don’t.
Producer: You mean you don’t know what watersports are?
Alan: I believe we’ve established that. Tell me what you mean by "watersports".
Producer: Well, it’s when people relieve themselves on each other.
Lynn: Perhaps those people were caught short?
Alan: They do it ON PURPOSE, Lynn!

You're missing the innuendo. Splashing out... cxming all over someone; water sports aka golden showers aka pxss play; bashing one out... a quick wxnk; "You've done a lot of..." "... and most recently 'Billy Elliott'," which is a Westend/Broadway musical based on the Oscar-nominated picture about a preteen boy from a blue collar family that wants to be a dancer (ballet if I remember correctly)
... so it sounds like he's stating Elton has "DONE" someone by the name of Billy Elliott; a lump in your throat... could be one of two things: a man's appendage during certain a act OR the load that is released from said appendage that one might ingest afterwards as a reward.

He did this for comic relief, massive respect to him for doing it. Comic Relief is an operating British charity, founded in 1985 by the comedy scriptwriter Richard Curtis and comedian Lenny Henry in response to the famine in Ethiopia. The concept of Comic Relief was to get British comedians to make the public laugh, while raising money to help people around the world and in the United Kingdom. So many famous people take part is sketches from British show created specifically for raising money for charity.

You commented on it in the video. You'll find for some reason that US celebrities often find that they are "above" certain things, like participating in a video game or children's cartoon after doing big movies might be some kind of "career damage". This same mindset doesn't exist as strongly in the UK. You'll see people of all kinds of fame in all kinds of projects.

Some of the jokes are gay sexual hints (you get a "lump" in your throat, you splash out) . Some of it is funny becase the guy in the blue costume, "Daffyd Thomas, the only gay in the village", a Welsh homophob gay guy, from an imaginary tiny village in Wales, is a recurring character, and things like "I am the only gay in the village", or " this question is from the old lady who works in the post office on Tuesdays, you know her, don't you" are recognized from Little Britain. When he says "Billy Elliot", the audience laugh because at the rehearsal, Matt Lucas laughed at his own exaggerated Welsh pronunciation several times, so they had to have another go. His rubber outfit, the newspaper "Llandewi Breffi Gazette" are all familiar from the show.

COMIC RELIEF is for charity. Elton John is good friends with David Walliams - they often summer together with their families. Walliams asked if Elton would like to participate in a charity sketch, and the answer (of course) was YES. Everything in the sketch is sexual innuendo about Elton's homosexuality, but because it's all for family related charityt, they must do the jokes in a pretty non-sexual way.

I like it when big celebs do this, shows their human side away from the glitz and glamour, I mean, Elton John has simply nothing to prove to anyone, he's not there doing this sketch to promote anything which in the most part is where you see celebs, on chat shows advertising a movie, album, book etc etc... There is one, and one good reason why he did this sketch, for the shits and giggles... And that's great if you ask me.
