I love how you took a standard feature in After Effects (The Stroke tool) and taught us how to use it through an awesome creative looking tutorial, not just here, make a box using the stroke tool or here the stroke tool can make lines. This tut makes learning this feature interesting. I've been using After Effects for approx. 20 years and there are still many features I haven't mastered. Cheers.

Yes! In Illustrator work in the same resolution of your video. Import the AI file into AE. Drag into timeline and go to LAYER Menu -> CREATE -> Create Shapes from Vector Layer. It should keep the different color strokes and width.
I love Saber plugin and I use it a lot. You can definitely use it for this technique as well. There's no right/wrong way, it all depends on the style/look or what you're trying to achieve.

When you double click on any footage, it will open up in the layer panel. You don't want to be in the layer panel to create the shape layer. You want to stay in the composition panel (main window/work area where you see what you're creating/animating). If you're in the composition panel, follow these steps. (1) deselect all layers by going to Edit -> Deselect All. You can also use the shortcut Ctrl+Shift+A. (2) Click on the Pen Tool. (3) Draw/Trace in the composition panel. At this point After Effects will create a new layer and it should be a Shape Layer. The layer name should start with the words "Shape Layer". Hope this helps! Let me know how it goes.

I think you can improve your speed if you use Adobe Illustrator, not only can it give you smoother and cleaner lines but it's easier to navigate around for painting. Then, import to After effects and convert to shapes.

Hi what software you are using for animation
I'm using After Effects. For frame by frame, I've used FlipaClip (it's great) and Scribbl (quick to make those neon light scribble).
