  • Clozapine is one of the atypical antipsychotic medication indicated for resistant schizophrenia. It is a dibenzodiazepine derivative with few advantages but still it can produce agranulocytosis as an important side effect.

    5-HT2A receptors are excitatory and not inhibitory. The fact that 5-HT2A stimulation can lead to less dopamine secretion is because those 5-HT2A are connected to Glutamate neurons and GABA interneurons and finally to dopaminergic neurons. When 5-HT2A is stimulated, the next glutamate neuron will secrete glutamate and thereby activating GABA interneurons. GABA is inhibitory and when activated, it inhibits dopaminergic neurons and leading to decreased dopamine. So antagonism of 5-HT2A leads to more dopamine.
