      What if you wanted to extract green tea caffeine for personal ingestion. Would the DCM become a problem even if it's been fully evaporated?

      Antony Liu; Well, yes and no.
      Pharmaceutical grade DCM wouldn't be an issue if COMPLETELY evaporated (this is MUCH harder to do than you might think. Even with a good vacuum, heating it for over a week and powderising the crystals every couple of days, the caffeine would still contain quite a bit of DCM that was incorporated into the crystal structue.).
      However, even a very pure AR grade of DCM, that you also fractionally distilled, MIGHT still contain some very toxic compounds (like carbon tetrachloride, mercury/lead salts, etc.), which would be VERY difficult to remove down to the required PPB level.

      Johnathan Griffin Ld50 of pure caffeine is like 8 - 10 grams, so no dying is quite unlikely

      Johnathan Griffin You never had pure caffeine? I have about 50 grams of it at home. It's not that bad but if you take too much (0,5 g at once is already too much), your heart rate goes up, you sweat and you'll get some really ugly headache while being nervous as hell. You need a 0,01g scale, measure out about 150 - 160 mg (equals about two coffees) and put it in whatever drink you want (it's bitter as fuck with a really unforgettable ugly taste), then you're good to go and start your day.

      .5 grams isn't that much for me, though it all depends on your tolerance. I60mg is about as much as your average 16oz energy drink, or 16oz of coffee. 200mg is a moderate dose for me, though I've done 1.1g with yohimbe HCl (a powerful natural stimulant) and that was fucking aweful. Though I know of someone who would do 3-5g per night while working overnights at walmart. Granted she started getting health problems.
      Though I agree completely on the taste that shit is TERRIBLE

    I would like to share the method which was done out of curiosity " how is caffeine in colour"I took 5 grams of tea sample and to it I added 500 ml water and kept for boiling.on boiling I added 10 grams of heavy Magnesium Oxide to it and kept for boiling again for 2 hours. Adding water in same not exceeding the 500 ml volume just for the purpose not let the conical dry.then cooled the whole solution and filtered through 201 filter paper.Filtrate was 500 ml.I then took 200 ml filtrate and added 20 ml Sulphuric acid (2 ml sulphuric acid : 18 ml water).Took the whole solution in separating funnel and extracted 6 times with chloroform.combined all these layers of chloroform and kept on water-bath till whole chloroform evaporates.And here I get white coloured caffeine in good amount.Thank you!!

    Have you tried extracting caffeine from soda pop with the same method? Basically just do a liquid-liquid extraction with a high-caffeine drink like Mountain Dew.
    I really like your videos. Keep up the good work.

    I have done this experiment using Chloroform as it was easier for me to obtain. It worked great and my final product was white and very pure. I don't know which would remove more caffeine, Chloroform or Dichloromethane, but I am sure the difference is negligible.

    2:17 so the caffeine is in the DCM layer? DCM/Methylene Chloride isnt listed as a solvent on the caffeine Pubchem page. Hmm

    So I followed your procedures to extract the caffeine from tea. I ran my caffeine through a 60MHz 1H NMR and I am seeing the normal caffeine peaks. However, I also see a peak around 1.75 ppm. Do you have any idea what this contamination may be?

    Helio my friend, your videos are amazing. You are a very gifted chemist. I have a suggestion for a video for you and I don't know how to contact you so please excuse me here. There is a huge market for e-cigarette devices right now and regulations are now implemented in the US which classify all cigarette products is a tobacco product. my suggestion is that because e-cigarette juices and components will become very difficult to buy, maybe you could help with one of the biggest problems. The nicotine used in the E-Cigarette liquid is extracted from tobacco plants and comes in various strengths, the most popular being 100 milligrams per milliliter which is then further diluted with PG and vegetable glycerine. Would you be willing to do a video about how somebody could use tobacco leaves to extract the nicotine and have it dissolved in propylene glycol? Thanks again from a fellow Canadian.

    If you had something dissolved in wax (carnuba wax specifically) that you needed to extract and convert to a water soluble salt, could you dissolve it from the wax using dcm/any nonpolar solvent then shake and vent like you did with an hcl solution? Brakleen didnt dissolve the wax but i am pretty sure it dissolved the something. It worked okay but i would like a professional's input for a more sound process.

      You could evaporate it at 80°c (accelerates evaporation without exactly boiling) for a while, then once it reaches about a fifth of the volume it once had disable the heat , transfer it to a test tube or something that is small but tall and let it sit at an almost freezing temperature (the closest to freeze the best, but don't let it freeze though, use your fridge or put it in a batch of ice, but the ice is probably gonna melt off and the tube has to not move).
      You can then gently remove all the sludge above and the liquid below is diluted caffeine and caffeine crystals. You could put it at boiling temperature, shake it around and let it sit once more.
      Otherwise, you can replace DMC by boiling water, ethanol but NOT methanol (1mg/ml and toxic) but DMC is recommended because of caffeine's solubility being of (140 mg/ml) AND is non polar (easy to separate from the sludge).
      Water dissolves it in (22mg/ml).
      the hotter the water, the greater the solubility of the caffeine.
      However, ethanol is a much better solvent for caffeine, and should be able to dissolve much more at room temperature than water can. Provided that your ethanol is reasonably pure (as in no methanol, no butyl alcohol, ... basically, medicinal alcohol).
      *If you have the separatory funnel* , measure your volume of liquid and solid, and weight it.
      Now, add to a recipient of the exact same volume enough water until it's of that specific volume. Measure the weight difference between the teasludge and your water:
      Volume of tea sludge - Volume of water = 0L (since they're equal)
      Weight of tea sludge - Weight of water = Caffeine (& junk) weight
      Add VERY GENTLY TO THE BOTTOM OF YOUR FUNNEL WHERE THE CAFFEINE SHOULD SIT 0.045.. ml of ethanol for every mg of difference.
      It's slightly less than caffeine's STP solubility in water, which should do the trick with ethanol. Then wait a minute so it dissolves (do not stir, it'll mix the water and ethanol together since they're miscible with eachother); then let the clearish bottom seep out from your funnel. You've got your raw, slightly contaminated caffeine mix. Please let the alcohol evaporate in less than three days as caffeine ends up unstable in alcohol after three days . You can use your radiator to do that job, as it's temperature averages 40-50°c ( which makes alcohol more volatile). In case you have no heating system in your lab/home, just lay it on a/multiple watchglass-es/flat,non-sticky glassy surfaces.
      *Otherwise,*
      you can simply put your solution in it , let it sit for a while (about an hour or so) at an ice cold temperature to minimize the solubility of caffeine, then add gently to the bottom using an eyedropper/pipette/straw some ethanol.
      Here is a clean protocol using ethanol WITHOUT THE FUNNEL:
      1. 1:40 Follow up to here, and if possible, up to 2:00 .
      2. Let your sludge of caffeine and tea extract evaporate at 80°c in the beaker for a while, until your volume (for example, 200 ml) diminushes by four fifths (so that it's 40ml). Then let it cool to STP (room temp. and pressure)
      3. Measure the volume of your oversaturated water-caffeine-tea-extract and it's weight. Weight will be refered to as "W" in my calculation and volume as "V" . I'll refer to it as "Liquid 1".
      4. Prepare the same volume of water at STP that i'll refer to as "Liquid 1". (room temp. and press.)
      5. Weight liquid 2. The difference between weight of Liquid 1 and Liquid 2 is your yield of caffeine (and other invasive particles.).
      Vliquid1-Vliquid2=0 (equality since same volume)
      Wliquid1-Wliquid2=Wcaffeine&etc
      To dilute your caffeine in ethanol, you'll pick it's solubility for caffeine which I wasn't able to find online , but knowing it's higher than Water's, we can deduce that it's above 22mg/ml and therefor, for every 22 miligrams of caffeine (&etc) you'll add add one mililitre of ethanol to your cold water solution to it's bottom (using a straw or a eyedropper/pipette) very gently. Then let it sit for one minute, and then, use said straw/eyedropper/pipette to pull the water out gently.
      You now have a ethanol/caffeine solution, hooray.
      Now boil off the ethanol at ~78°c until there's only the bottom left.
      What's left is a quite contaminated but pure enough caffeine end product.
      If you plan to consume it somehow which I don't recommend, you can wash it with REALLY COLD water since leftover alcohol and caffeine altogether won't do any good.

    One question, Why did you make such a strong effort to conserve the water from boiling out. Correct me if im wrong, but caffeine does not evaporate so it should not be affected by the boiling process

    When you repeated the process a second time at 1:30 , how much sodium carbonate did you add once again? 6 g? even if you only added 60 mL of water?       @@8915s understandable, they're both very good solvents but also quite toxic or anesthetic. If you doesn't have a rotavapor, it's difficult to carry out the evaporation of the chloroform (assuming you don't wanna fall asleep ahahaha)
      Btw, some procedures used another (non tossic) solvent. Let me check.

    Are polyphenols the same as tannins? If not can someone provide an example of a tannin in black tea that is not a polyphenol? Also, what about pigments, do they stay behind in the aq layer? What about theobromine and l theanine and the other stuff in the plant? Like, what will happen to the l-theanine when dcm is added

    I did this experiment in organic chemistry and the caffeine I extracted had a yellowish tinge to it and had the smell of Mountain Dew. Was my caffeine pure?

      No pure caffeine is white. Some impurities it seems. From my knowledge it must be white that is.

    Great video, I am basing a research experiment for general chemistry off of it and the paper included. What is the molarity of the dichloromethane and sodium carbonate you are using? The paper does not specify either. I need to include molarity in my materials required for a proposal. I'm assuming it is just 1 M but a reply would be very helpful.

      Honestly, that does look a little weird. However, it is because I washed it with saturated salt solution. It cleaned it up a little.

      Not really. You need the solvent to be immiscible with water. So you're really only left with diethyl ether, chloroform, ethyl acetate, and some others. Of those, DCM and chloroform work the best by far.

      Daily Insanity You really need to use a solvent that is not miscible with water and that can easily dissolve caffeine. DCM is a very good fit for both of these requirements. However, even with a lot of salt, I doubt the other solvents will work very well

      Dissolve it in small amounts of chloroform, wash it a few times with chloroform and transfer to the test time and then evaporate.
      You could also just scrape off the caffeine with a spatula and then transfer it dry, but some would be left behind.

      ***** A can of Monster would have about the same amount of caffeine but would also contain many other chemicals that are soluble in DCM which could be hard to separate later.

      I very much doubt that any soft drink contains as much caffeine as that tea concentrate. Tea is the most concentrated readily available natural souce of caffeine, containing about twice as much as coffee, gram by gram. A cup of coffee contains much more caffeine that a cup of tea, but that's just because using the same weight of tea per cup as we do for coffee would make an undrinkably astringent and bitter brew.

      Yes, it definitely is. There are a few procedures that call for NaOH instead of the carbonate. I am not sure how much to use, but it is definitely a low concentration.

      You can use a soxhlet, but I found a link that suggests that the solubility of caffeine in ethyl acetate is very close to water. Whereas the solubility in DCM or chloroform is >9x more.

      You can use ethyl acetate, but the yield will suffer because more will be left in the water layer. You might be able to fix that by increasing the number of washings. You could also evaporate all the water and dissolve the caffeine from the remnants using methanol or something.
