Maximizing Mycelium: How to Combine Jars of Mushroom Liquid Culture

  • Mushroom Liquid Culture Tip! Sometimes a jar of liquid culture is not full enough to make syringes with due to low nutrients. This method allows you to combine multiple jars of mycelium into one by discarding the excess liquid and leaving behind the mycelium in each jar. The jars of liquid culture need to be the same species.
    I use a laminar flow hood to ensure a sterile working environment but you can watch this video to make a still air box: • How to Make a Still Ai...
    Check the final jar of combined liquid culture on agar plates to verify that it is contamination-free before use.

    How could you get the mycelium to grow more instead of dumping out the excess liquid? Is there anything you could add?

    • @SporenSprout
      @SporenSprout  5 місяців тому +2

      You could probably add something like super concentrated LME water but it would have to be sterilized first.

    Can i ask I don't quite fully understand the difference between spore syringes and liquid culture. Can you explain in a video?
    Thank you, fenomenal work 👍

      Yo, a spore syringe has mushroom spores in it and it takes longer to grow, liquid culture is mycelium already growing!

    Nice, i wouldn't have thought of this. Can you add more sterile nutrients or is that a bad idea? ...nevermind i just read your answer below

    How about just injecting more corn syrup to the jar?

    • @SporenSprout
      @SporenSprout  5 місяців тому +1

      The corn syrup is not sterile so you would just be adding contaminants. You could probably try to sterilize some new lc media and inject that in there or replace the liquid with new more nutritious lc media.

      @@SporenSprout I guess I should have clarified that I did mean adding additional "sterilized Corn Syrup". My thought would be possibly make a jar of 50% water / 50% Corn Syrup, sterilized, that could be drawn from and adding a small amount at a time to the culture till getting the desired effect is achieved.

    I’m using your LC recipe today. Should I change the corn syrup or peptone, LME ratio? I also bought gourmet mushroom LC from you on Etsy. ❤

      You didn't really answer the question and I had the EXACT same one. I'm using your 1/64 tsp peptone, 1/64 tsp LME and 1/2 tsp corn syrup recipe and my cultures are about 1 week in. This makes me wonder if the recipe should be altered?

    • @SporenSprout
      @SporenSprout  5 місяців тому +1

      @@Subsonic-cd2en Oh yeah I glanced right over that question. The jars in this video were only corn syrup but yeah my LME + peptone video is also low nutrient and sometimes with certain species it is great for quick clear growth but not abundant growth. The reason I even used low nutrient to begin with was because I had a big problem with my LCs turning cloudy and I found out the reason was because of my all American sterilizer. So if you use a presto 23 quart pressure cooker you can use more nutrients (1 gram of LME + .5 gram peptone).

      I use the all American canner, I am incredibly curious what the difference is between the presto and mine. Other that one being stainless and the other aluminium and the only other difference I know of is the presto has a steam vent right at the top and the all American has a metal tube that goes down lower to ensure the canner is full of steam at a lower level. Is the metal or the tube and issue? I definitely do want plenty of nutrient content but I have had the same issue with getting cloudy LC and not knowing if it's contaminated due to the amount of sediment and haze. I'm wondering what there is that can be done besides buying a presto to enable me to use half a gram of peptone and a gram of LME, also, do you keep the corn syrup at half a tablespoon with the adage of more of the other two ingredients?

      You should put a lot more water in the all American than you normally would. The water should be touching the bottom 2 inches of the jars. Idk why but I think the water insulates the liquid, That should fix your problem other than switching to presto.

    This is the opposite of multiplication🤷‍♂️ A trick that can help is to attach a small neodymium magnet to the outside of the jar to capture the stir bar, so you can empty all the liquid culture and not loose any.
    So these were from before you gave up on LME?

    • @SporenSprout

      Yeah I guess you could call it “addition” because I added the mycelium together. These jars were made with corn syrup. But yeah low nutrient LME can do the same thing.
