If I order a reel of 100k+ nRF5340, can you pre-program them with my code at your factory?

Unfortunately no, but there are third-party companies that can do this for you. Please contact your RSM and they will help you find a company, or use the sales related contact form on our website to get in touch with RSM www.nordicsemi.com/About-us/Contact-Us

Can we access GPIOs of nRF5340 from this board ? Eg. if i want to measure some voltage from internal ADC and transmit ?

Yes, there are a few free GPIOs on the Thingy:53. Please see "chapter 4.5 Pin maps" in the Thingy:53 user guide: infocenter.nordicsemi.com/pdf/Thingy53_UG.pdf

Hi Zygfryd Homonto, the Nordic Thingy:53 is not trying to compete with minimal dev boards, it is a full-fledged prototyping platform with environmental sensors, color and light sensors, accelerometers, battery, and many more features. If you are looking for a comparable Nordic board our nRF52840 Dongle might be what you are looking for: www.nordicsemi.com/Products/Development-hardware/nRF52840-Dongle
