Yes, the wires must be in the correct position for the proper firing order to take place, otherwise the computer/engine will fire incorrectly and cause a misfire.
I'm going to be doing my spark plus and wires, should I also follow your instructions of wire first then plugs, then back to wire then plugs? So on....
What you can do is you can do one spark plug and wire at a time so that way they don't get mixed up and you should be fine. It doesn't matter which ones you do first as long as the order is the same as in the wires are put the same exact way as they were.
