My question is the solution being tested contains what and what as the constituents?

Ok good question.
The positive test (one one that changed from blue to green, yellow, orange then brown) is a glucose solutionglucose is a reducing sugars.
The other one that did not change (remained blue is just water)
It was the control for the experiment.

i thought we need to put the positive and negative tests in a boiling water in a beaker?

hello cooling is only necessary when testing for non reducing sugars.
This is because for reducing sugars, once you are done with heating, you just write the observation.
